    Capacidad antioxidante total y contenido de polifenoles de vinos argentinos

    Existe evidencia sobre los beneficios del consumo de dietas ricas en frutas y vegetales y una moderada cantidad de vinos tintos en situaciones de salud asociadas a la producción de riesgo oxidativo. Estos efectos protectores se atribuyen a la presencia en los alimentos citados de compuestos con actividad antioxidante, particularmente los polifenoles. En los vinos, los principales compuestos fenólicos son los ácidos galico y cafeico, epicatequina, catequina, cianidina y malvidina-3-glucósidos, rutina, miricetina, quercetina y resveratrol. La Argentina posee regiones muy aptas para la producción vitivinícola siendo el vino obtenido de la Vitis vinifera var. Malbec, uno de los más representativos del país. En la zona del Rio de La Plata y particularmente en Berisso se cultiva la Vitis labrusca var. Isabela de la cual se obtienen el denominado vino de la costa (VC). Existe escasa evidencia sobre la capacidad antioxidante total (CAT) de los vinos Malbec argentinos producidos en diferentes regiones y prácticamente ninguna sobre el vino de la costa. Objetivos: Como parte de un proyecto general del estudio de la capacidad antioxidante de productos naturales, en este trabajo se evalúa la CAT y el contenido de fenoles totales de vinos Malbec de 4 regiones vitivinícolas argentinas y del VC de Berisso utilizando diferentes modelos experimentales

    Antioxidant and cytotoxic properties of an aqueous extract from the Argentinean plant Hedeoma multiflorum

    Context: Hedeoma multiflorum Benth. (Lamiaceae) is widely used in Argentinean popular medicine for digestive and anti-spasmodic purposes. However, knowledge about its pharmacological properties has been poorly investigated. Objective: The antioxidant and cytotoxic properties of an aqueous extract from the plant were investigated for the first time. Material and methods: Scavenging of stable free radicals of 2,2-azino-bis (3-ethylbenzo-thiazoline-6-sulfonic acid) diammonium salt (ABTS+) and 2,2-diphenyl-1-picryl hydrazyl (DPPH), reducing of ferric (III) iron of ferric reducing ability of plasma (FRAP) reagent, and inhibition of lipid peroxidation (LP) of human plasma and rat brain homogenates were assessed. Cytotoxicity was tested on human polymorphonuclear (PMN) cells using the 3-(4,5-dimethylthiazol-2-yl)-2,5- diphenyl tetrazolium bromide (MTT) assay. Cytotoxicity was assessed by flow cytometric techniques. Results: Extract scavenged ABTS+ and DPPH (1.78 and 0.78 Âμ mol Trolox equivalent/mg dry extract, respectively) and reduced FRAP reagent (0.66 μmol ascorbic acid equivalent/mg dry extract). LP of human plasma and rat brain was also inhibited in a dose-dependent way (inhibitory concentration 50%=27.0 and 86.0 μg/mL, respectively). Extract is rich in polyphenol compounds (0.96±0.08 μmol equivalent caffeic acid/mg dry matter). Treatment of PMN decreased significantly the cell ability to reduce the MTT salt and increased the hypodiploid nuclei from 4 to 18% quantified using propidium iodide (PI). In the annexin V-Fluorescein isothiocyanate (annexin V-FITC) assay, 26% of treated cells were annexin V-FITC positive and PI negative. Using the 3,3′-dihexyloxacarbocyanine iodide uptake method, the negative fraction of cells was calculated as 29%. Discussion and conclusion: H. multiflorum extract was found to have a significant antioxidant and pro-apoptotic activities, and a great potential as a source of healthy products.Facultad de Ciencias Médica

    Is the protection against ischemia induced by red wine linked to its antioxidant capacity?

    Objective: To establish whether the total antioxidant capacity of nonalcoholic extracts of three Argentine red wines (RWE) is correlated with their protection against ischemia-reperfusion injury. Animals and methods: The antioxidant properties of three RWE were determined using different free radicalgenerating systems. To examine the effects of these RWE during a 20 min global ischemic period followed by 30 min of reperfusion, isolated rat hearts received 50 µg/mL of RWE 1 (cabernetsauvignon), RWE 2 (malbec) or RWE 3 (a commercial mixture of cabernet-sauvignon, malbec and merlot) 10 min before and after ischemia. Left ventricular developed pressure (LVDP), maximal velocity of rise of left ventricular pressure (+dP/dtmax) and left ventricular end-diastolic pressure (LVEDP) were used to assess contractility and diastolic function. Results: All RWE inhibited lipid peroxidation induced by the Cl4C/NADPH system in a similar proportion (42±4%, 47±9% and 43±14% for RWE 1, RWE 2 and RWE 3, respectively). The scavenging activity of superoxide anion and 2,2-diphenyl-1-picrylhydrazyl radical was about the same with the three RWE. In hearts without RWE treatment, LVDP and +dP/dtmax were 61±4% and 62±5%, respectively, at the end of the reperfusion period. Infusion of RWE 1 and RWE 2 significantly improved postischemic recovery (LVDP and +dP/dtmax were 102±4% and 101±4% for RWE 1 and 92±5% and 91±5% for RWE 2, respectively) and attenuated the increase of LVEDP. RWE 3 did not improve either systolic or diastolic dysfunction. Conclusion: These data show that although the three nonalcoholic RWE exhibit a similar total antioxidant capacity, only two of them protect the heart against myocardial stunning, suggesting that the protective effect is not primarily linked to the antioxidant properties of the extracts.Centro de Investigaciones Cardiovasculare

    En este trabajo se evaluó la citotoxicidad del isoespintanol y dos derivados del mismo, el bromuro de isoespintanol y el desmetil isoespintanol sobre PMN. Materiales y métodos: Se aislaron PMN de sangre de donantes sanos mediante centrifugación en gradiente de Ficoll-Paque. Los PMN se incubaron en presencia de los compuestos citados a 37 °C durante 3 horas en medio PBS completo. Los estudios de citotoxicidad se llevaron acabo utilizando los ensayos de MTT (actividad mitocondrial) y de exclusión del Ioduro de propicio (IP). Mediante ensayos con citometría de flujo (Annexina V-FITC) y Ioduro de propicio se determinó la naturaleza de la citotoxicidad.Facultad de Ciencias Médica
