    HIV-1 Vpr Causes Synaptodendritic Damage in Neurons

    HIV weakens the immune system by infecting and destroying T-cells, leaving the body vulnerable to infection and the development of AIDS. Conventional treatments for HIV, such as combined anti-rectroviral therapy (cART), fail to prevent the development of HIV-associated neurocognitive disorder (HAND). Neurological dysfunction has been directly related to the invasion of HIV in the central nervous system (CNS). HIV produces neurotoxic proteins, such as the Viral Protein R (Vpr), which contribute to HAND. Astrocytes are the most abundant cells in the brain and an important HIV target. We hypothesize that astrocytes expressing Vpr will cause neuronal damage in our co-culture system. Primary astrocytes were transfected with Vpr plasmid or control (pEGFP or mock) using electroporation. Astrocytes were then co-cultured with cortical neurons. At 48 and 72 hours we collected the primary astrocytes to confirm the Vpr expression via western blot analysis. We then measured structural damage in the neurons using immunofluorescence for cytoskeletal (MAP2, f-actin) and synaptic (synaptophysin) damage. Preliminary results showed strong staining of filamentous actin and MAP2 with weak detection of synaptophysin. The positive control for neurotoxicity (2.8µM acrylamide) showed substantial damage to the cellular structure. Results for Vpr expression are pending. After confirming that the immunofluorescence assays are working with our controls, we expect to detect any synaptodendritic damage in the neurons caused by Vpr in our upcoming experiments

    Minimum of η/s\eta/s and the phase transition of the Linear Sigma Model in the large-N limit

    We reexamine the possibility of employing the viscosity over entropy density ratio as a diagnostic tool to identify a phase transition in hadron physics to the strongly coupled quark-gluon plasma and other circumstances where direct measurement of the order parameter or the free energy may be difficult. It has been conjectured that the minimum of eta/s does indeed occur at the phase transition. We now make a careful assessment in a controled theoretical framework, the Linear Sigma Model at large-N, and indeed find that the minimum of eta/s occurs near the second order phase transition of the model due to the rapid variation of the order parameter (here the sigma vacuum expectation value) at a temperature slightly smaller than the critical one.Comment: 22 pages, 19 figures, v2, some references and several figures added, typos corrected and certain arguments clarified, revised for PR

    A Detailed Hydro-Economic Model for Assessing the Effects of Surface Water and Groundwater Policies: A Demonstration Model from Brazil

    Policymakers, managers of water use associations, and many others in developing countries are considering policy actions that will directly or indirectly change the costs and availability of groundwater and surface water for agricultural users. While in many cases such actions may bring about welcomed increases in water use efficiency, little is known about the likely effects of changes in irrigation costs or water access on farmer behavior, or on farmer incomes in the short or long runs, and virtually nothing is known about the detailed immediate or knock-on effects on water resources that such policy actions might cause. This paper reports the preliminary results of research aiming to fill these large scientific gaps by developing a detailed hydrologic model and a detailed economic model of agriculture in the context of the Buriti Vermelho (BV) sub-catchment area of the São Francisco River Basin in Brazil. A spatially explicit, farm-level, positive mathematical programming model capable of accommodating a broad array of farm sizes and farm/farmer characteristics is being developed to predict the effects of alternative water policies and neighbors water use patterns on agricultural production. Special attention is given to precisely defining and estimating the distinct variable costs (including labor and electrical energy costs) and capital costs of surface water and groundwater, which are considered perfect substitutes for irrigation. Shadow values for non-marketed inputs (land, family labor, and water) are estimated in the first step of the modeling process. A high-resolution, spatially distributed hydrologic model (MOD-HMS) is being developed to simulate three-dimensional, variably-saturated subsurface flow and solute transport. Subsurface flow is simulated using the three-dimensional Richards equation while accounting for a) application of water at the surface, b) precipitation, c) soil evaporation and crop transpiration, and d) agricultural pumping. Demonstration versions of both models are presented and tested: the economic model assesses the effects of increasing water scarcity on cultivated area, crop mix, input mix and farm profits; the hydrologic model uses two irrigation water use scenarios to demonstrate the effects of each on surface water flows and storage, and on groundwater storage and well depth. The models are not currently linked, but a detailed plan to do so is presented and discussed.     Cooperative foraging of Paratemnoides nidificator (Balzan, 1888) (Pseudoscorpiones: Atemnidae) on two species of ants (Hymenoptera:Formicidae) in the tropical dry Forest, Colombian Caribbean

    Two new cases of cooperative foraging by Paratemnoides nidificator (Balzan, 1888), in ants Atta cephalotes (Linnaeus, 1758) and Dolichoderus bispinosus Olivier, 1792 are documented. These events were observed in Santa Inés locality, municipality of San Marcos, department of Sucre, Colombian Caribbean. This represents the first record of this behavior of food searching on these species of ants by this social species of pseudoscorpions. It increases the number of known species as preys and that also are part of the diet of this pseudoscorpion).Dos nuevos casos de forrajeo cooperativo por parte de Paratemnoides nidificator (Balzan, 1888) en hormigas Atta cephalotes (Linnaeus, 1758) y Dolichoderus bispinosus (Oliver, 1792) son documentados. Estos eventos se observaron en la localidad de Santa Inés, municipio de San Marcos, departamento de Sucre, Caribe Colombiano. Esto representa el primer registro de este comportamiento de búsqueda de alimento en estas especies de hormigas por esta especie social de pseudoescorpiones.     Hair cell regeneration in the bullfrog vestibular otolith organs following aminoglycoside toxicity

    Adult bullfrogs were given single intraotic injections of the aminoglycoside antibiotic gentamicin sulfate and sacrificed at postinjection times ranging from 0.5 to 9 days. The saccular and utricular maculae of normal and injected animals were examined in wholemount and cross-section. Intraotic 200 (mu) M gentamicin concentrations resulted in the uniform destruction of the hair bundles and, at later times, the cell bodies of saccular hair cells. In the utriculus, striolar hair cells were selectively damaged while extrastriolar hair cells were relatively unaffected. Regenerating hair cells, identified in sectioned material by their small cell bodies and short, well-formed hair bundles, were seen in the saccular and utricular maculae as early as 24-48 h postinjection. Immature versions of mature hair cell types in both otolith organs were recognized by the presence of absence of a bulbed kinocilia and the relative lengths of their kinocilia and longest sterocilia. Utricular hair cell types with kinocilia longer than their longest stereocilia were observed at earlier times than hair cell types with shorter kinocilia. In the same sacculus, the hair bundles of gentamicin-treated animals, even at 9 days postinjection, were significantly smaller than those of normal animals. The hair bundles of utricular hair cells, on the other hand, reached full maturity within the same time period

    Radio Emission from SN 2001gd in NGC 5033

    We present the results of monitoring the radio emission from the Type IIb supernova SN 2001gd between 2002 February 8 and 2002 October 28. Most of the data were obtained using the Very Large Array at the five wavelengths of λλ\lambda \lambda1.3 cm (22.4 GHz), 2.0 cm (14.9 GHz), 3.6 cm (8.44 GHz), 6.2 cm (4.86 GHz), and 21 cm (1.4 GHz). Observations were also made with Giant Meterwave Radio Telescope at λ\lambda21 cm (1.4 GHz). The object was discovered optically well after maximum light, making any determination of the early radio evolution difficult. However, subsequent observations indicate that the radio emission has evolved regularly in both time and frequency and is well described by the SN shock/circumstellar medium interaction model.Comment: 16 pages 2 figures to appear in Astrophysical Journa

    A library of logic models to explain how interventions to reduce diagnostic error work

    OBJECTIVES: We aimed to create a library of logic models for interventions to reduce diagnostic error. This library can be used by those developing, implementing, or evaluating an intervention to improve patient care, to understand what needs to happen, and in what order, if the intervention is to be effective. METHODS: To create the library, we modified an existing method for generating logic models. The following five ordered activities to include in each model were defined: preintervention; implementation of the intervention; postimplementation, but before the immediate outcome can occur; the immediate outcome (usually behavior change); and postimmediate outcome, but before a reduction in diagnostic errors can occur. We also included reasons for lack of progress through the model. Relevant information was extracted about existing evaluations of interventions to reduce diagnostic error, identified by updating a previous systematic review. RESULTS: Data were synthesized to create logic models for four types of intervention, addressing five causes of diagnostic error in seven stages in the diagnostic pathway. In total, 46 interventions from 43 studies were included and 24 different logic models were generated. CONCLUSIONS: We used a novel approach to create a freely available library of logic models. The models highlight the importance of attending to what needs to occur before and after intervention delivery if the intervention is to be effective. Our work provides a useful starting point for intervention developers, helps evaluators identify intermediate outcomes, and provides a method to enable others to generate libraries for interventions targeting other errors

    Traveling with an invader: ectoparasitic mites of Hemidactylus frenatus (Squamata: Gekkonidae) in Colombia

    Information about natural history of invasive species (such as parasitism) is a relevant issue in order to consider potential threats to native fauna. In this work we report two pterygosomatid mites for the first time in Colombia, Geckobia bataviensis and G. keegani, parasites of the invasive house gecko Hemidactylus frenatus. Although it has not been demonstrated that these mites may infest native gecko lizards, our study highlights the need to explore the prevalence of internal and external parasites in native and invasive geckos, to better evaluate the potential impact of invasive Hemidactylus lizards in Colombia.Asociación Herpetológica Argentin
