    Teaching of Physical Education Course as a Fundamental Right of Students in Secondary Schools in Kenya: A Study of Bomet District

    The purpose of this study was to assess the gap in policy between the promise and reality in physical education in secondary schools in Bomet District - Kenya. The objective was to establish the extent to which the teaching of physical education formed a fundamental right for students in secondary schools. Biological Sciences Curriculum Study model guided the study. This study utilized descriptive survey method of research. Sampling designs used were simple random, purposive and stratified random sampling. Questionnaire, interview guide and observation schedules were used to collect data and a total of 281 respondents took part in the study. Data analysis was done using descriptive and inferential statistics. It was found that the students were denied their right to be educated physically in secondary schools in Bomet District. This research recommends that the Directorate of Quality Assurance should deploy specialist physical educators to district levels that are responsible for the periodic supervision and evaluation of physical education in secondary schools.     Assessment of phenotypes, physiological and behavioural responses associated with heat tolerance among Galla goats in North Eastern Kenya.

    Galla goats have great adaptation vigor to different environments. However, the outcomes of the ongoing climate change have subjected livestock including the Galla goats to harsh environmental conditions like high ambient temperatures. In this study, Galla goats were characterized in the selected areas based on phenotypic, physiological, and behavioural responses to high environmental temperature. One hundred and forty-nine Galla goats selected from Isiolo, Garissa and Tana River were scored for various features such as coat colour, horn colour, skin colour, horn shape, horn length, ear orientation, ear length, wattles and horn circumference. Data collected were entered in an excel spread sheet then analyzed in R statistical software version 4.0.4. White was found to be the dominant coat colour with a percentage frequency greater than 60.0% in the studied counties. The study revealed that both male and female Galla goats possess horns with more than 88.9% of goats being horned across the three counties. There was a strong relationship between horn presence and the environmental temperature with p=0.05. A positive correlation was also observed between environmental temperature and rectal temperature, horn length, ear length and horn circumference. The findings show the presence of an important gene pool from which guidelines on conservation of heat stress phenotypes as a result of the ongoing climate change can be put in place

    Operational Research Project Management, Experiences, Challenges and Lessons Learnt

    Introduction: Effective project management revolves around Strategic Management. Logistics seem simple and straight forward but, often the role  it plays in scientific undertakings is overlooked. It is usually assumed that research starts and ends in the laboratory. It is a fact that, for research activities to be successful, it requires exceptional planning to ensure that, resources are available as per the approved work-plan. This entails  determination of what, when, who, why and how it is to be done. Recent studies indicate that, logistics-related activities' impact on research  undertakings significantly. Objective: To document the project management experiences and lessons learnt in coordinating and implementation of East Africa Public Health Laboratories Networking Project –Operational Research (EAPHLNP-OR) activities in five East African countries, namely: Kenya, Rwanda, Burundi, Uganda and Tanzania. Methodology: The operational research component of the EAPHLNP, KEMRI established an OR Secretariat to coordinate the project activities in  Kenya and provide leadership to regional principal investigators. In consultation with the project Secretariat, the role of the administrator involved Work plan and budget preparation, planning, organizing, communicating, coordinating local and regional meetings, linking KEMRI research team with the study site (Hospital Administration) and Research Teams in the various counties. The site Teams obtained informed consent, recruited respondents, collected specimens, analyzed the specimens and shipped a portion of the same together with the results to KEMRI. Key Activities Of The Project: Managing financial aspects (budget and financial report preparations), logistical coordination, and procurement of training materials, organizing for meeting venues, taking minutes, travel arrangements and participation in scientific report writing. Control mechanism such as dairies, ledger books, work plan charts and schedules, managing and monitoring the progress of the project activities. Lesson Learnt & Challenges: Interpersonal skills were essential at all stages of the project. The critical stage was the forming, storming, and  norming stages. Here, group dynamics and conflicts took center stage. This threatened to stall the OR Project. Timely and constant communication with the study site coordinators, prioritization of scheduled project activities, was essential. Ensuring all parties are kept informed on the progress of the OR activities. The information in user-friendly format dairies and schedules provided the necessary feedback at administrative level, on project performance and at research findings. Key challenges included fluctuating funding, group dynamic conflicts and staff transfers. Discussion: EAPHLNP-OR was a Seven (7) years project undertaking, which for effective management involved understanding of the operating environment, strategic planning for short and long term goals, constant communication, review of priorities, documentation and practise of goodinterpersonal skills. Conclusion: Successful project management in OR required an administrator to coordinate the utilization of the available resources both capital and human.     Diversity of Sorghum (Sorghum bicolor L. Moench) Germplasm from Tanzania

    Sorghum (Sorghum bicolor L. Moench) is an important cereal crop cultivated in varied agro-ecological zones of Tanzania ranging from the southern highlands to coastal lowlands. Different cultivars and varieties of sorghum are needed for cultivation in such varied zones. Phenotypic properties of Tanzanian sorghums are not well-studied. Objective of this study was to phenotype some sorghum germplasm from Tanzania using morphological markers to establish their diversity for future use in breeding programs. Ninety-eight sorghum genotypes were evaluated at Bumala and Amagoro in Western Kenya during 2009 and 2010 seasons in a randomized complete block design. The International Plant Genetic Resource Centre descriptors of 1993 were used for data collection. Accession MCSR T29 was the earliest which took only 64 days while MCSR T80 was the latest taking 86 days to attain 50% flowering compared to the overall mean of 71 days. The MCSR T71 was the best yielder (114.6 g/ panicle) and MCSR T10 was the lowest (10.3 g/panicle) compared to overall mean of 92.4g/panicle. About 60% of the sorghums had brown and only 2.2% had black grains. MCSR T90 had bold grains with 1000 seed weight of 55.2g. Leaves per plant ranged from 7 in MCSR T69 to 17 in MCSR T25. Plant height ranged from 72.8 cm in MCSR T53 to 434.6 cm in MCSR T80. This study showed that, Tanzanian sorghums are diverse therefore could be used in future breeding programs for developing multipurpose and adapted cultivars.     Lethal Mutagenesis of Picornaviruses with N-6-Modified Purine Nucleoside Analogues

    RNA viruses exhibit extraordinarily high mutation rates during genome replication. Nonnatural ribonucleosides that can increase the mutation rate of RNA viruses by acting as ambiguous substrates during replication have been explored as antiviral agents acting through lethal mutagenesis. We have synthesized novel N-6-substituted purine analogues with ambiguous incorporation characteristics due to tautomerization of the nucleobase. The most potent of these analogues reduced the titer of poliovirus (PV) and coxsackievirus (CVB3) over 1,000-fold during a single passage in HeLa cell culture, with an increase in transition mutation frequency up to 65-fold. Kinetic analysis of incorporation by the PV polymerase indicated that these analogues were templated ambiguously with increased efficiency compared to the known mutagenic nucleoside ribavirin. Notably, these nucleosides were not efficient substrates for cellular ribonucleotide reductase in vitro, suggesting that conversion to the deoxyriboucleoside may be hindered, potentially limiting genetic damage to the host cell. Furthermore, a high-fidelity PV variant (G64S) displayed resistance to the antiviral effect and mutagenic potential of these analogues. These purine nucleoside analogues represent promising lead compounds in the development of clinically useful antiviral therapies based on the strategy of lethal mutagenesis

    Pandoraea fibrosis sp. Nov., a novel pandoraea species isolated from clinical respiratory samples

    © 2019 IUMS. Pandoraea species have been isolated from diverse environmental samples and are emerging important respiratory pathogens, particularly in people with cystic fibrosis (CF). In the present study, two bacterial isolates initially recovered from consecutive sputum samples collected from a CF patient and identified as Pandoraea pnomenusa underwent a polyphasic taxonomic analysis. The isolates were found to be Gram-negative, facultative anaerobic motile bacilli and subsequently designated as strains 6399 T (=LMG29626 T =DSM103228 T ) and 7641 (=LMG29627=DSM103229), respectively. Phylogenetic analysis based on 16S rRNA and gyrB gene sequences revealed that 6399 T and 7641 formed a distinct phylogenetic lineage within the genus Pandoraea. Genome sequence comparison analysis indicated that strains 6399 T and 7641 are clonal and share 100 % similarity, however, similarity to other type strains (ANIb 73.2–88.8 %, ANIm 83.5–89.9 % and OrthoANI 83.2– 89.3 %) indicates that 6399 T and 7641 do not belong to any of the reported type species. The major cellular fatty acids of 6399 T were C 16: 0 (32.1 %) C 17: 0 cyclo (18.7 %) and C 18: 1 !7c (14.5 %), while Q-8 was the only respiratory quinone detected. The major polar lipids identified were phosphatidylethanolamine, phosphatidylglycerol and diphosphatidylglycerol. The genomic DNA G+C content of 6399 T was 62.9 (mol%). Strain 6399 T can be differentiated from other members of Pandoraea by the absence of C 19: 0 !8c cyclo and by the presence of C 17: 0 !8c cyclo. Together our data show that the bacterial strains 6399 T and 7641 represent a novel species of the genus Pandoraea, for which the name Pandoraea fibrosis sp. nov. is proposed (type strain 6399 T )

    Cost-effectiveness of installing barriers at bridge and cliff sites for suicide prevention in Australia

    Importance: Installation of barriers has been shown to reduce suicides. To our knowledge, no studies have evaluated the cost-effectiveness of installing barriers at multiple bridge and cliff sites where suicides are known to occur. Objective: To examine the cost-effectiveness of installing barriers at bridge and cliff sites throughout Australia. Design, Setting, and Participants: This economic evaluation used an economic model to examine the costs, costs saved, and reductions in suicides if barriers were installed across identified bridge and cliff sites over 5 and 10 years. Specific and accessible bridge and cliff sites across Australia that reported 2 or more suicides over a 5-year period were identified for analysis. A partial societal perspective (including intervention costs and monetary value associated with preventing suicide deaths) was adopted in the development of the model. Interventions: Barriers installed at bridge and cliff sites. Main Outcomes and Measures: Primary outcome was return on investment (ROI) comparing cost savings with intervention costs. Secondary outcomes included incremental cost-effectiveness ratio (ICER), comprising the difference in costs between installation of barriers and no installation of barriers divided by the difference in reduction of suicide cases. Uncertainty and sensitivity analyses were undertaken to examine the association of changes in suicide rates with barrier installation, adjustments to the value of statistical life, and changes in maintenance costs of barriers. Results: A total of 7 bridges and 19 cliff sites were included in the model. If barriers were installed at bridge sites, an estimated US 145million(95145 million (95% uncertainty interval [UI], 90 to 160million)couldbesavedinpreventedsuicidesover5years,andUS160 million) could be saved in prevented suicides over 5 years, and US 270 million (95% UI, 176to176 to 298 million) over 10 years. The estimated ROI ratio for building barriers over 10 years at bridges was 2.4 (95% UI, 1.5 to 2.7); the results for cliff sites were not significant (ROI, 2.0; 95% UI, -1.1 to 3.8). The ICER indicated monetary savings due to averted suicides over the intervention cost for bridges, although evidence for similar savings was not significant for cliffs. Results were robust in all sensitivity analyses except when the value of statistical life-year over 5 or 10 years only was used. Conclusions and Relevance: In an economic analysis, barriers were a cost-effective suicide prevention intervention at bridge sites. Further research is required for cliff sites

    Mitochondrial Localized STAT3 Is Involved in NGF Induced Neurite Outgrowth

    Background: Signal transducer and activator of transcription 3 (STAT3) plays critical roles in neural development and is increasingly recognized as a major mediator of injury response in the nervous system. Cytokines and growth factors are known to phosphorylate STAT3 at tyrosine 705 with or without the concomitant phosphorylation at serine 727, resulting in the nuclear localization of STAT3 and subsequent transcriptional activation of genes. Recent evidence suggests that STAT3 may control cell function via alternative mechanisms independent of its transcriptional activity. Currently, the involvement of STAT3 mono-phosphorylated at residue serine 727 (P-Ser-STAT3) in neurite outgrowth and the underlying mechanism is largely unknown. Principal Findings: In this study, we investigated the role of nerve growth factor (NGF) induced P-Ser-STAT3 in mediating neurite outgrowth. NGF induced the phosphorylation of residue serine 727 but not tyrosine 705 of STAT3 in PC12 and primary cortical neuronal cells. In PC12 cells, serine but not tyrosine dominant negative mutant of STAT3 was found to impair NGF induced neurite outgrowth. Unexpectedly, NGF induced P-Ser-STAT3 was localized to the mitochondria but not in the nucleus. Mitochondrial STAT3 was further found to be intimately involved in NGF induced neurite outgrowth and the production of reactive oxygen species (ROS).
