    Techno-economic and greenhouse gas savings assessment of decentralized biomass gasification for electrifying the rural areas of Indonesia

    This study explored the feasibility of decentralized gasification of oil palm biomass in Indonesia to relieve its over-dependence on fossil fuel-based power generation and facilitate the electrification of its rural areas. The techno-feasibility of the gasification of oil palm biomass was first evaluated by reviewing existing literature. Subsequently, two scenarios (V1 and V2, and M1 and M2) were proposed regarding the use cases of the village and mill, respectively. The capacity of the gasification systems in the V1 and M1 scenarios are determined by the total amount of oil palm biomass available in the village and mill, respectively. The capacity of the gasification systems in the V2 and M2 scenarios is determined by the respective electricity demand of the village and mill. The global warming impact and economic feasibility (net present value (NPV) and levelized cost of electricity (LCOE)) of the proposed systems were compared with that of the current practices (diesel generator for the village use case and biomass boiler combustion for the mill use case) using life cycle assessment (LCA) and cost-benefit analysis (CBA). Under the current daily demand per household (0.4 kWh), deploying the V2 system in 104 villages with 500 households each could save up to 17.9 thousand tons of CO2-eq per year compared to the current diesel-based practice. If the electricity could be fed into the national grid, the M1 system with 100% capacity factor could provide yearly GHG emissions mitigation of 5.8 × 104 ton CO2-eq, relative to the current boiler combustion-based reference scenario. M1 had a positive mean NPV if the electricity could be fed into the national grid, while M2 had a positive mean NPV at the biochar price of 500 USD/ton. Under the current electricity tariff (ET) (0.11 kWh) and the biochar price of 2650 USD/ton, daily household demands of 2 and 1.8 kWh were required to reach the break-even point of the mean NPV for the V2 system for the cases of 300 and 500 households, respectively. The average LCOE of V2 is approximately one-fourth that of the reference scenario, while the average LCOE of V1 is larger than that of the reference scenario. The average LCOE of M1 decreased to around 0.06 USD/kWh for the case of a 100% capacity factor. Sensitivity analysis showed that the capital cost of gasification system and its overall electrical efficiency had the most significant effects on the NPV. Finally, practical system deployment was discussed, with consideration of policy formulation and fiscal incentives

    Trade-induced atmospheric mercury deposition over China and implications for demand-side controls

    Mercury (Hg) is of global concern because of its adverse effects on humans and the environment. In addition to long-range atmospheric transport, Hg emissions can be geographically relocated through economic trade. Here, we investigate the effect of China’s interregional trade on atmospheric Hg deposition over China, using an atmospheric transport model and multiregional input-output analysis. In general, total atmospheric Hg deposition over China is 408.8 Mg yr-1, and 32% of this is embodied in China’s interregional trade, with the hotspots occurring over Gansu, Henan, Hebei, and Yunnan provinces. Interprovincial trade considerably redistributes atmospheric Hg deposition over China, with a range in deposition flux from −104% to +28%. Developed regions, such as the Yangtze River Delta (Shanghai, Jiangsu, and Zhejiang) and Guangdong, avoid Hg deposition over their geographical boundaries, instead causing additional Hg deposition over developing provinces. Bilateral interaction among provinces is strong over some regions, suggesting a need for joint mitigation, such as the Jing-Jin-Ji region (Beijing, Tianjin, and Hebei) and the Yangtze River Delta. Transferring advanced technology from developed regions to their developing trade partners would be an effective measure to mitigate China’s Hg pollution. Our findings are relevant to interprovincial efforts to reduce trans-boundary Hg pollution in China

    High level soluble expression, one-step purification and characterization of HIV-1 p24 protein

    <p>Abstract</p> <p>Background</p> <p>P24 protein is the major core protein of HIV virus particle and has been suggested as a specific target for antiviral strategies. Recombinant p24 protein with natural antigenic activity would be useful for various studies, such as diagnostic reagents and multi-component HIV vaccine development. The aim of this study was to express and purify the p24 protein in soluble form in <it>E.coli</it>.</p> <p>Results</p> <p>According to the sequence of the p24 gene, a pair of primers was designed, and the target sequence of 700 bp was amplified using PCR. The PCR product was cloned into pQE30 vector, generating the recombinant plasmid pQE30-p24. SDS-PAGE analysis showed that the His-tagged recombinant p24 protein was highly expressed in soluble form after induction in <it>E. coli </it>strain BL21. The recombinant protein was purified by nickel affinity chromatography and used to react with HIV infected sera. The results showed that the recombinant p24 protein could specifically react with the HIV infected sera. To study the immunogenicity of this soluble recombinant p24 protein, it was used to immunize mice for the preparation of polyclonal antibody. Subsequent ELISA and Western-Blot analysis demonstrated that the p24 protein had proper immunogenicity in inducing mice to produce HIV p24 specific antibodies.</p> <p>Conclusion</p> <p>In this work, we report the high level soluble expression of HIV-1 p24 protein in <it>E. coli</it>. This soluble recombinant p24 protein specifically react with HIV infected sera and elicit HIV p24 specific antibodies in mice, indicating this soluble recombinant p24 protein could be a promising reagent for HIV diagnosis.</p

    Is inhibition involved in voluntary language switching? Evidence from transcranial direct current stimulation over the right dorsolateral prefrontal cortex.

    How do bilinguals freely switch languages in daily life (i.e., voluntary language switching)? Does this involve inhibition of cross-language interference, or does easier lexical access to specific words directly trigger switches to another language? To reveal the underlying mechanism of voluntary language switching, the current study applied transcranial direct current stimulation (tDCS) over the right dorsolateral prefrontal cortex (rDLPFC), an area associated with inhibitory control, to investigate whether and how inhibitory control is involved in voluntary language switching. Interestingly, during the A-tDCS and C-tDCS sessions, bilinguals showed faster naming latencies in repeat trials compared to switch trials, indicating a switch cost. Moreover, the cathodal stimulation exhibited a larger LPC in switching to L2 compared to switching to L1. In contrast, the S-tDCS (sham) session showed no switch cost, faster switch naming latencies compared to the A-tDCS and C-tDCS sessions, and little differences in LPC between switch and repeat trials. Together, these findings suggest that modulating inhibition-related brain regions interferes with voluntary language switching

    The importance of NOx control for peak ozone mitigation based on a sensitivity study using CMAQ‐HDDM‐3D model during a typical episode over the Yangtze River delta region, China.

    In recent years, ground-level ozone (O3) has been one of the main pollutants hindering air quality compliance in China's large city-clusters including the Yangtze River Delta (YRD) region. In this work, we utilized the process analysis (PA) and the higher-order decoupled direct method (HDDM-3D) tools embedded in the Community Multiscale Air Quality model (CMAQ) to characterize O3 formation and sensitivities to precursors during a typical O3 pollution episode over the YRD region in July 2018. Results indicate that gas-phase chemistry contributed dominantly to the ground-level O3 although a significant proportion was chemically produced at the middle and upper boundary layer before reaching the surface via diffusion process. Further analysis of the chemical pathways of O3 and Ox formation provided deep insights into the sensitivities of O3 to its precursors that were consistent with the HDDM results. The first-order sensitivities of O3 to anthropogenic volatile organic compounds (AVOC) were mainly positive but small, and temporal variations were negligible compared with those to NOx. During the peak O3 time in the afternoon, the first- and second-order sensitivities of O3 to NOx were significantly positive and negative, respectively, suggesting a convex response of O3 to NOx over most areas including Shanghai, Hangzhou, Nanjing and Hefei. These findings further highlighted an accelerated decrease in ground-level O3 in the afternoon corresponding to continuous decrease of NOx emissions in the afternoon. Therefore, over the YRD region including its metropolises, NOx emission reductions will be more important in reducing the afternoon peak O3 concentration compared with the effect of VOC emission control alone

    Alteration in Oral Microbiome Among Men Who Have Sex With Men With Acute and Chronic HIV Infection on Antiretroviral Therapy

    Despite the antiretroviral therapy (ART), human immunodeficiency virus (HIV)-related oral disease remains a common problem for people living with HIV (PLWH). Evidence suggests that impairment of immune function in HIV infection might lead to the conversion of commensal bacteria to microorganisms with increased pathogenicity. However, limited information is available about alteration in oral microbiome in PLWH on ART. We performed a longitudinal comparative study on men who have sex with men (MSM) with acute HIV infection (n=15), MSM with chronic HIV infection (n=15), and HIV-uninfected MSM controls (n=15). Throat swabs were collected when these subjects were recruited (W0) and 12 weeks after ART treatment (W12) from the patients. Genomic DNAs were extracted and 16S rRNA gene sequencing was performed. Microbiome diversity was significantly decreased in patients with acute and chronic HIV infections compared with those in controls at the sampling time of W0 and the significant difference remained at W12. An increased abundance of unidentified Prevotellaceae was found in patients with acute and chronic HIV infections. Moreover, increased abundances of Prevotella in subjects with acute HIV infection and Streptococcus in subjects with chronic HIV infection were observed. In contrast, greater abundance in Lactobacillus, Rothia, Lautropia, and Bacteroides was found in controls. After effective ART, Bradyrhizobium was enriched in both acute and chronic HIV infections, whereas in controls, Lactobacillus, Rothia, Clostridia, Actinobacteria, and Ruminococcaceae were enriched. In addition, we found that lower CD4(+) T-cell counts (< 200 cells/mm(3)) were associated with lower relative abundances of Haemophilus, Actinomyces, unidentified Ruminococcaceae, and Rothia. This study has shown alteration in oral microbiome resulting from HIV infection and ART. The results obtained warrant further studies in a large number of subjects with different ethnics. It might contribute to improved oral health in HIV-infected individuals

    Prevalence and Prognostic Significance of HPV in Laryngeal Squamous Cell Carcinoma in Northeast China

    Background/Aims: Human papillomavirus (HPV) is an etiological risk factor for a subset of head and neck squamous cell carcinomas. HPV has been proven to be a powerful prognostic biomarker for oropharyngeal cancer, but its role in the larynx has not been explored in depth. Here, we sought to evaluate the prevalence and genotype distribution of HPV in patients with laryngeal squamous cell carcinoma (LSCC) in northeast China. Methods: HPV DNA in specimens from 211 patients diagnosed with LSCC was analyzed by the polymerase chain reaction and in situ hybridization, and p16 overexpression was evaluated by immunohistochemistry. p16 expression was scored positive if strong and diffuse nuclear and cytoplasmic staining was present in &#x3e; 75% of tumor cells. Results: In this study, infection with HPV and p16 expression were not absolutely consistent. Among all patients, 132 (62.6%) were positive for HPV DNA (HPV+), while 23 (10.9%) were inconsistent for HPV and p16. Multivariate analysis indicated that HPV, but not p16, is an independent prognostic factor for overall survival in LSCC. Overall survival was significantly improved in HPV+ LSCC patients compared with the HPV-negative group (hazard ratio, 0.395; 95% confidence interval, 0.185–0.843; p = 0.016). Among the 132 HPV+ patients, 28 (21.2%) were HPV-16 single infection. Conclusion: This study indicates that HPV DNA is a more reliable surrogate marker than p16 for the prediction of survival in patients with LSCC

    Protocol for a Single-Center Randomized Controlled Trial of Percutaneous Coronary Intervention Via Distal Transradial Access Versus Transradial Access

    Background: Although transradial access (TRA) has become the main vascular access for coronary intervention, its high radial artery occlusion rate limits its application in some patients. Studies have shown that compared with TRA, distal transradial access (dTRA) with the snuffbox area or the Hegu acupoint area as the puncture point significantly decreases the incidence of radial artery occlusion. However, no randomized controlled study has confirmed the safety and efficacy of coronary artery intervention via dTRA in China. Methods and analyses: This single-center, prospective, randomized controlled, superiority open-label study will enroll 428 consecutive patients with coronary heart disease undergoing percutaneous coronary intervention as the study population. After preoperative evaluation, the participants will be randomly divided into a study group (dTRA) and control group (TRA) in a 1:1 ratio. The primary endpoint (radial artery occlusion at 24 hours after operation) and secondary endpoint events will be evaluated and recorded.
