V diplomové práci se zabývá konstrukčním návrhem automobilového zámku, který má hlavní komponenty v „plastovém“ provedení (rohatka, západka, tělesa). Tyto plastové díly jsem vhodně navrhl a optimalizoval na požadované zatížení. Zároveň srovnávám plastový zámek a ocelový zámek přední kapoty, srovnávám jak cenu zámků, tak hmotnost a zatížení zámků. V krátkosti porovnávám automobilové zámky přední kapoty v současném automobilovém průmyslu. Také se zabývám etapou výroby prototypového vzorku v současnosti.The diploma thesis deals with the design of the car lock, which main components such as claw, pawl and housing are made of plastic. My main goal was to optimized these plastic parts for a certain amount of power. The following part shows a comparison of plastic lock and the steel lock of front bonnet. The following part shows a comparison of the plastic lock and the steel lock of the front bonnet. The comparison is made in terms of the price, weight and load of the locks. Structure-activity relationship study of the neuritogenic potential of the glycan of starfish ganglioside LLG-3
LLG-3 is a ganglioside isolated from the starfish Linchia laevigata. To clarify the structure-activity relationship of the glycan of LLG-3 toward rat pheochromocytoma PC12 cells in the presence of nerve growth factor, a series of mono- to tetrasaccharide glycan derivatives were chemically synthesized and evaluated in vitro. The methyl group at C8 of the terminal sialic acid residue was crucial for neuritogenic activity, and the terminal trisaccharide moiety was the minimum active motif. Furthermore, the trisaccharide also stimulated neuritogenesis in human neuroblastoma SH-SY5Y cells via mitogen-activated protein kinase (MAPK) signaling. Phosphorylation of extracellular signal-regulated kinase (ERK) 1/2 was rapidly induced by adding 1 or 10 nM of the trisaccharide. The ratio of phosphorylated ERK to ERK reached a maximum 5 min after stimulation, and then decreased gradually. However, the trisaccharide did not induce significant Akt phosphorylation. These effects were abolished by pretreatment with the MAPK inhibitor U0126, which inhibits enzymes MEK1 and MEK2. In addition, U0126 inhibited the phosphorylation of ERK 1/2 in response to the trisaccharide dose-dependently. Therefore, we concluded that the trisaccharide promotes neurite extension in SH-SY5Y cells via MAPK/ERK signaling, not Akt signaling
Expression and characterization of honeybee, Apis mellifera, larva chymotrypsin-like protease
International audiencePreviously, we found three enzyme fractions containing activities for the hydrolysis of royal jelly proteins from honeybee queen larvae. In this study, we identified a honeybee chymotrypsin-like protease (HCLPase) by LC-MS/MS and expressed it as a recombinant protein in Escherichia coli. The protease had an estimated molecular weight of around 26 kDa and showed high specificity for succinyl-Ala-Ala-Pro-Phe p-nitroanilide as a proteolytic substrate. Furthermore, the protease had an optimal pH of 9, and the activity was markedly inhibited by phenylmethylsulfonyl fluoride but not tosyl phenylalanyl chloromethyl ketone, both of which are irreversible inhibitors of chymotrypsin-like serine proteases. These results suggested that this recombinant protease, HCLPase, was a chymotrypsin-like serine protease with different characteristics from mammalian chymotrypsin
