    Quantification of 'Candidatus Phytoplasma prunorum' in apricot trees exhibiting uneven European stone fruit yellows symptoms

    This study focused on quantification of 'Candidatus Phytoplasma prunorum' in apricot trees exhibiting uneven symptoms of European stone fruit yellows. For the research, 17 trees showing early leaf bud break in one part of the crown were selected in winter season, and 13 trees showing leafroll in older part of the shoots and no symptoms in younger part of the same shoots were selected in summer season. Absolute quantification of phytoplasmas was performed by real-time PCR in samples collected from asymptomatic and symptomatic parts. The results showed, that phytoplasma could not be always detected in asymptomatic parts, where 2 out of 17 samples from asymptomatic parts from winter season were negative while in summer season 6 out of 13 samples from asymptomatic parts were negative. Comparison of phytoplasma quantities between symptomatic and asymptomatic plant parts then showed significantly higher quantities in symptomatic parts from samples collected in summer season and no quantity difference in samples collected in winter season.O

    Antioxidant capacity, total phenolic compounds and fatty acids composition in walnut oil and bagasse pellets produced at different parameters of the screw press

    Different rotational speeds and nozzle diameters of screw press were used to process walnut kernels. To evaluate the influence of pressing conditions on the quality of walnut oil and bagasse pellets the oil pressing temperature, yield of oil, total phenolic compounds, antioxidant capacity and fatty acid composition were measured. The results show that the pressing at different conditions affected the antioxidant capacity and TPC of either bagasse pellets and pressed oil. Higher values of antioxidant capacity and TPC were measured in bagasse pellets than in pressed oils. Higher amount of pressed oil was yielded by using lower nozzle diameter, however, by lowering the nozzle diameter the oil pressing temperature was rising, which affected the fatty acids. Positive correlations with oil pressing temperatures were calculated at saturated fatty acids, while the correlation coefficients of polyunsaturated fatty acids were positive, but not significant. Monounsaturated fatty acids were negatively affected by higher oil pressing temperatures showing, that monounsaturated fatty acids were more susceptible to higher temperatures than polyunasturated and saturated fatty acids.O

    Cieľom tejto bakalárskej práce je poskytnúť informácie o základnej problematike dátových skladov, dátových kociek a OLAP analýzy. V teoretickej časti práce sa predstavuje etapa predspracovania dát a jeho elementárne časti prevedené pred ich samotným vkladaním do dátových skladov. Okrem toho sú stručne uvedené základné typy OLAP operácii a možnosti zobrazenia dátových kociek. Získané poznatky sú využité k vytvoreniu hlavného výsledku práce, ktorá má podobu aplikácie pre zobrazovanie OLAP dátových kociek vo vhodne zvolenej podobe. Vytvorené riešenie poskytuje možnosť prevedenia základných operácii OLAP analýzy nad výslednými dátovými kockami.The main aim of this thesis is to provide information about the basic issues of data warehouses, data cubes and OLAP analysis. Furthermore, in the theoretical part is introduced the stage of data preprocessing and its elementary parts performed before their insertion in to data warehouses. Additionally, there are listed the basic types of OLAP operations and the possible ways of displaying data cubes. Acquired information is used to create application for displaying OLAP data cubes in suitable format.

    Reduced graphene oxide-based nanometal-composite containing copper and silver nanoparticles protect tomato and pepper against Xanthomonas euvesicatoria infection

    Xanthomonas euvesicatoria (Xe) is of the genera Xanthomonas, which causes a bacterial spot disease that affects tomato and pepper plants, resulting in significant economic losses. Commonly used bactericides lead to pathogen resistance, environmental contamination and a current risk to human health. Herein, a nanocomposite consisting of Cu and Ag loaded onto reduced graphene oxide (rGO) was synthesized, characterized and tested. After confirmation of the antibacterial properties in vitro, nanocomposites at concentrations of 50 and 500 µg mL MINUS SIGN 1 were applied to protect tomato and pepper plants under controlled greenhouse conditions. The tomato and pepper plants indicated significantly lower disease severity when treated with the nanocomposite (15.6 and 16.7%, respectively) than when treated with the commercial copper-based bactericide Kocide(R) 2000. The nanocomposite was demonstrated as a high-efficiency biocide and has the potential for crop disease management with no phytotoxic effect.O

    Single-molecule observation of the induction of k-turn RNA structure on binding L7Ae protein

    AbstractThe k-turn is a commonly occurring structural motif that introduces a tight kink into duplex RNA. In free solution, it can exist in an extended form, or by folding into the kinked structure. Binding of proteins including the L7Ae family can induce the formation of the kinked geometry, raising the question of whether this occurs by passive selection of the kinked structure, or a more active process in which the protein manipulates the RNA structure. We have devised a single-molecule experiment whereby immobilized L7Ae protein binds Cy3-Cy5-labeled RNA from free solution. We find that all bound RNA is in the kinked geometry, with no evidence for transitions to an extended form at the millisecond timescale of the camera. Furthermore, real-time binding experiments provide no evidence for a more extended intermediate even at the earliest times, at a time resolution of 16 ms. The data support a passive conformational selection model by which the protein selects a fraction of RNA that is already in the kinked conformation, thereby drawing the equilibrium into this form

    GRBAlpha: the smallest astrophysical space observatory -- Part 1: Detector design, system description and satellite operations

    Aims. Since launched on 2021 March 22, the 1U-sized CubeSat GRBAlpha operates and collects scientific data on high-energy transients, making it the smallest astrophysical space observatory to date. GRBAlpha is an in-obit demonstration of a gamma-ray burst (GRB) detector concept suitably small to fit into a standard 1U volume. As it was demonstrated in a companion paper, GRBAlpha adds significant value to the scientific community with accurate characterization of bright GRBs, including the recent outstanding event of GRB 221009A. Methods. The GRB detector is a 75x75x5 mm CsI(Tl) scintillator wrapped in a reflective foil (ESR) read out by an array of SiPM detectors, multi-pixel photon counters by Hamamatsu, driven by two separate, redundant units. To further protect the scintillator block from sunlight and protect the SiPM detectors from particle radiation, we apply a multi-layer structure of Tedlar wrapping, anodized aluminium casing and a lead-alloy shielding on one edge of the assembly. The setup allows observations of gamma radiation within the energy range of 70-890 keV with an energy resolution of ~30%. Results. Here, we summarize the system design of the GRBAlpha mission, including the electronics and software components of the detector, some aspects of the platform as well as the current way of semi-autonomous operations. In addition, details are given about the raw data products and telemetry in order to encourage the community for expansion of the receiver network for our initiatives with GRBAlpha and related experiments.Comment: Accepted for publication in Astronomy & Astrophysics, 9 pages, 10 figure
