Sephadex je v biochemických experimentech hojně využívaným sorbentem k separacím molekul v širokém rozmezí molekulových hmotností. Využívá se také k purifikacím nebo odsolování izolovaných proteinů. Dosud byly chromatografické aplikace na tomto principu prováděny jen v kolonách velkých rozměrů, čemuž musela předcházet účinná izolace proteinů, aby bylo na kolonu naneseno dostatečné množství vzorku. Naplnění sephadexu do mikrokolony by snížilo potřebné množství vzorku a mohly by tak být dále zpracovávány obtížně izolovatelné proteiny, které se získávají v malém množství. V této bakalářské práci je s použitím suspenzního plnění mikrokolona opravdu naplněna. K vytvoření gelu jako takového je použito principu přípravy monolitických kolon, tedy naplnění a následné polymerizace, v našem případě hydratace a zgelovatěním.Sephadex is a common sorbent in biochemistry for separation of molecules of a wide range of molecular masses. V článku je prezentován průběh počasí po jednotlivých měsících kalendářního roku, který se vyznačoval značnou variabilitou provázenou suchem, sněhovou kalamitou, bouřkami, vichřicemi, teplotními rekordy i povodněmi.The course of the weather in individual months of the calendar year is presented in the paper. The year 2009 was characterized by considerable variability accompanied by drought, heavy snowfall, storms, windstorms, temperature records or floods.1

Změny ročního chodu srážek v České republice od roku 1961 =Changes in annual cycle of precipitation amount in the Czech Republic since 1961

Roční chod srážek má u nás obvykle maximum v červnu a minimum v únoru. Pro klimatické scénáře aktuálně zkonstruované do roku 2100 je roční chod jedním z důležitých parametrů. V příspěvku je roční chod popsán pro pozorované řady za období 1961-2010 i pro scénáře klimatu 2011-2100.Annual cycle of precipitation amount has usually maximum on June and minimum on February. Climate scenarios recently constructed for the period till 2100 use the annual cycle as one of important parameters. The description of annual cycle is presented in the paper both for the observational period 1961-2010 and for climate scenario 2011-2100.10410

Počasí v České republice v roce 2012 =Weather in the Czech Republic in 2012

Článek shrnuje průběh počasí a popis hlavních událostí v jednotlivých měsících roku 2012. Nejdůležitějšími událostmi roku byl studený začátek února s minimálními teplotami téměř -40°C a nové absolutní maximum teploty pro Českou republiku dosažené 20. srpna. Průměrná roční teplota byla mírně nadnormální, 1°C nad dlouhodobým průměrem a roční úhrn srážek normální (jen 17 mm pod dlouhodobým průměrem).Both the course of the weather and description of main events in individual months of the calendar are presented in the paper. The main events of the year 2012 were cold beginning of February with minimum temperature near -40 °C and new absolute maximum temperature for Czech Republic achieved on 20th August +40,4 °C. Yearly temperature was more than 1 °C above normal for whole Czech Republic and precipitation amount was normal (just 17 mm below normal).1
