Ultrafast dynamics of neutral superexcited Oxygen: A direct measurement of the competition between autoionization and predissociation

Using ultrafast extreme ultraviolet pulses, we performed a direct measurement of the relaxation dynamics of neutral superexcited states corresponding to the nl\sigma_g(c^4\Sigma_u^-) Rydberg series of O_2. An XUV attosecond pulse train was used to create a temporally localized Rydberg wavepacket and the ensuing electronic and nuclear dynamics were probed using a time-delayed femtosecond near-infrared pulse. We investigated the competing predissociation and autoionization mechanisms for superexcited molecules and found that autoionization is dominant for the low n Rydberg states. We measured an autoionization lifetime of 92+/-6 fs and 180+/-10 fs for (5s,4d)\sigma_g and (6s,5d)\sigma_g Rydberg state groups respectively. We determine that the disputed neutral dissociation lifetime for the \nu=0 vibrational level of the Rydberg series is 1100+/-100fs.Comment: 5 pages, 4 figure

Apoptosis in neurodegenerative diseases: to be or not to be? Absence of proof is not proof of absence

Apoptosis (from Greek falling off) is a term coined by Kerr, Wyllie and Currie in 1972 to describe a form of cell death associated with peculiar morphological changes. They contrasted apoptosis with necrosis, in which large numbers of cells undergo destruction and elicit a regional inflammatory response. In contrast, in apoptosis individual cells die and are being removed quickly, without inflammation, making their demise often difficult to detect. The initial concept of apoptosis was exclusively related to a morphological phenomenon.Biomedical Reviews 1995; 4: 103-108

Muscular diacylglycerol metabolism and insulin resistance

Failure of insulin to elicit an increase in glucose uptake and metabolism in target tissues such as skeletal muscle is a major characteristic of non-insulin dependent type 2 diabetes mellitus. A strong correlation between intramyocellular triacylglycerol concentrations and the severity of insulin resistance has been found and led to the assumption that lipid oversupply to skeletal muscle contributes to reduced insulin action. However, the molecular mechanism that links intramyocellular lipid content with the generation of muscle insulin resistance is still unclear. It appears unlikely that the neutral lipid metabolite triacylglycerol directly impairs insulin action. Hence it is believed that intermediates in fatty acid metabolism, such as fatty acyl-CoA, ceramides or diacylglycerol (DAG) link fat deposition in the muscle to compromised insulin signaling. DAG is identified as a potential mediator of lipid-induced insulin resistance, as increased DAG levels are associated with protein kinase C activation and a reduction in both insulin-stimulated IRS-1 tyrosine phosphorylation and PI3 kinase activity. As DAG is an intermediate in the synthesis of triacylglycerol from fatty acids and glycerol, its level can be lowered by either improving the oxidation of cellular fatty acids or by accelerating the incorporation of fatty acids into triacylglycerol. This review discusses the evidence that implicates DAG being central in the development of muscular insulin resistance. Furthermore, we will discuss if and how modulation of skeletal muscle DAG levels could function as a possible therapeutic target for the treatment of type 2 diabetes mellitus

Versatile silicon-waveguide supercontinuum for coherent mid-infrared spectroscopy

Infrared spectroscopy is a powerful tool for basic and applied science. The molecular spectral fingerprints in the 3 um to 20 um region provide a means to uniquely identify molecular structure for fundamental spectroscopy, atmospheric chemistry, trace and hazardous gas detection, and biological microscopy. Driven by such applications, the development of low-noise, coherent laser sources with broad, tunable coverage is a topic of great interest. Laser frequency combs possess a unique combination of precisely defined spectral lines and broad bandwidth that can enable the above-mentioned applications. Here, we leverage robust fabrication and geometrical dispersion engineering of silicon nanophotonic waveguides for coherent frequency comb generation spanning 70 THz in the mid-infrared (2.5 um to 6.2 um). Precise waveguide fabrication provides significant spectral broadening and engineered spectra targeted at specific mid-infrared bands. We use this coherent light source for dual-comb spectroscopy at 5 um.Comment: 26 pages, 5 figure

The Role of Visual Feedback and Creative Exploration for the Improvement of Timing Accuracy in Performing Musical Ornaments

    Get PDF
IN DEVELOPING A VISUAL FEEDBACK SYSTEM FOR A CREATIVE activity such as music performance, the objective is not just to reinforce one particular manner of performing. Instead, a desirable characteristic might be that the visual feedback enhances flexibility and originality, in addition to contributing to performance precision. In an experimental study focused on the timing of a musical ornament, we examined whether the instruction to explore ornament timing in training trials with or without visual feedback leads to improved temporal precision in imitating target performances of the ornament, and whether visual feedback enhances the performance diversity during training. The study uncovered distinct strategies of exploration of the performance of the musical ornament and highlighted the dynamics of exploration behavior during training. Visual feedback enhanced exploration of temporal characteristics and influenced imitation accuracy. This study opens up educational possibilities for the training of performance skills and provides direction for further investigation of creative processes in performance

Mutational analysis of BTAF1-TBP interaction: BTAF1 can rescue DNA-binding defective TBP mutants

    Get PDF
The BTAF1 transcription factor interacts with TATA-binding protein (TBP) to form the B-TFIID complex, which is involved in RNA polymerase II transcription. Here, we present an extensive mapping study of TBP residues involved in BTAF1 interaction. This shows that residues in the concave, DNA-binding surface of TBP are important for BTAF1 binding. In addition, BTAF1 interacts with residues in helix 2 on the convex side of TBP as assayed in protein-protein and in DNA-binding assays. BTAF1 drastically changes the TATA-box binding specificity of TBP, as it is able to recruit DNA-binding defective TBP mutants to both TATA-containing and TATA-less DNA. Interestingly, other helix 2 interacting factors, such as TFIIA and NC2, can also stabilize mutant TBP binding to DNA. In contrast, TFIIB which interacts with a distinct surface of TBP does not display this activity. Since many proteins contact helix 2 of TBP, this provides a molecular basis for mutually exclusive TBP interactions and stresses the importance of this structural element for eukaryotic transcription
