    Desmodium mottle virus, the first legumovirus (genus Begomovirus) from East Africa

    A novel bipartite legumovirus (genus Begomovirus, family Geminiviridae), that naturally infects the wild leguminous plant Desmodium sp. in Uganda, was molecularly characterized and named Desmodium mottle virus. The highest nucleotide identities for DNA-A, obtained from two field-collected samples, were 79.9% and 80.1% with the legumovirus, soybean mild mottle virus. DNA-B had the highest nucleotide identities (65.4% and 66.4%) with a typical non-legumovirus Old World begomovirus, African cassava mosaic virus. This is the first report of a legumovirus in East Africa and extends the known diversity of begomoviruses found infecting wild plants in this continent

    Musculoskeletal disorders in the elderly in rheumatology practice in Burkina Faso

    Objective: To study the frequency of musculoskeletal (MSK) disorders in the elderly in Ouagadougou, Burkina Faso. Patients and methods: A retrospective study was conducted from February 2006 to March 2011 in the University Hospital Yalgado Ouédraogo in Burkina Faso. All patients aged over 65 years seen at the Department of Rheumatology were included. Results: Four hundred and twenty-four patients (11.85%) aged over 65 years were identified among the total of 4084. The mean age was 70.15± 23.26 years with a range of 65-95 (mode: 70 years, median 69 years). There were 336 (69.4%) women and 148 men (30.6%). Rheumatic diseases were dominated by osteoarthritis(OA) and degenerative disease: low back pain / sciatica: 117 (24.17%) cases, OA of the knee: 111 (22.93%), tendinitis of the shoulder: 52 (10.74%), several OA (6%). Polymyalgia rheumatica was diagnosed in 15 (3%) patients. Only two cases of Rheumatoid Arthritis (RA) and one case of ankylosing spondylitis were noted. Osteoporosis was reported in three patients. Fifty-five (11.36%) had diabetes mellitus (type 2), 221 (45.66%) had hypertension and 15 (3%) heart disease. Conclusion: The elderly have an important place in rheumatology practice in Ouagadougou. Osteoarthritis and degenerative MSK disorders are common and osteoporosis, chondrocalcinosis and RA rare. Polymyalgia rheumatica was the most common inflammatory disorder.     Effects of sweet potato feathery mottle virus, sweet potato chlorotic stunt virus and their co-infection on sweet potato yield in Western Burkina Faso

    To determine the effects of sweet potato feathery mottle virus (SPFMV), Sweet potato chlorotic stunt virus (SPCSV) and their co-infection on sweet potato yield, twelve sweet potato varieties were assessed in a hotspot area in Western Burkina Faso. The experiment was carried out in a randomized complete-block design with the twelve varieties in three replications. Data were collected on plant growth parameters, plant virus symptoms and yield parameters. Additional testing for selected sweet potato viruses was done using a nitrocellulose membrane enzyme-linked immunosorbent assay (NCM-ELISA) and RT-PCR. SPFMV and SPCSV were the viruses detected in this study. Varieties Djakani and Ligri were virus-free and had the highest average yields out of twelve sweet potato varieties assessed. Field monitoring indicated that 58% of plants were found to be virus-infected. The results suggest that severe symptoms were associated with sweet potato virus disease (SPVD) and yield reduction. However, the interaction of SPCSV with other viruses, which may result in synergistic negative effects on sweet potato yield and quality, needs further research

    Mobile laboratory to improve response to meningitis epidemics, Burkina Faso epidemic season 2004

    A Mobile Laboratory was developed for use primarily during the epidemic meningitis season in Burkina Faso. This report describes the Mobile Laboratory characteristics, its use to date, problems encountered and their resolution, and future directions. During 2004, the mobile laboratory intervention in three remote Burkina Faso districts experiencing meningitis epidemics led to more speciïŹc case management and led directly to vaccination of one district. However, in a second district, the intervention occurred too late to allow vaccination. During 2006, the Mobile Laboratory was used to conduct an emergency carriage study that for the ïŹrst time occurred during the peak of a meningococcal serogroup A epidemic. This information is critical for the design of meningococcal conjugate vaccine schedules and vaccine approaches. During 2004-6, technicians in 11 district laboratories received training by Mobile Laboratory staff. Numerous problems with the initial prototype laboratory were identiïŹed, namely that the solar power cells could not provide enough energy to the refrigerator and incubator to maintain appropriate temperatures and having a single integrated unit required use of a separate vehicle for specimen transport. A second laboratory was developed during 2005-6 that used a generator or local energy source for power and that had a laboratory that could be detached from the vehicle. Currently the main limitation of the Mobile Laboratory is that it has not been integrated into routine Ministry of Health activities, limiting its use both during and between meningitis seasons

    Reconstructing the history of maize streak virus strain : a dispersal to reveal diversification hot spots and its origin in Southern Africa

    Maize streak virus strain A (MSV-A), the causal agent of maize streak disease, is today one of the most serious biotic threats to African food security. Determining where MSV-A originated and how it spread transcontinentally could yield valuable insights into its historical emergence as a crop pathogen. Similarly, determining where the major extant MSV-A lineages arose could identify geographical hot spots of MSV evolution. Here, we use model-based phylogeographic analyses of 353 fully sequenced MSV-A isolates to reconstruct a plausible history of MSV-A movements over the past 150 years. We show that since the probable emergence of MSV-A in southern Africa around 1863, the virus spread transcontinentally at an average rate of 32.5 km/year (95% highest probability density interval, 15.6 to 51.6 km/year). Using distinctive patterns of nucleotide variation caused by 20 unique intra-MSV-A recombination events, we tentatively classified the MSV-A isolates into 24 easily discernible lineages. Despite many of these lineages displaying distinct geographical distributions, it is apparent that almost all have emerged within the past 4 decades from either southern or east-central Africa. Collectively, our results suggest that regular analysis of MSV-A genomes within these diversification hot spots could be used to monitor the emergence of future MSV-A lineages that could affect maize cultivation in Africa

    Begomovirus disease complex: emerging threat to vegetable production systems of West and Central Africa

    Vegetables play a major role in the livelihoods of the rural poor in Africa. Among major constraints to vegetable production worldwide are diseases caused by a group of viruses belonging to the genus Begomovirus, family Geminiviridae. Begomoviruses are plant-infecting viruses, which are transmitted by the whitefly vector Bemisia tabaci and have been known to cause extreme yield reduction in a number of economically important vegetables around the world. Several begomoviruses have been detected infecting vegetable crops in West and Central Africa (WCA). Small single stranded circular molecules, alphasatellites and betasatellites, which are about half the size of their helper begomovirus genome, have also been detected in plants infected by begomoviruses. In WCA, B. tabaci has been associated with suspected begomovirus infections in many vegetable crops and weed species. Sequencing of viral genomes from crops such as okra resulted in the identification of two previously known begomovirus species (Cotton leaf curl Gezira virus and Okra yellow crinkle virus) as well as a new recombinant begomovirus species (Okra leaf curl Cameroon virus), a betasatellite (Cotton leaf curl Gezira betasatellite) and new alphasatellites. Tomato and pepper plants with leaf curling were shown to contain isolates of new begomoviruses, collectively referred to as West African tomato-infecting begomoviruses (WATIBs), new alphasatellites and betasatellites. To study the potential of weeds serving as begomovirus reservoirs, begomoviruses and satellites in the weed Ageratum conyzoides were characterized. Sequence analyses showed that they were infected by isolates of a new begomovirus (Ageratum leaf curl Cameroon virus) that belong to the WATIBs group, a new betasatellite (Ageratum leaf curl Cameroon betasatellite), an alphasatellite and two types of defective recombinants between a begomovirus and an alphasatellite. Putative recombinations were detected in begomovirus genomes for all four plant species studied, indicating that recombination is an important mechanism for their evolution. A close relationship between the begomoviruses infecting pepper and tomato and A. conyzoides and the detection of the same alphasatellite in them support the idea that weeds are important reservoirs for begomoviruses and their satellites. With this high diversity, recombination potential and transmission by B. tabaci, begomoviruses and ssDNA satellites pose a serious threat to crop production in West and Central Africa.This item is part of the UA Faculty Publications collection.
