    Passing the Stewardship Baton

    “Montana is wild, it’s our home, and it’s ours to learn from and care for.” This is the central theme FWP adopted to bring to life Montana WILD—the department’s first conservation education center located in Helena. Housed in a historic building adjacent to Spring Meadow Lake State Park, the facility, the exhibit, and the programs honor the deep connection people have with Montana’s fish and wildlife. Here we tell the story of how we came to have this richness today, how all citizens of the state have a part in this history and a stake in its future, and how through individual action we can achieve great things for ourselves and the future of fish and wildlife. Guided by core beliefs of the department and its hope for an informed and engaged citizenry, the statewide facility serves a variety of interests related to fish and wildlife. The objective for this presentation is to give an overview of what has been accomplished to date with regards to Montana WILD, to illuminate the challenges we face as professionals to help others commit to fish and wildlife, and to offer an inclusive framework for how to engender a compelling stewardship ethic

    This paper evaluates alternative indicators of global economic activity and other market fundamentals in terms of their usefulness for forecasting real oil prices and global petroleum consumption. We find that world industrial production is one of the most useful indicators that has been proposed in the literature. However, by combining measures from a number of di¤erent sources we can do even better. Our analysis results in a new index of global economic conditions and new measures for assessing future tightness of energy demand and expected oil price pressures. We illustrate their usefulness for quantifying the main factors behind the severe contraction of the global economy and the price risks faced by shale oil producers in early 2020

    Proteolysis of SNAP-25 by types E and A botulinal neurotoxins

    Clostridial neurotoxins, tetanus toxin (TeTx) and the seven related but serologically distinct botulinal neurotoxins (BoNT/A to BoNT/G), are potent inhibitors of synaptic vesicle exocytosis in nerve endings. Recently it was reported that the light chains of clostridial neurotoxins act as zinc-dependent metalloproteases which specifically cleave synaptic target proteins such as synaptobrevin/VAMPs, HPC-1/syntaxin (BoNT/C1), and SNAP-25 (BoNT/A). We show here that BoNT/E, like BoNT/A, cleaves SNAP-25, as generated by in vitro translation or by expression in Escherichia coli. BoNT/E cleaves the Arg180-Ile181 bond. This site is different from that of BoNT/A, which cleaves SNAP-25 between the amino acid residues Gln197 and Arg198. These findings further support the view that clostridial neurotoxins have evolved from an ancestral protease recognizing the exocytotic fusion machinery of synaptic vesicles whereby individual toxins target different members of the membrane fusion complex

    Use of long-term microdialysis subcutaneous glucose monitoring in the management of neonatal diabetes - A first case report

    In neonatal diabetes mellitus (NDM), a rare genetic disorder, insulin therapy is required but the management is difficult. Frequent blood glucose determinations are necessary in most cases. Microdialysis subcutaneous glucose monitoring (MSGM) is feasible in neonates and has been proposed to reduce painful blood sampling and blood loss. We have applied long-term MSGM to a small-fordate female newborn with transient NDM. We found a good correlation of subcutaneous and blood glucose concentration over a wide range of values. MSGM enabled a reduction in blood glucose determinations during optimization of intravenous insulin treatment and initiation of continuous subcutaneous insulin infusion. We conclude that long-term MSGM is feasible and may reduce painful blood sampling and blood loss in NDM. Furthermore, long-term MSGM may hold a potential for avoiding hypoglycemic episodes and earlier discharge. Copyright (C) 2006 S. Karger AG, Basel

    Pathfinder first light: alignment, calibration, and commissioning of the LINC-NIRVANA ground-layer adaptive optics subsystem

    We present descriptions of the alignment and calibration tests of the Pathfinder, which achieved first light during our 2013 commissioning campaign at the LBT. The full LINC-NIRVANA instrument is a Fizeau interferometric imager with fringe tracking and 2-layer natural guide star multi-conjugate adaptive optics (MCAO) systems on each eye of the LBT. The MCAO correction for each side is achieved using a ground layer wavefront sensor that drives the LBT adaptive secondary mirror and a mid-high layer wavefront sensor that drives a Xinetics 349 actuator DM conjugated to an altitude of 7.1 km. When the LINC-NIRVANA MCAO system is commissioned, it will be one of only two such systems on an 8-meter telescope and the only such system in the northern hemisphere. In order to mitigate risk, we take a modular approach to commissioning by decoupling and testing the LINC-NIRVANA subsystems individually. The Pathfinder is the ground-layer wavefront sensor for the DX eye of the LBT. It uses 12 pyramid wavefront sensors to optically co-add light from natural guide stars in order to make four pupil images that sense ground layer turbulence. Pathfinder is now the first LINC-NIRVANA subsystem to be fully integrated with the telescope and commissioned on sky. Our 2013 commissioning campaign consisted of 7 runs at the LBT with the tasks of assembly, integration and communication with the LBT telescope control system, alignment to the telescope optical axis, off-sky closed loop AO calibration, and finally closed loop on-sky AO. We present the programmatics of this campaign, along with the novel designs of our alignment scheme and our off-sky calibration test, which lead to the Pathfinder's first on-sky closed loop images
