    Ubiquitin Carboxyl-Terminal Hydrolase L1 in Parkinson’s Disease

    Ubiquitin plays the crucial roles to maintain the ubiquitin proteasome system (UPS) functions, which were suggested that involved in Parkinson’s diease (PD). Ubiquitin C-terminal hydrolase L1 (UCHL1), which was detected in Lewy bodies of nerve cells in PD brains, plays an important role for maintaining ubiquitin pool in UPS. The first UCHL1 mutation (UCHL1I93M) was found in two siblings of a PD family. By contrast, UCHL1S18Y mutation was recognized to reduce the risk of developing PD by its specific antioxidant protective function. The studies of UCHL1 in mouse models showed that lack of UCHL1 resulted in motor ataxia, degeneration of axons, and instability of free ubiquitin level. Transgenic mice expressing UCHL1I93M mutant exhibited dopaminergic neuron (DA) degeneration in MPTP-treated conditions. In this chapter, we provide a summary on recent findings related to roles of UCH-L1 in PD. Knockdown dUCH, a homolog of human UCHL1, in fly dopaminergic neuron resulted as some Parkinson’s disease—like phenotype such as: (1) the underdevelopment and/or degeneration of DA neurons; (2) the shortage of dopamine in the brain; and (3) the locomotor dysfunctions. Those finding indicated that dUCH (ortholog of human UCH-L1 in Drosophila) plays an important role in Parkinson’s disease

    UCH-L1 (ubiquitin carboxyl-terminal hydrolase L1) is a protein, which plays an important role in ubiquitin-proteasome system. Many previous reports showed the relation between UCH-L1 and neurodegenerative diseases, diabetes, as well as cancer. However, the mechanism still remains unclear. In the aim to investigate the functions and regulatory mechanism of UCH-L1 in living organism, Drosophila melanogaster model was utilized to examine the role of UCH-L1. This chapter provides a summary on recent findings related to the roles of UCH-L1 based on the model. First, abnormal expression of Drosophila ubiquitin carboxyl-terminal hydrolase (dUCH) leads to the defects on fly tissue development and function. Gain function of dUCH in the eye imaginal discs induced a rough eye phenotype in the adult, partly resulting from induction of caspase-dependent apoptosis, upset of photoreceptor cell distribution and ommatidium apical mispatterning. Interestingly, the dUCH overexpression of induced rough eye phenotype was completely recused by co-expression either Sevenless or Draf of the mitogen-activated protein kinase pathway. Besides, knockdown dUCH in dopaminergic neurons resulted in some Parkinson’s disease—like phenotypes in fly. Taken together, those findings in Drosophila model contributed a significant dUCH in tissue development and function

    On Building a Community to Exchange and Share Sustainable Fashion Applications

    Sustainable development is a standard plan for peace and prosperity for people and the planet, now and in the future, set forth by the 2030 Agenda for Sustainable Development, which was adopted by all United Nations member states and agreed to in 2015. The Sustainable Development Goals (SDGs) are an urgent call to action for all developed and developing countries in relation to global cooperation. Many factors cause environmental pollution, but the fashion industry is considered by the United Nations Conference on Trade and Development (UNCTAD) as the second most polluting industry in the world. It accounts for 10% of global carbon emissions due to the energy used in production, manufacturing, and transportation. In particular, the factors that strongly affect the environment in the fashion industry are the textile materials, the fabric dyeing industry, and the rapid development of fast fashion. The data was collected through an online survey of 101 fashion consumers and interviews with nine target audiences who need to buy and sell used clothes or are interested in buying and selling used clothes or sustainable fashion. Sustainable design is not a new concept, but it is still quite strange to the Vietnamese. Consumer ignorance about sustainable consumption and the harmful effects of the fashion industry on planet Earth is a big reason why consumers constantly plunge into the shopping vortex. The problem of wasteful and unsustainable fashion consumption is a big global problem at a time when the fashion industry is gradually emerging into the sustainable fashion movement. This is an opportunity to build a community by connecting consumers through their essential needs, helping them raise awareness, change their behavior, and make each individual part of a sustainable fashion community.     Nghiên cứu hoạt tính kháng khuẩn của cao chiết nước từ một số loại cây dùng làm thuốc dân gian tại Vườn Quốc gia Bidoup - núi Bà, tỉnh Lâm Đồng

    Cây thuốc dân gian được người dân tộc tại Vườn Quốc Gia Bidoup – Núi Bà, Lâm Đồng sử dụng để điều trị một số bệnh như: tiêu chảy, viêm, làm lành vết thương …. Sử dụng cây thuốc hiện chủ yếu dựa vào kinh nghiệm nên nghiên cứu này hướng đến việc đánh giá hoạt tính kháng khuẩn của cao chiết nước để làm cơ sở cho các nghiên cứu tiếp theo. Tách chiết cao nước và đánh giá hoạt tính kháng khuẩn bằng phương pháp khuếch tán trên giếng thạch đối với 16 chủng vi khuẩn chỉ thị của 9 cây thuốc dân gian được người dân tộc K’Ho sử dụng để trị bệnh tiêu chảy. Kết quả nghiên cứu cho thấy rằng hiệu suất tách chiết cao nước khá cao (11,99% - 29,52%), đồng thời cao chiết nước từ cây xidra nguôn (Medinilla septentrionalis) ở nồng độ 100 mg/mL có hoạt tính kháng khuẩn mạnh nhất (kháng 16/16 chủng) với đường kính vòng kháng khuẩn từ 9,3 cm – 11,0 cm; 4/9 mẫu cao chiết có hoạt tính yếu (kháng được từ 1 chủng đến 6/16 chủng vi khuẩn chỉ thị) và 4/9 mẫu cao chiết nước không kháng khuẩn. Kết quả xác định sơ bộ thành phần hóa học của cao chiết nước có tính kháng khuẩn cho thấy có sự hiện diện của alkaloid, steroid, saponin, tannin và flavonoid. Kết quả cũng cho thấy rằng nước có thể được sử dụng để tách chiết cao nhưng không phải tất cả các cao chiết nước đều có hoạt tính tốt, do đó cần phải có các nghiên cứu chuyên sâu hơn

    Detailed investigation of factors affecting the synthesis of SiO2@Au for the enhancement of Raman spectroscopy

    The reaction time, temperature, ratio of precursors, and concentration of sodium citrate are known as the main factors that affect the direct synthesis process of SiO2@Au based on the chemical reaction of HAuCl4 and sodium citrate. Hence, we investigated, in detail, and observed that these factors played a crucial role in determining the shape and size of synthesized nanoparticles. The significant enhancement of the SERS signal corresponding to the fabrication conditions is an existing challenge. Our study results show that the optimal reaction conditions for the fabrication of SiO2@Au are a 1:21 ratio of HAuCl4 to sodium citrate, with an initial concentration of sodium citrate of 4.2 mM, and a reaction time lasting longer than 6 h at a temperature of 80 degrees C. Under optimal conditions, our synthesis process result is SiO2@Au nanoparticles with a diameter of approximately 350 nm. In particular, the considerable enhancement of Raman intensities of SiO2@Au compared to SiO2 particles was examined.Web of Science1217art. no. 308

    Isolation an agar degradation Bacillus sp. AT6 and preliminary application for seaweed saccharification

    Background: Agar is a common polysaccharide found in nature. However, agar is strongly resisted to the degradation processing, leading to limitation of its application in various areas. Thus, finding an effective solution for agar saccharification significantly improves the economically effects of agar based substrates.Methods: Soil samples were collected from TienPhong Forestry Ltd. Company, ThuyXuan District, ThuaThien Hue province, Vietnam. Potential agar degrading bacteria were screened on a mineral salt agar medium. The isolate was identified based on 16S rRNA nucleotide sequence, morphological, physiological, and biochemical characteristics. Agarase production was evaluated by modification culture conditions including incubation time, shaking speed, and initial inoculum size. Molecular mass of extracellular agarase was determined by native SDS-PAGE. The effect of pH, temperature, metal ions, and organic solvents were conducted for enzyme characterization. Application of enzyme was investigated on seaweed saccharification.Result: An agar degrading bacterial strain was isolated from soils and identified as Bacillus sp. AT6. Maximal agarase accumulation obtained in the culture containing an inoculum size of 10% (v/v), shaking speed of 210 rpm, and 96 hours incubation. The agarase revealed a single band on zymogram analysis with an apparent molecular weight of 180 kDa. The optimal temperature and pH were 40°C and pH 8.0, respectively. All tested metal ions and organic solvents partially decreased enzyme activity. Treatment seaweed by agarase resulted in reducing sugars release present in the reaction, indicating the saccharification of seaweed was succeeded.Conclusion: Bacillus sp. AT6 is a new report of agarolytic bacteria that produces extracellular agarase enzymes. The present results promise strain AT6 is a great candidate for agar saccharification for industrial application

    DETERMINATION THE PRIORITY OF FACTORS INFLUENCING OVERSEAS INVESTMENT DESTINATIONS OF BUSINESSES USING AN ANALYTIC HIERARCHY PROCESS METHOD

    Because of the great benefits overseas investment brings, countries and businesses are increasingly concerned with the question of an ideal investment destination. Many studies have shown that, factors of the consumption market, the infrastructure and resources, economic and political environment have a significant impact on the decision to choose an investment location, however the priority order of these factors are different for various research spaces. By studying the specific case of Viettel Group - one of the largest telecommunications groups in Vietnam that is having many international investment activities in recent years, the study has been verified the priority order of factors influencing the decision to choose an overseas investment location of this corporation.     Clinical Outcomes of Patients With Drug-Resistant Tuberculous Meningitis Treated With an Intensified Antituberculosis Regimen.

    Drug-resistant tuberculous meningitis (TBM) is difficult to diagnose and treat. Mortality is high and optimal treatment is unknown. We compared clinical outcomes of drug-resistant and -susceptible TBM treated with either standard or intensified antituberculosis treatment. We analyzed the influence of Mycobacterium tuberculosis drug resistance on the outcomes of patients with TBM enrolled into a randomized controlled trial comparing a standard, 9-month antituberculosis regimen (containing rifampicin 10 mg/kg/day) with an intensified regimen with higher-dose rifampicin (15 mg/kg/day) and levofloxacin (20 mg/kg/day) for the first 8 weeks. The primary endpoint of the trial was 9-month survival. In this subgroup analysis, resistance categories were predefined as multidrug resistant (MDR), isoniazid resistant, rifampicin susceptible (INH-R), and susceptible to rifampicin and isoniazid (INH-S + RIF-S). Outcome by resistance categories and response to intensified treatment were compared and estimated by Cox regression. Of 817 randomized patients, 322 had a known drug resistance profile. INH-R was found in 86 (26.7%) patients, MDR in 15 (4.7%) patients, rifampicin monoresistance in 1 patient (0.3%), and INH-S + RIF-S in 220 (68.3%) patients. Multivariable regression showed that MDR (hazard ratio [HR], 5.91 [95% confidence interval {CI}, 3.00-11.6]), P < .001), was an independent predictor of death. INH-R had a significant association with the combined outcome of new neurological events or death (HR, 1.58 [95% CI, 1.11-2.23]). Adjusted Cox regression, corrected for treatment adjustments, showed that intensified treatment was significantly associated with improved survival (HR, 0.34 [95% CI, .15-.76], P = .01) in INH-R TBM. Early intensified treatment improved survival in patients with INH-R TBM. Targeted regimens for drug-resistant TBM should be further explored

    Evaluation of the Luminex xTAG Respiratory Viral Panel FAST v2 assay for detection of multiple respiratory viral pathogens in nasal and throat swabs in Vietnam.

    BACKGROUND: Acute respiratory infections (ARI) are among the leading causes of hospitalization in children ≤5 years old. Rapid diagnostics of viral pathogens is essential to avoid unnecessary antibiotic treatment, thereby slowing down antibiotic-resistance. We evaluated the diagnostic performance of the Luminex xTAG Respiratory Viral Panel FAST v2 against viral specific PCR as reference assays for ARI in Vietnam. METHODS: Four hundred and forty two nose and throat swabs were collected in viral transport medium, and were tested with Luminex xTAG Respiratory Viral Panel FAST v2. Multiplex RT-PCR and single RT-PCR were used as references.    Results: Overall, viral pathogens were detected in a total count of 270/294 (91.8%, 95% CI 88.1-94.7) by the Luminex among reference assays, whilst 112/6336 (1.8%, 95% CI, 1.4-2.1) of pathogens were detected by the Luminex, but not by reference assays. Frequency of pathogens detected by Luminex and reference assays was 379 and 292, respectively. The diagnostic yield was 66.7% (295/442, 95%CI 62.1-71.1%) for the Luminex assay and 54.1% (239/442, 95% CI, 49.3-58.8%) for reference assays. The Luminex kit had higher yields for all viruses except influenza B virus, respiratory syncytial virus, and human bocavirus. High agreements between both methods [mean (range): 0.91 (0.83-1.00)] were found for 10/15 viral agents. CONCLUSIONS: The Luminex assay is a high throughput multiplex platform for rapid detection of common viral pathogens causing ARI. Although the current high cost may prevent Luminex assays from being widely used, especially in limited resource settings where ARI are felt most, its introduction in clinical diagnostics may help reduce unnecessary use of antibiotic prescription

    Viral Metagenomic Analysis of Cerebrospinal Fluid from Patients with Acute Central Nervous System Infections of Unknown Origin, Vietnam.

    Central nervous system (CNS) infection is a serious neurologic condition, although the etiology remains unknown in >50% of patients. We used metagenomic next-generation sequencing to detect viruses in 204 cerebrospinal fluid (CSF) samples from patients with acute CNS infection who were enrolled from Vietnam hospitals during 2012-2016. We detected 8 viral species in 107/204 (52.4%) of CSF samples. After virus-specific PCR confirmation, the detection rate was lowered to 30/204 (14.7%). Enteroviruses were the most common viruses detected (n = 23), followed by hepatitis B virus (3), HIV (2), molluscum contagiosum virus (1), and gemycircularvirus (1). Analysis of enterovirus sequences revealed the predominance of echovirus 30 (9). Phylogenetically, the echovirus 30 strains belonged to genogroup V and VIIb. Our results expanded knowledge about the clinical burden of enterovirus in Vietnam and underscore the challenges of identifying a plausible viral pathogen in CSF of patients with CNS infections
