Análise in silico de proteínas quinases expressas em Coffea arabica.
Apesar da indiscutível importância agronômica do café, particularmente para o Brasil, esta cultura ainda é alvo de diferentes estresses que causam perda em sua produtividade. A resposta do cafeeiro aos diferentes sinais como patógenos e de condições ambientais adversas é ainda desconhecida. Com o objetivo de identificar proteínas quinases, o banco de seqüências expressas de Café (CAFEST) foi investigado. A análise do banco de dados CafEST revelou 479 seqüências identificadas como quinases que formaram um conjunto de 153 unigenes. As proteínas codificadas pelos segmentos expressos correspondem a diferentes e importantes categorias de quinases (MAPK, MAPKK, MAPKKK, receptores, etc). A análise filogenética mostrou a conservação das proteínas expressas no cafeeiro comparado a outras plantas e fungos. Os presentes resultados indicam que a resposta a estresses seja conservada em cafeeiro, provavelmente mediada pelos mecanismos de fosforilação em cascata. Além disso, apontam para o aprofundamento dos estudos destas vias como contribuição aos fenômenos relacionados à interação planta-patógeno
Evaluation of Microencapsulation of The UFV-AREG1 Bacteriophage in Alginate-Ca Microcapsules using Microfluidic Devices
The indiscriminate use of antibiotics and the emergence of resistant
microorganisms have become a major challenge for the food industry. The purpose
of this work was to microencapsulate the bacteriophage UFV-AREG1 in a calcium
alginate matrix using microfluidic devices and to study the viability and
efficiency of retention. The microcapsules were added to gel of propylene
glycol for use as an antimicrobial in the food industry. The technique showed
the number of the phage encapsulation, yielding drops with an average 100-250
m of diameter, 82.1 2% retention efficiency and stability in the gel
matrix for 21 days. The gel added to the microencapsulated phage showed
efficiency (not detectable on the surface) in reducing bacterial contamination
on the surface at a similar level to antimicrobial chemicals (alcohol 70%).
Therefore, it was possible to microencapsulate bacteriophages in alginate-Ca
and apply the microcapsules in gels for use as sanitizers in the food industry.Comment: 8 pages, 5 figure

Host adaptation to viruses relies on few genes with different cross-resistance properties

However, the genetics of these direct and correlated responses remains poorly studied. The overlap between these responses is instrumental for the understanding of host evolution in multiparasite environments. We determined the genetic and phenotypic changes underlying adaptation of Drosophila melanogaster to Drosophila C virus (DCV). Within 20 generations, flies selected with DCV showed increased survival after DCV infection, but also after cricket paralysis virus (CrPV) and flock house virus (FHV) infection. Whole-genome sequencing identified two regions of significant differentiation among treatments, from which candidate genes were functionally tested with RNAi. Three genes were validated--pastrel, a known DCV-response gene, and two other loci, Ubc-E2H and CG8492. Knockdown of Ubc-E2H and pastrel also led to increased sensitivity to CrPV, whereas knockdown of CG8492 increased susceptibility to FHV infection.
