    Improved synchronous production of Plasmodium falciparum gametocytes in vitro.

    The sexual stages of the Plasmodium falciparum life cycle are attractive targets for vaccines and transmission blocking drugs. Difficulties in culturing and obtaining large amounts of sexual stage P. falciparum parasites, particularly early stages, have often limited research progress in this area. We present a new protocol which simplifies the process of stimulating gametocytogenesis leading to improved synchronous gametocyte production. This new method can be adapted to enrich for early stage gametocytes (I and II) with a higher degree of purity than has previously been achieved, using MACS magnetic affinity columns. The protocol described lends itself to large scale culturing and harvesting of synchronous parasites suitable for biochemical assays, northern blots, flow cytometry, microarrays and proteomic analysis

    Surface and subsurface water contributions during snowmelt in a small Precambrian Shield watershed

    ABSTRACT The rates and pathways of snowmelt runoff on Precambrian Shield watersheds are not fully understood. The results of a detailed field study and numerical analysis of these processes are presented for a small watershed located in Muskoka , Ontario, that has been gauged both chemically and physically since 1976 by the Ontario Ministry of the Environmentas a part of its Acid Precipitation in Ontario Study. Field data show that in many areas soils with very high hydraulic conductivities contributed signifcant amounts of subsurface flow to the hydrograph during and shortly after melt. Quickflow in the first phase of snowmelt was generated by a combination of saturation overland flow and saturated-unsaturated subsurface storm flow from shallow side slopes; however, most of the total water volume was delivered via slower ground-water flow. In the final phase of the melt period, the streamflow was mainly due to ground-water flow recharged on aforested area and delivered over limited distances through shallow, highly permeable overburden materials. Quickjow peaks superimposed on this general trend could be accounted for mainly as surface runoff over groundwater effuent areas mapped throughout the watershed. In support of ongoing hydrochemical studies by the Ontario Ministry of the Environment, these data and interpretations wil aid in modifing and improving hydrologic submodels by adding to our understanding of water movement in acidifed watersheds.     The effect of hydrogen peroxide on uranium oxide films on 316L stainless steel

    For the first time the effect of hydrogen peroxide on the dissolution of electrodeposited uranium oxide films on 316L stainless steel planchets (acting as simulant uranium-contaminated metal surfaces) has been studied. Analysis of the H2O2-mediated film dissolution processes via open circuit potentiometry, alpha counting and SEM/EDX imaging has shown that in near-neutral solutions of pH 6.1 and at [H2O2] 0.1 mol dm(-3) the uranium oxide film, again in analogy to common corrosion processes, behaves as if in a transpassive state and begins to dissolve. This transition from passive to transpassive behaviour in the effect of peroxide concentration on UO2 films has not hitherto been observed or explored, either in terms of corrosion processes or otherwise. Through consideration of thermodynamic solubility product and complex formation constant data, we attribute the transition to the formation of soluble uranyl-peroxide complexes under mildly alkaline, high [H2O2] conditions - a conclusion that has implications for the design of both acid minimal, metal ion oxidant-free decontamination strategies with low secondary waste arisings, and single step processes for spent nuclear fuel dissolution such as the Carbonate-based Oxidative Leaching (COL) process.     Modelling rotational failure in confined geometries using DLO

    Discontinuity layout optimisation (DLO) is a generally applicable numerical limit analysis procedure that can be used to identify critical plastic collapse mechanisms in engineering problems. Considering the modelling of in-plane failure, the authors have previously presented a formulation capable of identifying rotational failure mechanisms in non-dilating media. However, the formulation presented did not explicitly address cases involving confined geometries, where curved slip lines could potentially intersect boundaries. In this paper, methods are outlined which permit efficient modelling of such cases. Details of the kinematic and equilibrium formulations are provided, which are then verified through application to various geotechnical and structural mechanics problems. It is shown that results of high accuracy can be obtained, both in terms of the predicted collapse load and the corresponding failure mechanism

    Cellular plasticity facilitates phenotypic change in a dominant coral’s Symbiodiniaceae assemblage

    Coral-associated dinoflagellates (Symbiodiniaceae) are photosynthetic endosymbionts that influence coral acclimation, as indicated by photo-endosymbiotic phenotypic variance across different environmental conditions. Symbiont shuffling (shifts in endosymbiont community composition), changes in endosymbiont cell density, and cellular plasticity have all been proposed as acclimation mechanisms. However, few studies have been able to partition which of the three strategies were responsible for observed phenotypic variance. Using a combination of metabarcoding and flow cytometry, we simultaneously characterized Acropora pulchra-associated Symbiodiniaceae assemblages at the community, population, and individual level under natural environmental conditions to deduce whether seasonal phenotypic change and site-related phenotypic variation of Symbiodiniaceae assemblages is a product of symbiont shuffling or cellular plasticity. Symbiodiniaceae assemblages displayed season-specific phenotypic variance, while Symbiodiniaceae community composition was geographically structured and cell density showed limited data structure. Based on these patterns, we reveal that cellular plasticity of Symbiodiniaceae was the source of a phenotypic variation, thus indicating that cellular plasticity is a mechanism for acclimation to mild environmental change

    Simulation of Neptunium Extraction in an Advanced PUREX Process — Model Improvement

    Routing neptunium to a single product in spent nuclear fuel reprocessing is a significant challenge. In this work, we have further improved the simulation of neptunium extraction in an advanced PUREX flowsheet by applying a revised model of the Np(V)-Np(VI) redox reaction kinetics, a new nitric acid radiolysis model and by evaluating various models for the nitrous acid distribution coefficient. The Np disproportionation reaction is shown to have a negligible effect. The models are validated against published ‘cold test’ experimental results; the ‘hot test’ simulation suggests high neptunium radiolysis could help to achieve high recoveries using this flowsheet

    The magnitude of educational disadvantage amongst indigenous minority groups in Australia.

    Indigenous groups are amongst the most disadvantaged minority groups in the developed world. This paper examines the educational disadvantage of indigenous Australians by assessing academic performance at a relatively early age. We find that, by the age of 10, indigenous Australians are substantially behind non-indigenous Australians in academic achievement. Their relative performance deteriorates further over the next 2 years. School and locality do not appear to be important determinants of the indigenous to non-indigenous achievement gap. However, geographic remoteness, indigenous ethnicity and language use at home have a marked influence on educational achievement. A current focus of Australian indigenous policy is to increase school resources. Our results suggest that this will not eliminate indigenous educational disadvantage on its own
