    More than 100 million animals every year suffer and die by fierce chemical test, medical, food, and cosmetic by giving poisonous, blinding and killing million animals every year for irresponsible companies. In medical world, all procedures that are done against the animals called animal testing.Animal testing happens in Indonesia against civet cat animal. The animal eats coffee fruit and digests it to be coffee fruit seed that is put out with its feces. This seed that has unique taste and high price so that there is attraction for the coffee producers to produce civet cats coffee seed on a large scale.Hence, the problem is coffee producer method by doing animal testing against civet cat where the process can cause civet cat extinction. Therefore, PETA is one of environmental organization that tries to prevent animal testing method by focusing on main attention with the term of four areas.This research uses anilities descriptive method that explains a thing based on analyze and the data is collected and proved the truth. This research also uses deductive method that the conclusion is made general-specifically.     Enforcement of developmental lineage specificity by transcription factor Oct1

    Embryonic stem cells co-express Oct4 and Oct1, a related protein with similar DNA-binding specificity. To study the role of Oct1 in ESC pluripotency and transcriptional control, we constructed germline and inducible-conditional Oct1-deficient ESC lines. ESCs lacking Oct1 show normal appearance, self-renewal and growth but manifest defects upon differentiation. They fail to form beating cardiomyocytes, generate neurons poorly, form small, poorly differentiated teratomas, and cannot generate chimeric mice. Upon RA-mediated differentiation, Oct1-deficient cells induce lineage-appropriate developmentally poised genes poorly while lineage-inappropriate genes, including extra-embryonic genes, are aberrantly expressed. In ESCs, Oct1 co-occupies a specific set of targets with Oct4, but does not occupy differentially expressed developmental targets. Instead, Oct1 occupies these targets as cells differentiate and Oct4 declines. These results identify a dynamic interplay between Oct1 and Oct4, in particular during the critical window immediately after loss of pluripotency when cells make the earliest developmental fate decisions

    POU2F1 activity regulates HOXD10 and HOXD11 promoting a proliferative and invasive phenotype in head and neck cancer

    HOX genes are master regulators of organ morphogenesis and cell differentiation during embryonic development, and continue to be expressed throughout post-natal life. To test the hypothesis that HOX genes are dysregulated in head and neck squamous cell carcinoma (HNSCC) we defined their expression profile, and investigated the function, transcriptional regulation and clinical relevance of a subset of highly expressed HOXD genes. Two HOXD genes, D10 and D11, showed strikingly high levels in HNSCC cell lines, patient tumor samples and publicly available datasets. Knockdown of HOXD10 in HNSCC cells caused decreased proliferation and invasion, whereas knockdown of HOXD11 reduced only invasion. POU2F1 consensus sequences were identified in the 5’ DNA of HOXD10 and D11. Knockdown of POU2F1 significantly reduced expression of HOXD10 and D11 and inhibited HNSCC proliferation. Luciferase reporter constructs of the HOXD10 and D11 promoters confirmed that POU2F1 consensus binding sites are required for optimal promoter activity. Utilizing patient tumor samples a significant association was found between immunohistochemical staining of HOXD10 and both the overall and the disease-specific survival, adding further support that HOXD10 is dysregulated in head and neck cancer. Additional studies are now warranted to fully evaluate HOXD10 as a prognostic tool in head and neck cancers

Kamadjaja Logistics yang bergelut di dunia jasa pengeiriman barang dan gudang tidak sedikit mengalami kendala rusak atau pecah barang, sehingga barang yang rusak atau pecah harus diclaim kepihak pengirim. Dalam pengklaiman barang yang rusak atau pecah harus memiliki data yang lengkap dan data-data penunjang yang bisa meyakinkan pihak pengirim agar barang yang rusak atau pecah dapat di claim. Tujuan dari penelitian ini adalah untuk memperoleh data yang diperlukan dalam rangka membangun sistem informasi penyelesaian claim produk Unilever pada PT. Kamadjaja Logistics Samarinda dan untuk memperoleh gambaran sistem yang sedang berjalan dan memastikan bahwa sistem yang akan dibuat jauh lebih baik dan dapat menunjang kinerja. Adapun keguanaan dari penelitian ini adalah untuk merancang dan membangun sistem informasi penyelesaian claim produk Unilever pada PT. Kamadjaja Logistics Samarinda, dan untuk menyusun sistem basis data sistem informasi penyelesaian claim produk pada PT. Kamadjaja Logistics Samarinda ke PT. Unilever. Dengan adanya sistem informasi penyelesaian claim produk Unilever pada PT. Kamadjaja Logistics Samarinda ini, maka dalam  pembuatan Quality Incident Report menjadi lebih efektif dan efisien dan data-data penunjang terpenuhi sehingga claim dapat terlaksana

    Road infrastructure has a function as one of the supporters of the development of an area. The Road infrastructure still used today is a flexible pavement composed of a mixture of asphalt and aggregate, which is heated and then compacted. Asphalt serves as a binder in the flexible pavement to maintain density and resist the weathering of aggregates caused by weather. It is an innovative additive used in concrete waste to replace rough aggregates. Adding LGA B50/30 in a base mixture is expected to be an alternative to improving the quality of the base mix and supporting the government in the use of domestic products. The study aims to determine how marshall is characterized if concrete waste is used in the AC-BC Ashburton mix as a substitute for rough aggregates and whether it meets the requirements of the Binna Marga Specification. The study was conducted experimentally with marshal characteristic parameters density, VIM (Void In Mix), VMA (Void In Mineral Aggregate), VFA (Void Filled with Asphalt), stability, and Marshall Quotient. (MQ). The Optimum Asphalt Rate used is 6.5%. The result is the density decreases with the addition of concrete waste levels, with the highest value at the 30% concrete waste level of 2,408 gr/cc. The increase in the VIM value with the addition of the concrete residue level has a higher value at a rate of 70% or 4.66%. The highest MQ is at a 50% concrete waste rate of 334.04 kg/mm. The study results show that waste concrete innovation materials can be used in Asbuton AC-BC mixtures with levels not exceeding 60%. Based on this research, concrete waste can be an alternative to natural aggregate substitutes

    Analysis Index of Retained Strenght on AC-WC Asphalt Mixture with the Addition of Cigarette Filter Waste as an Alternative for Asphalt Modification

    In order to provide an alternative asphalt mixture that is more durable, various efforts have been made to improve the quality of asphalt, therefore research is needed on polymer modified asphalt by replacing some of the asphalt with cigarette filter waste. In this study, cigarette filter waste was substituted into asphalt with proportions of 2%, 5%, and 8%. The purpose of this study was to determine the percentage of cigarette filter waste that affects the AC-WC mixture based on marshall characteristics and residual stability. The optimum asphalt content (KAO) used was 5.1%, with the results showing that the addition of cigarette filter waste affected the marshall characteristic values, namely the specific gravity value, Voids Filled with Asphalt (VFA), and the highest stability was at a variation of 2%, while the Voids in Mix (VIM), Void in Mineral Aggregate (VMA), and the highest flow. were at a variation of 8%. The residual stability index test showed that water affected the durability of the AC-WC mixture, as evidenced by the highest value in the 24-hour immersion variation with a cigarette filter content of 2% which decreased with increasing immersion time. The addition of cigarette filter waste into the AC-WC mixture has met the Bina Marga Specifications in the Marshall characteristic test, but the change in the residual stability index was only at a soaking time of 24 hours, while for other periods it did not comply with the Bina Marga specifications

    Diabetes mellitus is a systemic disease characterized by insulin deficiency. Common symptoms in people with diabetes is elevated blood glucose is called hyperglycemia. Manifestations in the oral cavity are inflammation of gingival tissue or periodontitis. The bacteria that play a role in the severity of periodontal disease is A. actinomycetemcomitans. The purpose of this study was to determine the relationship of blood glucose levels with the growth of A. actinomycetemcomitans in DM rats treated with insulin therapy. Methode 20 three month old male Wistar rats with weight of 170-200 grams were divided into two groups. First, periodontitis and DM were manipulated into all groups through aloksan injection with dose of 170 mg/kg. Those rats in group I were treated with insulin, those in group II were without treatment. In the third and seventh weeks, the blood glucose levels and the number of A. actinomycetemcomitans was measured. The data was tested by using Pearson test. The results showed that there is a strong relationship between blood glucose levels with the growth of A. actinomycetemcomitans. Conclusion: The increasing blood glucose levels are very strongly associated with an increase in the number of A. actinomycetemcomitans and otherwise in DM rats with insulin therapy

    Inhibition activity of Robusta coffee beans polyphenol extract on the production of TNF-α neutrophil cells

    Polyphenols are one of the active substances in the Robusta coffee beans with various benefits for humans’ health including anti-inflammation. neutrophil cell (polymorphonuclear PMN) plays a significant role as the primary immune response against foreign agent. Inflammatory response is characterized by the production of pro-inflammatory cytokines such as tumor necrosis factor-α (TNF-α). Hence, the purpose of this study is to determine anti-inflammation capacity of Robusta coffee beans polyphenols extract on the TNF-α production in neutrophil cells. Neutrophil was derived from human peripheral venous blood by means of gradient density method. One hundred microliter of neutrophil  (1,5x103 cell) was incubated with 3.13%, 6.25%, 12.5%, 25% polyphenol extract and exposed with 100 μl of Lipopolysaccharides (LPS) 25 mg/ml. The production of TNF-α were determined by ELISA. Data were analyzed by one way Anova. Polyphenols of Robusta coffee bean extract was shown to inhibit TNF-α production in LPS-exposed neutrophil cells compared to controls. Whereas, LPS-exposed neutrophil increased TNF-α production. The most effective concentration to inhibit TNF-α production was 12.5%. It was concluded that polyphenols of Robusta coffee beans has anti-inflammatory properties as indicated by its ability to decrease TNF-α levels

    Fusobacterium nucleatum is a bacteria found in normal flora of the oral cavity that plays a role in the occurrence of periodontal disease. The prevalence of periodontal disease in Indonesia reaches 70%. One treatment by using medicinal plants is robusta coffee beans. Robusta coffee beans contain volatile acids, chlorogenic acids, caffeine acids, phenols and caffeine suspected to be antibacterial. The aim was to determine the inhibition of coffee robusta extract on growth of Fusobacterium nucleatum. This study used disc diffusion method with 4 samples in each study group. The study group consisted of 6 treatment groups (12,5%, 25%, 50% and 100% coffee robusta extract), positive control group (chlorhexidine gluconate 0,2%), and negative control group (sterile aquades). Data were analyzed using One Way Anova test and LSD (Least Significant Difference) test. Coffee robusta extract has the ability to inhibit the growth of Fusobacterium nucleatum. The concentration of coffe robusta extract 100%, 50%, 25%, 12,5% have same ability to inhibit the growth of Fusobacterium nucleatum

    Kawasan Kalurahan Melikan termasuk dalam kawasan Pegunungan Sewu. Kawasan ini termasuk dalam kawasan lindung nasional dan menjadi bagian dari Global Geopark Network UNESCO sejak 2015. Sebagaimana kawasan karst lainnya, kawasan ini memiliki potensi sumber daya alam, sumber daya manusia, dan sosial ekonomi budaya. Penelitian ini memetakan seluruh potesi tersebut menjadi dasar dalam perancangan Kawasan Taman Budaya Banglipuran yang merupakan fokus pengembangan kawasan Kalurahan Melikan. Pendekatan yang digunakan dalam penelitian ini adalah action research yang dilaksanakan dalam tiga tahap. Tahap penelitian meliputi pemetaan, perencanaan dan pengembangan. Pemetaan digital menggunakan Quantum GIS. Perencanaan dilaksanakan dengan FGD. Dan pengembangan meliputi perncangan kawasan dengan visual 3D.
