    Expression and processing of fluorescent fusion proteins of amyloid precursor protein (APP)

    AbstractProcessing of β-amyloid precursor protein (APP) by β- and γ-secretases in neurons produces amyloid-β (Aβ), whose excess accumulation leads to Alzheimer's disease (AD). Knowledge on subcellular trafficking pathways of APP and its fragments is important for the understanding of AD pathogenesis. We designed fusion proteins comprising a C-terminal fragment of APP (app) and fluorescent proteins GFP (G) and DsRed (D) to permit the tracking of the fusion proteins and fragments in cells. CAD cells expressing these proteins emitted colocalized green and red fluorescence and produce ectodomains, sGapp and sRapp, and Aβ, whose level was reduced by inhibitors of β- and γ-secretases. The presence of GappR in endosomes was observed via colocalization with Rab5. These observations indicated that the fusion proteins were membrane inserted, transported in vesicles and proteolytically processed by the same mechanism for APP. By attenuating fusion protein synthesis with cycloheximide, individual fluorescent colors from the C-terminus of the fusion proteins appeared in the cytosol which was strongly suppressed by β-secretase inhibitor, suggesting that the ectodomains exit the cell rapidly (t1/2 about 20min) while the C-terminal fragments were retained longer in cells. In live cells, we observed the fluorescence of the ectodomains located between parental fusion proteins and plasma membrane, suggesting that these ectodomain positions are part of their secretion pathway. Our results indicate that the native ectodomain does not play a decisive role for the key features of APP trafficking and processing and the new fusion proteins may lead to novel insights in intracellular activities of APP

    Construction of a doxycycline inducible adipogenic lentiviral expression system

    To provide a tool for research on regulating adipocyte differentiation, tetracycline inducible (Tet on) lentiviral expression vectors under the control of an adipose-specific promoter were constructed. The lowest basal expression in the absence of doxycycline and most efficient dose-dependent, doxycycline-induced transient overexpression was observed using vectors constructed with a combination of Tetracycline Responsive Element (TRE) and reverse tetracycline-controlled TransActivator advanced (rtTAadv), transfected in white (3T3-L1) and brown (HIB-1B) preadipocytes cell lines. The results demonstrate that doxycycline adipogenic inducible expression can be achieved using a pLenti TRE / rtTA adv under the control of the truncated aP2 promoter in HIB-1B preadipocytes

    Studies of Prototype CsI(Tl) Crystal Scintillators for Low-Energy Neutrino Experiments

    Crystal scintillators provide potential merits for the pursuit of low-energy low-background experiments. A CsI(Tl) scintillating crystal detector is being constructed to study low-energy neutrino physics at a nuclear reactor, while projects are underway to adopt this technique for dark matter searches. The choice of the geometrical parameters of the crystal modules, as well as the optimization of the read-out scheme, are the results of an R&D program. Crystals with 40 cm in length were developed. The detector requirements and the achieved performance of the prototypes are presented. Future prospects for this technique are discussed.Comment: 32 pages, 14 figure

    Health and social care service utilisation and associated expenditure among community-dwelling older adults with depressive symptoms

    AIMS: Late-life depression has substantial impacts on individuals, families and society. Knowledge gaps remain in estimating the economic impacts associated with late-life depression by symptom severity, which has implications for resource prioritisation and research design (such as in modelling). This study examined the incremental health and social care expenditure of depressive symptoms by severity. METHODS: We analysed data collected from 2707 older adults aged 60 years and over in Hong Kong. The Patient Health Questionnaire-9 (PHQ-9) and the Client Service Receipt Inventory were used, respectively, to measure depressive symptoms and service utilisation as a basis for calculating care expenditure. Two-part models were used to estimate the incremental expenditure associated with symptom severity over 1 year. RESULTS: The average PHQ-9 score was 6.3 (standard deviation, s.d. = 4.0). The percentages of respondents with mild, moderate and moderately severe symptoms and non-depressed were 51.8%, 13.5%, 3.7% and 31.0%, respectively. Overall, the moderately severe group generated the largest average incremental expenditure (US5886;955886; 95% CI 1126-10 647 or a 272% increase), followed by the mild group (US3849; 95% CI 2520-5177 or a 176% increase) and the moderate group (US1843;951843; 95% CI 854-2831, or 85% increase). Non-psychiatric healthcare was the main cost component in a mild symptom group, after controlling for other chronic conditions and covariates. The average incremental association between PHQ-9 score and overall care expenditure peaked at PHQ-9 score of 4 (US691; 95% CI 444-939), then gradually fell to negative between scores of 12 (US35;95 - 35; 95% CI - 530 to 460) and 19 (US -171; 95% CI - 417 to 76) and soared to positive and rebounded at the score of 23 (US$601; 95% CI -1652 to 2854). CONCLUSIONS: The association between depressive symptoms and care expenditure is stronger among older adults with mild and moderately severe symptoms. Older adults with the same symptom severity have different care utilisation and expenditure patterns. Non-psychiatric healthcare is the major cost element. These findings inform ways to optimise policy efforts to improve the financial sustainability of health and long-term care systems, including the involvement of primary care physicians and other geriatric healthcare providers in preventing and treating depression among older adults and related budgeting and accounting issues across services

    Inquiry web-based learning to enhance information problem solving competences in science

    Early research on using web information indicates that secondary students fail to explore much web tools, use them naively and have serious difficulties to understand and integrate web information. In response to these challenges, the main goal of this research has been to design, implement and evaluate an instructional approach that helps students learn from web information. We have developed on-line learning materials which focus on specific curricular contents and provide specific scaffolds to help students accomplish web-based tasks and develop specific information problem-solving competencies. These scaffolds have intended to give support to students involved in information-seeking activities as they were asked questions, searched for information, organised and assessed their findings, and created rich representations of their newly-constructed understandings. We have designed a one year long study to investigate the depth and accuracy of 127 secondary students, as regards their content understanding as well as their development of information problem-solving competencies when using on-line resources to solve instructional tasks. Our research demonstrates that the experimental group performed computer-based activities statistically better than the control group. Our findings also suggest that students were able to develop accurate and in-depth understanding from web information if they could appropriately use search and managerial strategies. This research lends evidence to questions regarding the value of students engaging in on-line inquiry web-based learning to enhance content understanding and to develop more efficient information problem-solving competencies in secondary education

    Surveillance of emerging drugs of abuse in Hong Kong: Validation of an analytical tool

Objective: To validate a locally developed chromatography-based method to monitor emerging drugs of abuse whilst performing regular drug testing in abusers. Design: Cross-sectional study. Setting: Eleven regional hospitals, seven social service units, and a tertiary level clinical toxicology laboratory in Hong Kong. Participants: A total of 972 drug abusers and high-risk individuals were recruited from acute, rehabilitation, and high-risk settings between 1 November 2011 and 31 July 2013. A subset of the participants was of South Asian ethnicity. In total, 2000 urine or hair specimens were collected. Main outcome measures: Proof of concept that surveillance of emerging drugs of abuse can be performed whilst conducting routine drug of abuse testing in patients. Results: The method was successfully applied to 2000 samples with three emerging drugs of abuse detected in five samples: PMMA (paramethoxymethamphetamine), TFMPP [1-(3-trifluoromethylphenyl)piperazine], and methcathinone. The method also detected conventional drugs of abuse, with codeine, methadone, heroin, methamphetamine, and ketamine being the most frequently detected drugs. Other findings included the observation that South Asians had significantly higher rates of using opiates such as heroin, methadone, and codeine; and that ketamine and cocaine had significantly higher detection rates in acute subjects compared with the rehabilitation population. Conclusions: This locally developed analytical method is a valid tool for simultaneous surveillance of emerging drugs of abuse and routine drug monitoring of patients at minimal additional cost and effort.     Measurements of the Mass and Full-Width of the ηc\eta_c Meson

    In a sample of 58 million J/ψJ/\psi events collected with the BES II detector, the process J/ψγηc\psi\to\gamma\eta_c is observed in five different decay channels: γK+Kπ+π\gamma K^+K^-\pi^+\pi^-, γπ+ππ+π\gamma\pi^+\pi^-\pi^+\pi^-, γK±KS0π\gamma K^\pm K^0_S \pi^\mp (with KS0π+πK^0_S\to\pi^+\pi^-), γϕϕ\gamma \phi\phi (with ϕK+K\phi\to K^+K^-) and γppˉ\gamma p\bar{p}. From a combined fit of all five channels, we determine the mass and full-width of ηc\eta_c to be mηc=2977.5±1.0(stat.)±1.2(syst.)m_{\eta_c}=2977.5\pm1.0 ({stat.})\pm1.2 ({syst.}) MeV/c2c^2 and Γηc=17.0±3.7(stat.)±7.4(syst.)\Gamma_{\eta_c} = 17.0\pm3.7 ({stat.})\pm7.4 ({syst.}) MeV/c2c^2.Comment: 9 pages, 2 figures and 4 table.     Direct Measurements of the Branching Fractions for D0Ke+νeD^0 \to K^-e^+\nu_e and D0πe+νeD^0 \to \pi^-e^+\nu_e and Determinations of the Form Factors f+K(0)f_{+}^{K}(0) and f+π(0)f^{\pi}_{+}(0)

    The absolute branching fractions for the decays D0Ke+νeD^0 \to K^-e ^+\nu_e and D0πe+νeD^0 \to \pi^-e^+\nu_e are determined using 7584±198±3417584\pm 198 \pm 341 singly tagged Dˉ0\bar D^0 sample from the data collected around 3.773 GeV with the BES-II detector at the BEPC. In the system recoiling against the singly tagged Dˉ0\bar D^0 meson, 104.0±10.9104.0\pm 10.9 events for D0Ke+νeD^0 \to K^-e ^+\nu_e and 9.0±3.69.0 \pm 3.6 events for D0πe+νeD^0 \to \pi^-e^+\nu_e decays are observed. Those yield the absolute branching fractions to be BF(D0Ke+νe)=(3.82±0.40±0.27)BF(D^0 \to K^-e^+\nu_e)=(3.82 \pm 0.40\pm 0.27)% and BF(D0πe+νe)=(0.33±0.13±0.03)BF(D^0 \to \pi^-e^+\nu_e)=(0.33 \pm 0.13\pm 0.03)%. The vector form factors are determined to be f+K(0)=0.78±0.04±0.03|f^K_+(0)| = 0.78 \pm 0.04 \pm 0.03 and f+π(0)=0.73±0.14±0.06|f^{\pi}_+(0)| = 0.73 \pm 0.14 \pm 0.06. The ratio of the two form factors is measured to be f+π(0)/f+K(0)=0.93±0.19±0.07|f^{\pi}_+(0)/f^K_+(0)|= 0.93 \pm 0.19 \pm 0.07.Comment: 6 pages, 5 figure
