    First Two-Micron Imaging Polarimetry of Beta Pictoris

    High-resolution K band imaging polarimetry of the beta Pic dust disk has been conducted with adaptive optics and a coronagraph using the Subaru 8.2-m telescope. Polarization of ~10 % is detected out to r ~ 120 AU with a centro-symmetric vector pattern around the central star, confirming that the disk is seen as an infrared reflection nebula. We have modeled our near-infrared and previous optical polarization results in terms of dust scattering in the disk and have found that both the degrees of polarization and the radial intensity profiles are well reproduced. We argue that the observed characteristics of the disk dust are consistent with the presence of ice-filled fluffy aggregates consisting of submicron grains in the beta Pic system. There is a gap around 100 AU in both the intensity and polarization profiles, which suggests a paucity of planetesimals in this region. The radial intensity profile also shows ripple-like structures, which are indicative of the presence of multiple planetesimal belts, as in the case of the M-type Vega-like star AU Mic.Comment: 18 pages, 6 figures, accepted for Jp

    Long-duration nano-second single pulse lasers for observation of spectra from bulk liquids at high hydrostatic pressures

    AbstractThe influence of laser pulse duration on the spectral emissions observed from bulk ionic solutions has been investigated for hydrostatic pressures between 0.1 and 30MPa. Transient pressure, shadowgraph imaging and spectroscopic measurements were performed for single pulses of duration 20 and 150ns. The transient pressure measurements show that for hydrostatic pressures up to 30MPa, propagation of the high-pressure shockwave generated by the focused laser causes the local pressure to reduce below ambient levels during the time frame that spectroscopic measurements can be made. The pressure impulse and subsequent reduction in pressure are larger, with the latter lasting longer for the 150ns pulse compared to a 20ns pulse of the same energy. The 150ns pulse generates larger cavities with significant enhancement of the spectral emissions observed compared to the 20ns duration pulse for pressures up to 30MPa. The results demonstrate that laser-induced breakdown using a long ns duration pulse offers an advantage over conventional, short ns duration pulses for the analysis of bulk ionic solutions at hydrostatic pressures between 0.1 and 30MPa

    Elimination of Specific miRNAs by Naked 14-nt sgRNAs

    tRNase ZL-utilizing efficacious gene silencing (TRUE gene silencing) is a newly developed technology to suppress mammalian gene expression. TRUE gene silencing works on the basis of a unique enzymatic property of mammalian tRNase ZL, which is that it can recognize a pre-tRNA-like or micro-pre-tRNA-like complex formed between target RNA and artificial small guide RNA (sgRNA) and can cleave any target RNA at any desired site. There are four types of sgRNA, 5′-half-tRNA, RNA heptamer, hook RNA, and ∼14-nt linear RNA. Here we show that a 14-nt linear-type sgRNA against human miR-16 can guide tRNase ZL cleavage of miR-16 in vitro and can downregulate the miR-16 level in HEK293 cells. We also demonstrate that the 14-nt sgRNA can be efficiently taken up without any transfection reagents by living cells and can exist stably in there for at least 24 hours. The naked 14-nt sgRNA significantly reduced the miR-16 level in HEK293 and HL60 cells. Three other naked 14-nt sgRNAs against miR-142-3p, miR-206, and miR-19a/b are also shown to downregulate the respective miRNA levels in various mammalian cell lines. Our observations suggest that in general we can eliminate a specific cellular miRNA at least by ∼50% by using a naked 14-nt sgRNA on the basis of TRUE gene silencing

    Development of a deep-sea laser-induced breakdown spectrometer for in situ multi-element chemical analysis

    Spectroscopy is emerging as a technique that can expand the envelope of modern oceanographic sensors. The selectivity of spectroscopic techniques enables a single instrument to measure multiple components of the marine environment and can form the basis for versatile tools to perform in situ geochemical analysis. We have developed a deep-sea laser-induced breakdown spectrometer (ChemiCam) and successfully deployed the instrument from a remotely operated vehicle (ROV) to perform in situ multi-element analysis of both seawater and mineral deposits at depths of over 1000 m. The instrument consists of a long-nanosecond duration pulse-laser, a spectrometer and a high-speed camera. Power supply, instrument control and signal telemetry are provided through a ROV tether. The instrument has two modes of operation. In the first mode, the laser is focused directly into seawater and spectroscopic measurements of seawater composition are performed. In the second mode, a fiber-optic cable assembly is used to make spectroscopic measurements of mineral deposits. In this mode the laser is fired through a 4 m long fiber-optic cable and is focused onto the target’s surface using an optical head and a linear stage that can be held by a ROV manipulator. In this paper, we describe the instrument and the methods developed to process its measurements. Exemplary measurements of both seawater and mineral deposits made during deployments of the device at an active hydrothermal vent field in the Okinawa trough are presented. Through integration with platforms such as underwater vehicles, drilling systems and subsea observatories, it is hoped that this technology can contribute to more efficient scientific surveys of the deep-sea environment

    Tumor stem cell assay for detecting metastases of human lung cancer.

    We applied a tumor stem cell assay using an enriched double-layered soft agar system for the detection of metastatic sites of lung cancer. Lung cancer colonies grew from 7 of 10 effusions cytologically positive for tumor cells and 7 of 10 bone marrow aspirates cytologically and histologically positive for tumor cells. Twenty-six of 29 bone marrow aspirates cytologically and histologically negative for tumor cells showed no colony growth. However, the remaining three bone marrow aspirates, which were obtained from patients with small cell lung cancer, formed colonies in soft agar. These results indicate that the tumor stem cell assay is useful for detecting metastatic sites of lung cancer.</p

    Astrometry of Water Maser Sources in Nearby Molecular Clouds with VERA - II. SVS 13 in NGC 1333

    We report on the results of multi-epoch VLBI observations with VERA (VLBI Exploration of Radio Astrometry) of the 22 GHz H2O masers associated with the young stellar object SVS 13 in the NGC 1333 region. We have carried out phase-referencing VLBI astrometry and measured an annual parallax of the maser features in SVS 13 of 4.25+/-0.32 mas, corresponding to the distance of 235+/-18 pc from the Sun. Our result is consistent with a photometric distance of 220 pc previously reported. Even though the maser features were detectable only for 6 months, the present results provide the distance to NGC 1333 with much higher accuracy than photometric methods. The absolute positions and proper motions have been derived, revealing that the H2O masers with the LSR (local standard of rest) velocities of 7-8 km s-1 are most likely associated with VLA4A, which is a radio counterpart of SVS 13. The origin of the observed proper motions of the maser features are currently difficult to attribute to either the jet or the rotating circumstellar disk associated with VLA4A, which should be investigated through future high-resolution astrometric observations of VLA4A and other radio sources in NGC 1333.Comment: 9 pages, 5 figures.     Cytological and histological correlation of primary lung cancer: a preliminary study of 106 cases with resectable tumors.

    In order to increase the accuracy of diagnosis in lung cancer, analysis concerning cytological and histological correlation was attempted. The present study consists of 106 patients, who were seen during the past approximately five years and underwent radical surgery to remove tumors completely; mere biopsy specimens were excluded. These patients were 63 years old on the average, 78 males and 28 females, 29 cases of the hilar type (H) and 77 of the peripheral type (P), and 27 and 76 cases of the clinical stage I in H and P, respectively. Histologically, there were 53 adenocarcinomas (Ad), 38 squamous cell carcinomas (Sq), 4 adenosquamous cell carcinomas (Ad + Sq), 5 large cell carcinomas (LCC), and 6 small cell carcinomas (SCC); among them, 3 Ad and 21 Sq in H, and 50 Ad and 17 Sq in P. The overall positive percentages were 65.5 (H) and 26.0 (P) by combination of spontaneous, airsol-induced and Saccomanno's methods, against 96.6 (H) and 72.8 (P) with inclusion of brushing method. 94.8% of Sq in H and 66.7% of Ad and 70.6% of Sq in P were positive by the brushing. A comparative study of these four methods, performed at least once on the same patient, also confirmed the superiority of brushing. Cyto- and histological agreement was 21/21 (100%) for Sq in H, whereas 30/34 (88.2%) for Ad and 13/15 (86.7%) for Sq in P. In conclusion, cyto- and histological findings in H and P corresponded well, and as far as cytology of peripheral type is concerned, a combined method, especially with brushing, is strongly recommended.</p

    Astrometry of Galactic Star Forming Region Sharpless 269 with VERA : Parallax Measurements and Constraint on Outer Rotation Curve

    We have performed high-precision astrometry of H2O maser sources in Galactic star forming region Sharpless 269 (S269) with VERA. We have successfully detected a trigonometric parallax of 189+/-8 micro-arcsec, corresponding to the source distance of 5.28 +0.24/-0.22 kpc. This is the smallest parallax ever measured, and the first one detected beyond 5 kpc. The source distance as well as proper motions are used to constrain the outer rotation curve of the Galaxy, demonstrating that the difference of rotation velocities at the Sun and at S269 (which is 13.1 kpc away from the Galaxy's center) is less than 3%. This gives the strongest constraint on the flatness of the outer rotation curve and provides a direct confirmation on the existence of large amount of dark matter in the Galaxy's outer disk.Comment: 7 pages and 4 figures, Accepted by PASJ (Vol. 59, No. 5, October 25, 2007 issue
