    QuantEYE: The Quantum Optics Instrument for OWL

    QuantEYE is designed to be the highest time-resolution instrument on ESO:s planned Overwhelmingly Large Telescope, devised to explore astrophysical variability on microsecond and nanosecond scales, down to the quantum-optical limit. Expected phenomena include instabilities of photon-gas bubbles in accretion flows, p-mode oscillations in neutron stars, and quantum-optical photon bunching in time. Precise timescales are both variable and unknown, and studies must be of photon-stream statistics, e.g., their power spectra or autocorrelations. Such functions increase with the square of the intensity, implying an enormously increased sensitivity at the largest telescopes. QuantEYE covers the optical, and its design involves an array of photon-counting avalanche-diode detectors, each viewing one segment of the OWL entrance pupil.     Mycobacterium tuberculosis Drives Expansion of Low-Density Neutrophils Equipped With Regulatory Activities

    In human tuberculosis (TB) neutrophils represent the most commonly infected phagocyte but their role in protection and pathology is highly contradictory. Moreover, a subset of low-density neutrophils (LDNs) has been identified in TB, but their functions remain unclear. Here, we have analyzed total neutrophils and their low-density and normal-density (NDNs) subsets in patients with active TB disease, in terms of frequency, phenotype, functional features, and gene expression signature. Full-blood counts from Healthy Donors (H.D.), Latent TB infected, active TB, and cured TB patients were performed. Frequency, phenotype, burst activity, and suppressor T cell activity of the two different subsets were assessed by flow cytometry while NETosis and phagocytosis were evaluated by confocal microscopy. Expression analysis was performed by using the semi-quantitative RT-PCR array technology. Elevated numbers of total neutrophils and a high neutrophil/lymphocyte ratio distinguished patients with active TB from all the other groups. PBMCs of patients with active TB disease contained elevated percentages of LDNs compared with those of H.D., with an increased expression of CD66b, CD33, CD15, and CD16 compared to NDNs. Transcriptomic analysis of LDNs and NDNs purified from the peripheral blood of TB patients identified 12 genes differentially expressed: CCL5, CCR5, CD4, IL10, LYZ, and STAT4 were upregulated, while CXCL8, IFNAR1, NFKB1A, STAT1, TICAM1, and TNF were downregulated in LDNs, as compared to NDNs. Differently than NDNs, LDNs failed to phagocyte live Mycobacterium tuberculosis (M. tuberculosis) bacilli, to make oxidative burst and NETosis, but caused significant suppression of antigen-specific and polyclonal T cell proliferation which was partially mediated by IL-10. These insights add a little dowel of knowledge in understanding the pathogenesis of human TB

    The molecular size continuum of soil organic phosphorus and its chemical associations

    The chemical nature of most organic P (Porg) in soil remains ‘unresolved’ but is accounted for by a broad signal in the phosphomonoester region of solution 31P nuclear magnetic resonance (NMR) spectra. The molecular size range of this broad NMR signal and its molecular structure remain unclear. The aim of this study was to elucidate the chemical nature of Porg with increasing molecular size in soil extracts combining size exclusion chromatography (SEC) with solution 31P NMR spectroscopy. Gel-filtration SEC was carried out on NaOH-EDTA extracts of four soils (range 238-1135 mg Porg/kgsoil) to collect fractions with molecular sizes of 70 kDa. These were then analysed by NMR spectroscopy. Organic P was detected across the entire molecular size continuum from 70 kDa. Concentrations of Porg in the >10kDa fraction ranged from 107 to 427 mg P/kgsoil and exhibited on average three to four broad signals in the phosphomonoester region of NMR spectra. These broad signals were most prominent in the 10-20 and 20-50 kDa fractions, accounting for on average 77 % and 74 % of total phosphomonoesters, respectively. Our study demonstrates that the broad signal is present in all investigated molecular size fractions and comprises on average three to four components of varying NMR peak line width (20 to 250 Hz). The stereoisomers myo- and scyllo-inositol hexakisphosphates (IP6) were also present across multiple molecular size ranges but were predominant in the 5-10 kDa fraction. The proportion of IP associated with large molecular size fractions >10 kDa was on average 23 % (SD=39 %) of total IP across all soils. These findings suggest that stabilisation of IP in soil includes processes associated with the organic phase

    Bioleaching of Valuable Elements from Red Mud: A Study on the Potential of Non-Enriched Biomass

    Red mud (RM) is the main residue produced by the alkaline extraction of alumina (Al2O3) from bauxite, and it contains valuable metals such as iron (Fe), aluminium (Al), titanium (Ti), magnesium (Mg), manganese (Mn), rare earth elements, etc. This research aimed to investigate the biologically induced leaching of some valuable elements from raw RM without preliminary biomass enrichment and inoculum, simultaneously reducing RM polluting potential and extracting metals for their subsequent recovery within a circular economy-based approach. In addition to the missing inoculum, such an approach is challenging since high RM alkalinity and pH, as well as the absence of any sulphides, constrain the use of the most common biohydrometallurgical techniques. Red Muds from two European locations were tested (RM-I and RM-II, respectively). Bioleaching tests were performed at different temperatures (T = 22 & DEG;C and 28 & DEG;C; and also 15 & DEG;C for RM-II) and solid-to-liquid ratios (S/L = 2%, 5%). A sudden drop in pH from alkaline to constant neutral/acidic values was observed in almost all tests, and such results were attributed to biological activity since abiotic tests did not show any pH decrease. The best results in terms of extraction were achieved with RM-II, in particular for Al, Mg and Mn (17%, 42% and 47%, respectively). At 2% S/L, the highest temperature allowed for a better metal release, while at 5% S/L, the highest extraction of Al, Mg and Ti was observed at 22 & DEG;C. As expected, iron was less available to leach at the achieved pH values, as it was mostly present as hematite in both RMs. Finally, the first microbiological characterisation of the autochthonous biomass selected during the bioleaching treatment of RM was provided

    Lignin biodegradation in pulp-and-paper mill wastewater by selected white rot fungi

    An investigation has been carried out to explore the lignin-degrading ability of white rot fungi, as B. adusta and P. crysosporium, grown in different media containing (i) glucose and mineral salts; (ii) a dairy residue; (iii) a dairy residue and mineral salts. Both fungi were then used as inoculum to treat synthetic and industrial pulp-and-paper mill wastewater. On synthetic wastewater, up to 97% and 74% of lignin degradation by B. adusta and P. crysosporium, respectively, have been reached. On industrial wastewater, both fungal strains were able to accomplish 100% delignification in 8-10 days, independent from pH control, with a significant reduction of total organic carbon (TOC) of the solution. Results have confirmed the great biotechnological potential of both B. adusta and P. crysosporium for complete lignin removal in industrial wastewater, and can open the way to next industrial applications on large scale

    Impacts of Anthropogenic Pollutants on Benthic Prokaryotic Communities in Mediterranean Touristic Ports

    Ports and marinas are central nodes in transport network and play a strategic role in coastal development. They receive pollution from land-based sources, marine traffic and port infrastructures on one side and constitute a potential pollution source for the adjacent coastal areas on the other. The aim of the present study was to evaluate the effects of organic and inorganic co-contamination on the prokaryotic communities in sediments from three Mediterranean ports. The structure and composition of the bacterial and archaeal communities were assessed by targeted metagenomic analysis of the 16S rRNA gene, and the links of prokaryotic communities with environmental and pollution variables were investigated. The harbors presented pronounced site-specificity in the environmental properties and pollution status. Consistently, the structure of archaeal and bacterial communities in surface sediments exhibited a strong spatial variation among the three investigated ports. On the contrary, a wide overlap in composition of prokaryotic assemblages among sites was found, but local variation in the community composition and loss of prokaryotic diversity was highlighted in a heavily impacted port sector near a shipyard. We provided evidences that organic matter, metals and PAHs as well as temperature and salinity play a strong role in structuring benthic bacterial communities significantly contributing to the understanding of their responses to anthropogenic perturbations in marine coastal areas. Among metals, copper was recognized as strongly associated with the observed changes in bacterial assemblages. Overall, this study provides the first assessment of the effects exerted by multiple organic and inorganic contaminations on benthic prokaryotes in ports over a large spatial scale and designates bacterial community as a candidate tool for the monitoring of the sediment quality status in harbors

    New bioassays reveal susceptibility of stone-fruit rootstocks to capnodis tenebrionis larvae

    Larvae of Capnodis tenebrionis (L.) (Coleoptera Buprestidae) feed and develop in roots of stone-fruit trees, thereby decreasing their efficiency, which can lead to plant death. The control of these larvae is critical, due to their localization in the root, and the management of this pest is focused on adults, mainly by using non-specific synthetic insecticides. Less susceptible Prunus rootstocks might be applied as a preventative management of larval infestation by this pest. The current research investigated the susceptibility to C. tenebrionis larvae of the most commonly used rootstocks by combining two bio-assays during two-year trials: development of larvae assayed on semi-artificial substrates containing rootstock bark flour; infestation by neonate larvae on rootstock twigs. The rearing assay on semi-artificial substrates made it possible to distinguish (1) a rootstock cluster (Montclar and GF677) in which larvae developed faster and heavier and produced larger adults, (2) a cluster (Adesoto, CAB6P, Colt and MaxMa60) in which larval growth was less efficient as well as adult size, and (3) a cluster (Garnem and Myrabolan 29C) with intermediate responses in larval development and adult size. The twig infestation assay by neonates showed the most infested (Colt) and least infested (Barrier, MaxMa60 and Marianna 26) rootstocks. When the results of both assays are combined, GF677 and Myrabolan 29C appear more susceptible, while Adesoto and MaxMa60 less susceptible to C. tenebrionis larvae, although Barrier and Marianna 26 require further investigation. The experimental model applied in the current trials can enable processing of a large number of tests on different rootstocks, thereby allowing the accumulation of a large quantity of data on the potential susceptibility of rootstocks. The possibility of rearing larvae on a substrate can allow comparison of additional compounds that could interact with larval growth

    Technical results, clinical efficacy and predictors of outcome of intercostal arteries embolization for hemothorax: A two-institutions’ experience

    Background: To evaluate the clinical efficacy and identify the predictors of outcome of intercostal arterial embolization for hemothorax caused by intercostal artery (ICA) injuries. Methods: A retrospective multi-institutional study was conducted. Outcomes were analyzed in 30 consecutive patients presenting with hemothorax caused by active ICA hemorrhage undergoing transcatheter arterial embolization (TAE). Clinical and procedural parameters were compared between outcomes groups. Results: Overall technical success rate was 87% (n=26). Among the 4 failed cases, 2 underwent repeated TAE and 2 underwent additional surgery. Overall 30-day mortality rate was 23%. Low haemoglobin levels and haematocrit, hepatic comorbidities and more than one artery undergoing embolization increased technical failure rate significantly. Survival was poorer in patients with massive bleeding. Conclusions: ICA embolization was found to be a safe and effective method in treating hemothorax caused by active ICA haemorrhage. Careful pre-embolization evaluation may be required for patient with low haemoglobin levels and haematocrit, hepatic comorbidities and active haemorrhage from more than one artery

    An innovative speech-based user interface for smarthomes and IoT solutions to help people with speech and motor disabilities

    A better use of the increasing functional capabilities of home automation systems and Internet of Things (IoT) devices to support the needs of users with disability, is the subject of a research project currently conducted by Area Ausili (Assistive Technology Area), a department of Polo Tecnologico Regionale Corte Roncati of the Local Health Trust of Bologna (Italy), in collaboration with AIAS Ausilioteca Assistive Technology (AT) Team. The main aim of the project is to develop experimental low cost systems for environmental control through simplified and accessible user interfaces. Many of the activities are focused on automatic speech recognition and are developed in the framework of the CloudCAST project. In this paper we report on the first technical achievements of the project and discuss future possible developments and applications within and outside CloudCAST

    Alcohol from tropical fruit residues

    Foram estudados os rendimentos em álcool de cascas de frutos tropicais (banana, abacaxi e tangerina) em extratos contendo açúcares fermentáveis obtidos mediante extração aquosa a quente e em solução ácida diluída (6,7%) e concentrada (35% e 41%). Na fermentação - feita em escala de laboratório - empregou-se a levedura Saccharomyces cerevisiae. O aproveitamento dos açúcares variou de 96% a 100% no caso da extração em água quente, sendo estes valores inferiores na extração com ácido clorídrico diluído. Foi estimada uma média de 12,6 l de álcool por t de casca de banana, de 68,7 l/t de casca de tangerina e de 100,6 l por t de casca de abacaxi, nas extrações com água quente. Estes valores caíram, na extração com ácido diluído, para 12 l/t no caso de banana, 10 l/t no abacaxi, e 26 l/t no da casca de tangerina. Num processo de extração combinado de água com ácido concentrado foram obtidos rendimentos da ordem de 13 l/t (abacaxi), 9,5 l/t (tangerina) e 7,8 l/t (banana). O rendimento em álcool foi de 72,1% para banana, 98,2% para abacaxi e 98,0% para tangerina no caso de extração com água quente. Estes valores foram reduzidos para 59%, 34% e 43%, respectivamente, na extração ácida diluída e no processo combinado, para 64,3%, 32,9% e 65,1% na mesma ordem. Alcohol production from peel extracts of tropical fruits (banana, pineapple and mandarin oranges) having fermentable sugars extracted with hot water and HCl solutions (6.7% and a combined 35% and 41%) was studied. For the fermentation of the extract the yeast Saccharomyces cerevisiae was used. The sugar utilization varied from 96% to 100% for hot water extraction being this value lower for the HCl extractions. Alcohol productions were 12.6 liters per ton in the case of banana, 68.7 liters per ton for mandarins, and 100.6 liters per ton for pineapple, for the hot water extractions. These values dropped to 12 liters per ton in banana, 10 liters per ton in pineapple and 26 liters per ton in orange peels for the diluted acid extraction. For an extraction process using a combination of hot water and concentrated acid, yields were 13 l/ton for pineapple, 9.5 l/ton for mandarin and 7.8 l/ton for banana peels. The alcoholic yields were estimated as 72.1% for banana, 98.2% for pineapple and 98.0% for mandarins, for the hot water extraction. These values were reduced to 59%, 34% and 43%, respectively, in the diluted acid extraction, and to 64.3%, 32.9% and 65.1% in the same order, for the combined process.
