El silenciamiento genético en célulasde Schwann del gen Nae1 (llamado NAe1 cKO), la enzima que inicia la cascada de neddyaltion, causanotables defectos en los nervios periféricos mostrando síntomas similares a los observados en neuropatías graves. Estos defectos incluyen: la reducción de la velocidad de conducción nerviosa, la ausencia de mielina en los nervios periféricos y la degradación de la mielina en células de Schwann . Al mismo tiempo, pudimos detectar en los ratones silenciados grandes cambios en el perfil transcriptómico y proteómico de las células de Schwann. Estos resultados sugieren que la actividad de las Cullin RingLigases (CRL) podría estar afectada, lo que podría estar impidiendo la degradación de reguladores negativos de la mielinización como c-Jun y Sox2. Por otro lado, no fuimos capaces de relacionar directamente la neddylation con el mantenimiento de las vainas de mielina en ratones adultos ni con papel de las células de Schwann en la regeneración de nervios periféricos en respuesta a daño en los mismosCICbioGUN

Amino Functionalized Micro-Mesoporous Hybrid Particles for the Sustained Release of the Antiretroviral Drug Tenofovir

© 2020 by the authors.The sustained release of an antiretroviral agent to women mucosa has been proved as an excellent strategy to reduce the sexual transmission of HIV. Hybrid micro-mesoporous particles have been synthesized and functionalized with a silane coupling agent followed by loading the antiretroviral tenofovir. It has been observed that the disposition of the silane molecule on the surface of the particles determines the interaction mechanism with the antiretroviral molecule loaded independently on the surface area of the particles. In this sense, available and free amino groups are required to achieve a smart pH-responsive material, a condition that is only achieved in those materials containing a silane chemisorbed monolayer. Moreover, the modulation of the release kinetics attributed to the presence of the silane monolayer covering the mesopores has been confirmed by fitting the releasing curves to the first order and Weibull models. Identification of factors associated with diagnostic error in primary care

Background Missed, delayed or incorrect diagnoses are considered to be diagnostic errors. The aim of this paper is to describe the methodology of a study to analyse cognitive aspects of the process by which primary care (PC) physicians diagnose dyspnoea. It examines the possible links between the use of heuristics, suboptimal cognitive acts and diagnostic errors, using Reason’s taxonomy of human error (slips, lapses, mistakes and violations). The influence of situational factors (professional experience, perceived overwork and fatigue) is also analysed.Methods Cohort study of new episodes of dyspnoea in patients receiving care from family physicians and residents at PC centres in Granada (Spain). With an initial expected diagnostic error rate of 20%, and a sampling error of 3%, 384 episodes of dyspnoea are calculated to be required. In addition to filling out the electronic medical record of the patients attended, each physician fills out 2 specially designed questionnaires about the diagnostic process performed in each case of dyspnoea. The first questionnaire includes questions on the physician’s initial diagnostic impression, the 3 most likely diagnoses (in order of likelihood), and the diagnosis reached after the initial medical history and physical examination. It also includes items on the physicians’ perceived overwork and fatigue during patient care. The second questionnaire records the confirmed diagnosis once it is reached. The complete diagnostic process is peer-reviewed to identify and classify the diagnostic errors. The possible use of heuristics of representativeness, availability, and anchoring and adjustment in each diagnostic process is also analysed. Each audit is reviewed with the physician responsible for the diagnostic process. Finally, logistic regression models are used to determine if there are differences in the diagnostic error variables based on the heuristics identified.Discussion This work sets out a new approach to studying the diagnostic decision-making process in PC, taking advantage of new technologies which allow immediate recording of the decision-making process.The authors gratefully acknowledge funding of this research from the Spanish Research Agency. Layer-by-layer vaginal films for acyclovir controlled release to prevent genital herpes

Genital herpes is one of the most common sexually transmitted infections worldwide. It mainly affects women, as the rate of sexual transmission from male-to-female is higher than from female-to-male. The application of vaginal antivirals drugs could reduce the prevalence of genital herpes and prevent future infections. Layer-bylayer vaginal films were prepared by the solvent evaporation method using iota-carrageenan, hydroxypropyl methylcellulose and the polymethacrylates Eudragit® RS PO and Eudragit® S100, for the controlled release of acyclovir. The films were characterized by texture analysis and Raman spectroscopy. Swelling, mucoadhesion, and drug release studies were conducted in simulated vaginal fluid. The results show that Layer-by-Layer films exhibited adequate mechanical properties. The structuring of the layer-by-layer films allowed the controlled release of acyclovir and produced a prolonged mucoadhesion residence time of up to 192 h. The films formed in layer 2 by the combination of Eudragit® RS PO and S100 showed a controlled release of acyclovir for eight days, and adequate mechanical properties. These promising formulations for the prevention of genital herpes deserve further evaluation

El Instituto Unidad de Investigaciones Jurídico-Sociales Gerardo Molina (Unijus) promueve y apoya el desarrollo e implementación de programas y proyectos de investigación que aporten al conocimiento de la realidad nacional e internacional, de las instituciones jurídicas, políticas y sociales, así como de las teorías jurídicas y políticas que contribuyan a la construcción de nuevas propuestas para la solución de las problemáticas que aquejan la sociedad. Por tal motivo, es fundamental el apoyo constante a la formación de semilleros de investigación en la Facultad de Derecho, Ciencias Políticas y Sociales de la Sede Bogotá, así como la consolidación de los semilleros ya existentes. En el año 2013, se desarrollaron las Convocatorias No. 01 y 02 de 2013 para la Selección de Semilleros de Investigación de la Facultad de Derecho, Ciencias Políticas y Sociales. Como resultado de dicho proceso fueron seleccionadas trece propuestas de investigación integradas por un total de 47 estudiantes, quienes a su vez se encontraban vinculados a 7 grupos de investigación. Hepatic levels of S-adenosylmethionine regulate the adaptive response to fasting

26 p.-6 fig.-1 tab.-1 graph. abst.There has been an intense focus to uncover the molecular mechanisms by which fasting triggers the adaptive cellular responses in the major organs of the body. Here, we show that in mice, hepatic S-adenosylmethionine (SAMe)—the principal methyl donor—acts as a metabolic sensor of nutrition to fine-tune the catabolic-fasting response by modulating phosphatidylethanolamine N-methyltransferase (PEMT) activity, endoplasmic reticulum-mitochondria contacts, β-oxidation, and ATP production in the liver, together with FGF21-mediated lipolysis and thermogenesis in adipose tissues. Notably, we show that glucagon induces the expression of the hepatic SAMe-synthesizing enzyme methionine adenosyltransferase α1 (MAT1A), which translocates to mitochondria-associated membranes. This leads to the production of this metabolite at these sites, which acts as a brake to prevent excessive β-oxidation and mitochondrial ATP synthesis and thereby endoplasmic reticulum stress and liver injury. Buffer layer morphology effects on the ordering of epitaxial FePd(001) thin films

The structural properties of thin FePd(001) films grown on MgO(100) substrates by UHV triode-sputtering have been studied as a function of the morphology of the substrate/seed layer interface. The films formed in layer 2 by the combination of Eudragit® RS PO and S100 showed a controlled release of acyclovir for eight days, and adequate mechanical properties.
