    JAK2 V617F-Dependent Upregulation of PU.1 Expression in the Peripheral Blood of Myeloproliferative Neoplasm Patients

    Myeloproliferative neoplasms (MPN) are multiple disease entities characterized by clonal expansion of one or more of the myeloid lineages (i.e. granulocytic, erythroid, megakaryocytic and mast cell). JAK2 mutations, such as the common V617F substitution and the less common exon 12 mutations, are frequently detected in such tumor cells and have been incorporated into the diagnostic criteria published by the World Health Organization since 2008. However, the mechanism by which these mutations contribute to MPN development is poorly understood. We examined gene expression profiles of MPN patients focusing on genes in the JAK–STAT signaling pathway using low-density real-time PCR arrays. We identified the following 2 upregulated genes in MPN patients: a known target of the JAK–STAT axis, SOCS3, and a potentially novel target, SPI1, encoding PU.1. Induction of PU.1 expression by JAK2 V617F in JAK2-wildtype K562 cells and its downregulation by JAK2 siRNA transfection in JAK2 V617F-positive HEL cells supported this possibility. We also found that the ABL1 kinase inhibitor imatinib was very effective in suppressing PU.1 expression in BCR-ABL1-positive K562 cells but not in HEL cells. This suggests that PU.1 expression is regulated by both JAK2 and ABL1. The contribution of the two kinases in driving PU.1 expression was dominant for JAK2 and ABL1 in HEL and K562 cells, respectively. Therefore, PU.1 may be a common transcription factor upregulated in MPN. PU.1 is a transcription factor required for myeloid differentiation and is implicated in erythroid leukemia. Therefore, expression of PU.1 downstream of activated JAK2 may explain why JAK2 mutations are frequently observed in MPN patients

    The favorite foods of obese women were snack and cereal before the dietary counseling class started. The obese women took care of having a low intake of oily foods and making a taste with low salt concentration. We often observed that they took shortage of foods as well as excess of ones. They repeated trials of the diet and had much amount of healthful food ingestions for concerning of their healthy life. After the class, the characteristics of the weight reduction group were decrease of taking snack foods and increase of the QOL, based on the balance sheet of food intake. These improvements were depended on the cooperation with their families. On the other hand, the non-weight reduction group was not improved because of the non-cooperation with their families

    The present thesis reports of the activities of a Team project“ Assessment Project for Criteria for Human Life Quality”, established by several members of the Department of Life Environmental Sciences. Preliminary research was mainly conducted in areas of consumer products use, housing facilities, as well as the cultivation of public opinion and attitudes towards low energy life. The high accumulation of systems and functions, the rational and rigid differentiations of tasks and areas, typical of city life, aggravate the alienation of the individual and accelerate the consumption rate of material and energy resource. Further research is necessary, therefore, to relocate the areas in which the needs for privacy and property can be reformulated to embrace a more organic and cyclic concept of life

    Sensitive measurement of positron emitters eluted from HPLC

    For sensitive analysis of the radioactive-metabolite in human PET, a radio-HPLC system coupled to a newly designed positron detector was constructed. The detector had the advantages of low noise level(1.7+/-1.0cpm)and high sensitivity(32+/-1%)due to coincidence counting and large BGO crystals. Furthermore, the detector was easy to move,since a pair of the BGO housings coupled to photomultipliers was effectively arranged in parallel and a HPLC cell with different volume could be inserted between the BGO housing. This radio-HPLC system was useful for analyzing samples with low radioactivity. It was applied to the measurement of [11C]FLB457in plasma,having high affinity and high selectivity with dopamine D2 receptors. Extremely low radioactivity of [11C]FLB457(2500dpm)could be analzed by using the radio-HPLC system. The performance of this detector was compared with those of commercially available systems that had been used as sensitive detectors for HPLC. (C)2001 Elsevier Science Ltd. All rights reserved
