    The relationship between histochemical enzyme activities of brain tumors and clinical features of the patients

    Human brain turnors removed from 126 patients were histochemically examined and following results were obtained. 1. In general, alkaline phosphatase activity is decreased in poorly differentiated gliomas, but is not related to the tumor cell infiltration. 2. All the cases of alkaline phosphatase negative gliomas have poor reconvalescent course and most of the positive cases show good reconvalescence. 3. Alkaline phosphatase, leucine aminopeptidase and acid phosphatase activities are remarkable in fibroblastic meningioma, moderate or feeble in meningocytic meningioma, and negative in malignant meningioma. 4. The activities of alkaline phosphatase, &#946;-esterase, leucine aminopeptidase and acid phosphatase are decreased in most of meningocytic meningiomas when the duration of symptoms and signs is short. 5. Succinic dehydrogenase, malic dehydrogenase, isocitric dehydrogenase and &#946;-glucuronidase are strongly reactive in malignant meningioma; from strong to moderate in meningocytic meningioma and from moderate to feeble in fibroblastic meningioma. 6. There is a slight increasing tendency of the activities of succinic dehydrogenase, malic dehydrogenase, isocitric dehydrogenase in fibroblastic meningioma and p·glucuronidase for a short duration of symtoms and signs. 7. In the case of acoustic neurinomas the higher the alkaline phosphatase activity, the longer is the duration of symptoms and signs.</p

    Histochemical studies on enzyme activities of gastric carcinoma. II. Dehydrogenases

    With gastric carcinomas the activities of eight dehydrogenases; succmlC, lactic, malic, &#945; glycerophosphate, glutamic, &#946;-hydroxybutyric, glucose-6-phosphate and isocitric dehydrogenase were statistically estimated. Principal findings may be briefly summarized as follows. These enzymatic activities differed considerably even in the same classification of carcinomas and generally ranged from strong to weak in the following order: lactic, malic, glucose-6-phosphate, isocitric, succinic, &#945;-glycerophosphate, glutamic and &#946;-hydroxybutyric dehydrogenase. The activities of adenocarcinomas were stronger than those in simple ones, and these were not related appreciably to cell differentiation in adenocarcinomas except succinic, glutamic, glucose-6-phosphate and isocitric dehydrogenase. As for succinic dehydrogenase and NAD-linked dehydrogenases except for lactic dehydrogenase, the activities were strongest in intestinal metaplasia and early mucosal carcinomas, the next being in benign adenomatous polyps and weakest in the other carcinomas. As for NADP-linked dehydrogenases and lactic dehydronase, the activities were also strongest in intestinal metaplasia and early carcinomas, the second in the other carcinomas and the third in the benign polyps. Generally, these dehydrogenase activities were strongest in free carcinoma cells in blood and lymph vessels and in actively growing part of several carcinomas and weakest in the central area of tumors, especially almost negative in the central necrotic area.</p

    Histochemical studies on enzyme activities of gastric carcinoma. I. Hydrolytic enzymes

    The activities of five hydrolytic enzymes, alkaline and acid phosphataSe, beta-esterase, leucine aminopeptidase and beta-glucuronidase, of human gastric carcinomas from 180 patients were investigated histochemically. Alkaline phosphatase activity was almost negative in the carcinoma but was weakly positive in this tumor at times (about 10 to 20 per cent). Acid phosphatase activity which displayed a slightly increasing tendency of the reaction in poorly differentiated tumor was variegated and mainly from feeble to moderate in activity. Beta-esterase reaction was in varying degrees with each case, but more malignant the carcinomas, the weaker was the activity. Leucine aminopeptidase was positive in about 30 to 60 per cent of the specimens observed but the reaction was founded to be localized often in some areas and generally similar to alkaline phosphatase reaction. The activities of leucine aminopeptidase, alkaline phosphatase and beta-esterase were positive at a higher rate in mucinous carcinomas than in non-mucin producing one. Beta-glucuronidase activity was slight or moderate in general but rather strong in the early stage of carcinomas.</p

    A histochemical study on hydrolytic and oxidative enzymes in human sarcomas

    Histochemical evaluations of human sarcomas such as reticulum cell sarcoma, fibrosarcoma, lymphosarcoma and neurofibrosarcoma, were carried out with five hydrolytic enzymes and eight oxidative enzymes. The activities of acid phosphatase and beta-glucuronidase were slightly positive in the neoplastic cells observed. Beta-esterase activity was also positive but varied according to the kind of sarcomas. Alkaline phosphatase activity was faint or negative in sarcoma cells, though positive in capillary walls. Leucine aminopeptidase activity was negative giving not any appreciable coloration of the cell as far as the method employed is concerned. Among the activities of dehydrogenases, the most intense activity was observed in lactic dehydrogenase. The activities of succinic and beta-hydroxybutyric dehydrogenases were slight. The activities of alpha-glycerophosphate, glutamic and betahydroxybutyric dehydrogenases were faint or slight. The activities of NADPlinked dehydrogenases, glucose-6-phosphate and isocitric dehydrogenase were all faint or slight in these sarcoma cells.</p

    Baseline tumour necrosis factor alpha levels predict the necessity for dose escalation of infliximab therapy in patients with rheumatoid arthritis

    Objectives: To investigate the possible role of baseline plasma tumour necrosis factor alpha levels (baseline-TNF) on the clinical response to infliximab in patients with rheumatoid arthritis (RA). Methods: Patients with RA refractory to methotrexate received 3, 6, or 10 mg/kg of infliximab every 8 weeks, in a randomised, double-blind manner: the RISING study. Clinical response (disease activity score in 28 joints based on C-reactive protein or American College of Rheumatology core set) at week 54 and serum infliximab levels were compared in three patient groups with low, intermediate, or high baseline-TNF (TNF-low, TNF-int, or TNF-high). Results: In TNF-low patients, the clinical response to different doses of infliximab was comparable, whereas TNF-int patients exhibited a dose-dependent trend. In contrast, TNF-high patients (approximately 13% of the total patients) had a clinical response to 10 mg/kg significantly better than the response to 3 and 6 mg/kg of infliximab. In TNF-high patients, the median trough serum levels of infliximab were below the detection limit (<0.1 μg/ml) at 3 and 6 mg/kg but were greater than 2 μg/ml at 10 mg/kg, whereas the levels were approximately 1 μg/ml for each dosage group in TNF-low patients. Conclusion: In patients with RA, baseline-TNF is significantly associated with the clinical response to infliximab in patients with a high baseline-TNF. A higher dose of infliximab may be necessary in these patients, whereas lower doses of infliximab are sufficient for those with a low baseline-TNF. Baseline-TNF may be a useful measure for personalising the treatment of RA using infliximab

    Development of an intravital imaging system for the synovial tissue reveals the dynamics of CTLA-4 Ig in vivo

    There have been many attempts to visualize the inflamed joints using multiphoton microscopy. However, due to the hypervascular and multilayered structure of the inflamed synovium, intravital imaging of the deep synovial tissue has been difficult. Here, we established original intravital imaging systems to visualize synovial tissue and pathological osteoclasts at the pannus–bone interface using multiphoton microscopy. Combined with fluorescence-labeling of CTLA-4 Ig, a biological agent used for the treatment of rheumatoid arthritis, we identified that CTLA-4 Ig was distributed predominantly within the inflamed synovium and bound to CX3CR1+ macrophages and CD140a+ fibroblasts 6 h after injection, but not to mature osteoclasts. Intravital imaging of blood and lymphatic vessels in the inflamed synovium further showed that extravasated CTLA-4 Ig was immediately drained through lymphatic vessels under acute arthritic conditions, but the drainage activity was retarded under chronic conditions. These results indicate that this intravital synovial imaging system can serve as a platform for exploring the dynamics of immune cells, osteoclasts, and biological agents within the synovial microenvironment in vivo.Hasegawa T., Kikuta J., Sudo T., et al.     Clinical features of asthma patients with bronchiolar obstruction (type Ⅱ asthma) were studied in relation to the proportion of neutrophils in bronchoalveolar lavage (BAL) fluid. Of 13 subjects studied, 7 were accompanied with BAL neutrophilia (53.5%) (BALn(+)) and 6 were without BAL neutrophilia (3.5%) (BALn(-)). 1. The mean age was higher in BALn(-) (66.0 years) than in BALn(+) patients (55.0 years). 2. Bronchial reactivity to methacholine was slightly higher in BALn(-) patients than in those with BALn(+). 3. The value of FEV1.0% was significantly lower in BALn(+) patints than in those with BALn(-) (p<0.01). 4. The proportion of BAL lymphocytes was signicantly more decreased in BALn(+) patients compared to the proportion in those with BALn(-) (p<0.001). 5. the values of serum IgG, IgA, and IgM were not significantly different between BALn(+) and BALn(-) patients, however, the value of IgG was more decreased in BALn(+) patients than in those with BALn(-). These results suggest that two kinds of type Ⅱ asthma ; one is with BAL neutrophilia related to suppressed immunity, and another is without BAL neutrophilia in part due to aging.細気管支閉塞型(Ⅱ型)喘息の臨床的特徴が,BAL液中の好中球頻度との関連のもとに検討された。対象13例のうち,7例がBAL液中好中球増加(平均好中球頻度;53.5%)をともなう症例(BALn(+))で,残りの6例はBAL液中好中球増加 をともなわない(3.5%)症例(BALn(-))であった。1.平均年齢は,BALn(+)症例(55.0才)に比べ,BALn(-)症例(66.0才)でより高い傾向が見られた。2.メサコリンに対する気道過敏性は,BALn(+)症例に比べBALn(-)症例でやや高い傾向が見られたが,両者間に有意の差は見られなかった。3.FEV1.0%値は,BALn(-)症例に比べBALn(+)症例で有意に低い値を示した(P<0.05)。4.BAL液中リンパ球頻度はBALn(+)症例でBALn(-)症例に比べ有意に低い値を示した(P<0.001)。5.血清IgG,IgAおよびIgM値には両者間に有意の差は見られなかったが,IgG値はBALn(+)症例でより低い傾向が見られた。これらの結果より,Ⅱ型喘息にはBAL液中好中球増加を示す症例と示さない症例の2種類があること,そして前者は免疫能の低下と,そして後者は加齢とある程度の関連があることが示唆された

The patient, a 5-year-old girl, had repeated episodes of productive cough after contracting the common cold at the age of 1 year and 6 months.     Residual Strain Dependence on Matrix Structure in RHQ-Nb3Al Wires by Neutron Diffraction Measurement

    We prepared three types of non-Cu RHQ-Nb3Al wire samples with different matrix structures: an all-Ta matrix,a composite matrix of Nb and Ta with a Ta inter filament, and an all-Nb matrix. Neutron diffraction patterns of the wire samples were measured at room temperature in J-PARC "TAKUMI". To obtain residual strains of materials, we estimated lattice constant a by multi-peak analysis in the wire. Powder sample of each wire was measured, where the powder was considered to be strain-free. The grain size of all the powder samples was below 0.02 mm. For wire sample with the all-Nb matrix, we also obtained lattice spacing d by a single-peak analysis. Residual strains of Nb3Al filament were estimated from the two analysis results and were compared. Result, residual strains obtained from the multi-peak analysis showed a good accuracy with small standard deviation. The multi-peak analysis results for the residual strains of Nb3Al filament in the three samples were all tensile residual strain in the axial direction, they are 0.12%, 0.12%, and 0.05% for the all-Ta matrix, the composite matrix, and the all-Nb matrix, respectively. Difference in the residual strain of Nb3Al filament between the composite and all-Nb matrix samples indicates that type of inter-filament materials show a great effect on the residual strain.     Novel scheme for Lane-Bates\u27 blind deconvolution : Determinant conditions for the zeros of blurs and a simple algorithm for eliminating blurs

     The Lane-Bates method of blind deconvolution makes it possible to analytically recover the original image without prior knowledge of blurs convolved in a given image. The method utilizes the zeros of the z-transform of the given image. Its implementation, however, requires highly nontrivial analysis of the zeros. We have developed a novel scheme that considerably simplifies the analysis of the zeros. We have developed two versions of the scheme, i.e., determinant conditions (DCs) for the zeros of blurs and a search algorithm (SA) of blur images. The DCs consist of two forms, i.e., a derivative form and a multi-point form. The derivative form is given as a determinant form of conditions on derivatives of the zeros of assumed blurs that can be evaluated by using zeros of the z-transform of the given image. On the other hand, the multi-point form is given as a determinant form of conditions on the zeros of assumed blurs that are evaluated at multiple points in z space. The scheme is particularly powerful when the blurs have multiple structures as we illustrate. The SA is given as a form of simultaneous equations for blur elements of an assumed blur. The method is powerful when we try to find a single blur. This method is robust for compressed gray-scale images. These methods have been experimentally tested with model blurred images and shown to be powerful. In this report we illustrate how they are useful for the Lane-Bates blind deconvolution
