    Different carbohydrate sources affect swine performance and post-prandial glycaemic response

    The type of starch and fibre in the diet affects several parameters, including glycaemic and insulin response, that are involved in pig growth performance. Four experimental diets for growing pigs differing for carbohydrates source (corn, barley, faba bean and pea) were tested. The diets were analysed in vitro to assess the carbohydrates characteristics, and they were administered to 56 crossbreed growing pigs (Landrace × Large White) randomly divided into four groups (mean age of 95 ± 6 days; body weight 80 kg ± 4 days). Clinical examination and average daily gain were performed before recruitment and after 40 days of experiment. The metabolic effects were investigated by blood count and serum biochemical parameters and by the glycaemic and insulin post-prandial response. The study revealed substantial differences among the diets, suggesting that alternative feedstuffs for swine affect several parameters, including glycaemic and insulin response, with no negative effects on growing performance. The Barley group showed the highest daily weight gain (p <.05) associated with the highest glycaemic (p <.05) and insulin response at 1 and 2 h post-prandial (p <.01), suggesting that the barley-based diet can support performance comparable to that of the corn-based diet in growing pig. By contrast, the lowest glycaemia was observed in the Faba bean group (p <.05), confirming the capacity of this legume to modulate post-prandial glucose levels. Moreover, the ability of some ingredients in lowering glucose and insulin response enriches the knowledge on functional nutrients for animal diets and to prevent the incidence of enteric diseases.Highlights The type of starch and fibre used in the diet highly affected some blood parameters, such as glycaemic and insulin responses.     Abstract: Aim: The aim of this study was to assess if discrepancy between power-Doppler voiding urosonography (PD-VUS) and voiding cystourethrography (VCUG) affects the management of patients with primary vesicoureteric reflux (VUR). Materials and Methods: Fifty-six children with suspected or known VUR were assessed both by PD-VUS and VCUG. Two independent observers, both pediatric surgeons, each aware of the results of only one imaging modality, advised children's management according to present care standards. Agreement between diagnostic findings at the two imaging modalities and between therapeutic advice of the two observers was evaluated Using K statistics. Results: PD-VUS diagnosed VUR in 3 patients and 6 ureteral units more than VCUG. VCUG showed VUR in 2 ureteral units, but in no patient more than PD-VUS. Accuracy of PD-VUS compared with VCUG was 92.8% and 94.6% considering ureteral units and patients, respectively. The two observers disagreed about the management of 4 (7%) of 56 cases. Agreement was significant (P < .001) both between findings at the two imaging modalities and between management options advised by the two independent observers

    Effect of a high forage: Concentrate ratio on milk yield, blood parameters and oxidative status in lactating cows

    A feeding strategy that requires a forage: concentrate ratio equal to 70: 30, with at least five different herbs in the forage and the use of silages prohibited, has recently been introduced in Italy. Despite the benefits in terms of human health (lower ω6: ω3 ratio, higher conjugated linoleic acid level) of the obtained milk, little information regarding the possible effects on cows' health is available. The aim of this study was to evaluate the effects of such a feeding strategy in dairy cows (90 days in milk at the beginning of the trial) on milk yield and composition, and blood metabolic profile, including the evaluation of oxidative stress. The proposed feeding strategy, compared with a semi-intensive strategy, resulted in an improvement of animal oxidative status (lower levels of reactive oxygen metabolites, higher levels of antioxidant potential and anti-reactive oxygen metabolites) and a significant increase of milk urea only in the first part of the trial. No differences in milk yield and composition were detected throughout the trial

    In vitro fermentation of ten cultivars of barley silage.

    The fermentation characteristics of whole-crop barley silages from ten different cultivars were evaluated by the in vitro gas production technique. The organic matter degradability of barley silage (62.9% in average) was comparable to those reported in our previous trials for oat (59.7%) and sorghum silages (65.5%); while the maximum gas production rate (5.38 ml/h in average) was slightly lower respect to oat (6.71 ml/h) and sorghum silage (6.74 ml/h). The mean nutritive value (4.00 MJ/kg DM) calculated on the basis of both chemical composition and in vitro fermentation data was comparable to that (4.16 MJ/kg DM) obtained in our previous research performed on corn silage, from crop sowed in the same area

    Detection of plant species-specific dna (barley and soybean) in blood, muscle tissue, organs and gastrointestinal contents of rabbit

    The aim of this study was to detect plant DNA sequences from low copy number genes of barley grain and soybean meal, the latter being subjected to solvent extraction process, in blood, liver, kidney, spleen, muscle tissue and digesta (duodenum, caecum and faeces from rectal ampulla) of rabbits. For fattening, Hyla rabbits (20 males and 20 females) were fed a diet including barley grain (15%) and soybean meal (12%). Animals were slaughtered at 74 d of age (2 ± 0.2 kg live weight) and samples collected from each animal. The quality of each DNA sample was verified using the UNIV P/Q primers used to amplify a mammalian specifi c portion of mtDNA 16S rRNA gene. The presence of plant DNA was subsequently ascertained on the same DNA samples, as well as on barley and soybean (control). Two classes of plant DNA sequences were monitored via real-time PCR, using SYBR(R) Green I Dye: a high copy number chloroplast gene (trnl) and a low copy number specific for barley (metal-dependent hydrolase-like protein) and soybean (lectin) genes. Melting curve analysis was used to identify the PCR products. The chloroplast fragment detection frequency was higher (P<0.01) in muscle (90%), liver (80%), kidney (80%) and spleen (80%) than in blood (40%) and digesta samples. In the latter, chloroplast DNA was found in 40 and 30% of duodenum and caecum contents respectively, and in 30% of faeces. The specificity of the amplicons obtained was checked by sequencing and annotation. In the samples positive for chloroplast fragments, the frequency of detection of barley specific sequence was higher (P<0.01) in liver (62.5%), kidney (62.5%), spleen (62.5%) and digesta (100%) than in blood (25%) and muscle (22.2%) samples. The soybean lectin gene was not detected in animal samples, although it was seen in plant samples.     Effects of ecotrofin™ on milk yield, milk quality and serum biochemistry in lactating goats

    A nutritional supplement (Ecotrofin™, by Vetoquinol Italia S.r.l) recommended in ruminants feeding to strengthen the physiological condition and improve digestive performance was tested in 20 pluriparae grazing goats divided in two groups (control and treated) to assess its possible effects on milk yield and quality and to assess eventual adverse effects. Animals from both groups also received 400&nbsp;g/day of corn meal, and the treated group was supplemented with 20&nbsp;g/head/day of the nutritional supplement. At the doses suggested by the manufacturer, despite a transient increase after 30&nbsp;days of supplementation, Ecotrofin™ did not show significant effects on milk yield and, although some changes were found in the fatty acids profile, no significant improvement of MUFA and PUFA, as well as of omega-6:omega-3 ratio and CLA content were seen. Therefore, in our experimental conditions the supplementation of diet with Ecotrofin™ did not appear useful to improve goat's performance. A significant effect on kidney health markers (27 vs. 22.5 for urea and 0.83 vs. 0.76 for creatinine, p&nbsp;&lt;&nbsp;0.05) suggested a beneficial effect on renal function but, since levels fell in the normal ranges in both groups, such hypothesis would need further studies to be addressed

    In vitro and in vivo studies of Cucurbita pepo L. flowers: chemical profile and bioactivity

    Edible flowers consumption has increased in recent years due to their rich content of healthy phytochemicals. The aim of this study was to analyse the chemical profile of Cucurbita pepo L. flowers, and to explore their antioxidant and hypoglycaemic prop- erties. Moreover, in order to assess in vivo effects, biochemical analysis, Reactive Oxygen Metabolites (d-ROMs) and Biological Antioxidant Potential (BAP) tests were performed on mice serum. High Performance Liquid Chromatography-Diode Array Detection (HPLC-DAD) analyses revealed the presence of (þ)-catechin, ()-epicatechin, rutin, and syringic acid as main constituents. 2,20- Azino-bis (3-ethylbenzothiazoline-6-sulphonic acid) (ABTS) and Ferric Reducing Antioxidant Power (FRAP) tests showed interest- ing results. The extract exhibited the strongest inhibitory effect on a-glucosidase (IC50 of 144.77lg/mL). In vivo results confirmed the hypoglycaemic effects, also affecting lipid metabolism but did not revealed benefits on ROS production. These results may add some information supporting the use of C. pepo flowers as func- tional foods and/or nutraceuticals

    Influence of dietary hydrogenated palm oil supplementation on serum biochemistry and progesterone levels in dairy goats

    The aim of this research was to investigate the influence of hydrogenated palm oil (HPO) added to a dairy goat diet on serum biochemistry and progesterone levels. Thirty pregnant Cilentana dairy goats were equally divided into 2 groups (control [CTR] and HPO groups). After kidding, concentrated feed for both groups was gradually increased up to 400 g/(animald),andtheHPOgroupreceived50g/(animald), and the HPO group received 50 g/(animald) of HPO. Supplementation with HPO significantly increased cholesterol levels (mg/dL, 63.80 vs. 54.68 at 30 d, P 0.05; 78.20 vs. 58.00 at 60 d, P 0.05; 83.80 vs. 57.83 at 120 d, P 0.01) compared with the CTR group although no significant differences were detected for liver and kidney function indicators. Moreover, other biochemical parameters were not affected by HPO supplementation thus suggesting no change occurred in lipid and protein metabolism. Furthermore, a significant correlation was found be- tween progesterone levels and serum cholesterol (r 1⁄4 0.65, P 0.01) although these were not signifi- cantly higher in HPO supplemented goats. The dose and time of HPO supplementation appears critical as regards assessing the limits between the risks and benefits of HPO supplementation in dairy goats. At the tested dose, HPO was well tolerated by the animals and may represent a useful tool to increase energy availability during highly demanding periods
