    Radar‐Sounding Characterization of the Subglacial Groundwater Table Beneath Hiawatha Glacier, Greenland

    Radar-sounding surveys associated with the discovery of a large impact crater beneath Hiawatha Glacier, Greenland, revealed bright, flat subglacial reflections hypothesized to originate from a subglacial groundwater table. We test this hypothesis using radiometric and hydrologic analysis of those radar data. The dielectric loss between the reflection from the top of the basal layer and subglacial reflection and their reflectivity difference represent dual constraints upon the complex permittivity of the basal material. Either ice-cemented debris or fractured, well-drained bedrock explain the basal layer's radiometric properties. The subglacial reflector's geometry is parallel to isopotential hydraulic head contours, located 7.5–15.3 m below the interface, and 11 ± 7 dB brighter than the ice–basal layer reflection. We conclude that this subglacial reflection is a groundwater table and that its detection was enabled by the wide bandwidth of the radar system and unusual geologic setting, suggesting a path for future direct radar detection of subglacial groundwater elsewhere

    Incorporating a Hyperspectral Direct-Diffuse Pyranometer in an Above-Water Reflectance Algorithm

    In situ hyperspectral remote-sensing reflectance (Rrs(λ)) is used to derive water quality products and perform autonomous monitoring of aquatic ecosystems. Conventionally, above-water Rrs(λ) is estimated from three spectroradiometers which measure downwelling planar irradiance(Ed(λ)), sky radiance (Ls(λ)), and total upwelling radiance (Lt(λ)), with a scaling of Ls(λ)/Ed(λ)used to correct for surface-reflected radiance. Here, we incorporate direct and diffuse irradiance,(Edd(λ)) and Eds(λ)), from a hyperspectral pyranometer (HSP) in an Rrs(λ) processing algorithm from a solar-tracking radiometry platform (So-Rad). HSP measurements of sun and sky glint (scaled Edd(λ)/Ed(λ) and Eds(λ)/Ed(λ)) replace model-optimized terms in the 3C (three-glint component) Rrs(λ) algorithm, which estimates Rrs(λ) via spectral optimization of modelled atmospheric and water properties with respect to measured radiometric quantities. We refer to the HSP-enabled method as DD (direct-diffuse) and compare differences in Rrs(λ) and Rrs(λ) variability (assessed over 20 min measurement cycles) between 3C and DD as a function of atmospheric optical state using data from three ports in the Western Channel. The greatest divergence between the algorithms occurs in the blue part of the spectrum where DD has significantly lower Rrs(λ) variability than 3C in clearer sky conditions. We also consider Rrs(λ) processing from a hypothetical two-sensor configuration (using only the Lt(λ) spectroradiometer and the HSP and referred to as DD2) as a potential lower-cost measurement solution, which is shown to have comparable Rrs(λ) and Rrs(λ) variability to DD in clearer sky conditions. Our results support that the HSP sensor can fulfil a dual role in aquatic ecosystem monitoring by improving precision in Rrs(λ) alongside its primary function to characterize aerosols

    Ipl1/aurora kinase suppresses S-CDK-driven spindle formation during prophase I to ensure chromosome integrity during meiosis

    Cells coordinate spindle formation with DNA repair and morphological modifications to chromosomes prior to their segregation to prevent cell division with damaged chromosomes. Here we uncover a novel and unexpected role for Aurora kinase in preventing the formation of spindles by Clb5-CDK (S-CDK) during meiotic prophase I and when the DDR is active in budding yeast. This is critical since S-CDK is essential for replication during premeiotic S-phase as well as double-strand break induction that facilitates meiotic recombination and, ultimately, chromosome segregation. Furthermore, we find that depletion of Cdc5 polo kinase activity delays spindle formation in DDR-arrested cells and that ectopic expression of Cdc5 in prophase I enhances spindle formation, when Ipl1 is depleted. Our findings establish a new paradigm for Aurora kinase function in both negative and positive regulation of spindle dynamics

    Spatial structure of in situ reflectance in coastal and inland waters: implications for satellite validation

    Validation of satellite-derived aquatic reflectance involves relating meter-scale in situ observations to satellite pixels with typical spatial resolution ∌ 10–100 m within a temporal “match-up window” of an overpass. Due to sub-pixel variation these discrepancies in measurement scale are a source of uncertainty in the validation result. Additionally, validation protocols and statistics do not normally account for spatial autocorrelation when pairing in situ data from moving platforms with satellite pixels. Here, using high-frequency autonomous mobile radiometers deployed on ships, we characterize the spatial structure of in situ Rrs in inland and coastal waters (Lake Balaton, Western English Channel, Tagus Estuary). Using variogram analysis, we partition Rrs variability into spatial and intrinsic (non-spatial) components. We then demonstrate the capacity of mobile radiometers to spatially sample in situ Rrs within a temporal window broadly representative of satellite validation and provide spatial statistics to aid satellite validation practice. At a length scale typical of a medium resolution sensor (300 m) between 5% and 35% (median values across spectral bands and deployments) of the variation in in situ Rrs was due to spatial separation. This result illustrates the extent to which mobile radiometers can reduce validation uncertainty due to spatial discrepancy via sub-pixel sampling. The length scale at which in situ Rrs became spatially decorrelated ranged from ∌ 100–1,000 m. This information serves as a guideline for selection of spatially independent in situ Rrs when matching with a satellite image, emphasizing the need for either downsampling or using modified statistics when selecting data to validate high resolution sensors (sub 100 m pixel size)

    Recommendations for exercise adherence measures in musculoskeletal settings : a systematic review and consensus meeting (protocol)

    Background: Exercise programmes are frequently advocated for the management of musculoskeletal disorders; however, adherence is an important pre-requisite for their success. The assessment of exercise adherence requires the use of relevant and appropriate measures, but guidance for appropriate assessment does not exist. This research will identify and evaluate the quality and acceptability of all measures used to assess exercise adherence within a musculoskeletal setting, seeking to reach consensus for the most relevant and appropriate measures for application in research and/or clinical practice settings. Methods/design: There are two key stages to the proposed research. First, a systematic review of the quality and acceptability of measures used to assess exercise adherence in musculoskeletal disorders; second, a consensus meeting. The systematic review will be conducted in two phases and reported in accordance with the Preferred Reporting Items for Systematic Reviews and Meta-Analyses (PRISMA) guidelines to ensure a robust methodology. Phase one will identify all measures that have been used to assess exercise adherence in a musculoskeletal setting. Phase two will seek to identify published and unpublished evidence of the measurement and practical properties of identified measures. Study quality will be assessed against the COnsensus-based Standards for the selection of health Measurement Instruments (COSMIN) guidelines. A shortlist of best quality measures will be produced for consideration during stage two: a meeting of relevant stakeholders in the United Kingdom during which consensus on the most relevant and appropriate measures of exercise adherence for application in research and/or clinical practice settings will be sought. Discussion: This study will benefit clinicians who seek to evaluate patients’ levels of exercise adherence and those intending to undertake research, service evaluation, or audit relating to exercise adherence in the musculoskeletal field. The findings will impact upon new research studies which aim to understand the factors that predict adherence with exercise and which test different adherence-enhancing interventions.     Shedding light on the elusive role of endothelial cells in cytomegalovirus dissemination.

    Cytomegalovirus (CMV) is frequently transmitted by solid organ transplantation and is associated with graft failure. By forming the boundary between circulation and organ parenchyma, endothelial cells (EC) are suited for bidirectional virus spread from and to the transplant. We applied Cre/loxP-mediated green-fluorescence-tagging of EC-derived murine CMV (MCMV) to quantify the role of infected EC in transplantation-associated CMV dissemination in the mouse model. Both EC- and non-EC-derived virus originating from infected Tie2-cre(+) heart and kidney transplants were readily transmitted to MCMV-naĂŻve recipients by primary viremia. In contrast, when a Tie2-cre(+) transplant was infected by primary viremia in an infected recipient, the recombined EC-derived virus poorly spread to recipient tissues. Similarly, in reverse direction, EC-derived virus from infected Tie2-cre(+) recipient tissues poorly spread to the transplant. These data contradict any privileged role of EC in CMV dissemination and challenge an indiscriminate applicability of the primary and secondary viremia concept of virus dissemination

    Temporal changes in frequency of severe hypoglycemia treated by emergency medical services in types 1 and 2 diabetes:a population-based data-linkage cohort study

    Background  Almost 20 years ago, the frequencies of severe hypoglycemia requiring emergency medical treatment were reported in people with types 1 and 2 diabetes in the Tayside region of Scotland. With subsequent improvements in the treatment of diabetes, concurrent with changes in the provision of emergency medical care, a decline in the frequency of severe hypoglycemia could be anticipated. The present population-based data-linkage cohort study aimed to ascertain whether a temporal change has occurred in the incidence rates of hypoglycemia requiring emergency medical services in people with types 1 and 2 diabetes.  Methods  The study population comprised all people with diabetes in Tayside, Scotland over the period 1 January 2011 to 31 December 2012. Patients’ data from different healthcare sources were linked anonymously to measure the incidence rates of hypoglycemia requiring emergency medical services that include treatment by ambulance staff and in hospital emergency departments, and necessitated hospital admission. These were compared with data recorded in 1997–1998 in the same region.  Results  In January 2011 to December 2012, 2029 people in Tayside had type 1 diabetes and 21,734 had type 2 diabetes, compared to 977 and 7678, respectively, in June 1997 to May 1998. In people with type 2 diabetes, the proportion treated with sulfonylureas had declined from 36.8 to 22.4% (p<0.001), while insulin-treatment had increased from 11.7 to 18.7% (p<0.001). The incidence rate of hypoglycemia requiring emergency medical treatment had significantly fallen from 0.115 (95% CI: 0.094–0.136) to 0.082 (0.073–0.092) events per person per year in type 1 diabetes (p<0.001), and from 0.118 (0.095–0.141) to 0.037 (0.003–0.041) in insulin-treated type 2 diabetes (p=0.008). However, the absolute annual number of hypoglycemia events requiring emergency treatment was 1.4-fold higher.  Conclusions  Although from 1998 to 2012 the incidences of hypoglycemia requiring emergency medical services appeared to have declined by a third in type 1 diabetes and by two thirds in insulin-treated type 2 diabetes, because the prevalence of diabetes was higher (2.7 fold), the number of severe hypoglycemia events requiring emergency medical treatment was greater
