    Effects of vitrification and post-thawing interval on the cytoskeleton and subsequent fertilization rate of in vitro derived bovine oocytes

    Vitrification may alter the cytoskeleton (microtubule, meiotic spindle, microfilament, etc.) and the subsequent fertilization rate of in vitro derived bovine oocytes. This study was conducted to evaluate the effect of vitrification and post-thawing incubation periods on the cytoskeleton and fertilization rate of in vitro matured (IVM) bovine oocytes. Following 22 h of IVM, 184 fresh matured oocytes (MO) were immediately fertilized in vitro and served as a control. The remaining MO (1009) were then vitrified by the solid surface vitrification method. Immediately after thawing, MO were incubated in maturation medium in 5% CO2 at 39 °C for 0, 30, 60, 90 and 120 min respectively. Following incubation, half of the MO from each vitrified-thawed treatment group (0, 30, 60, 90, and 120 min) was stained with fluorescein isothiocyanate conjugated (FITC) and propidium iodide (PI) to evaluate the microtubule and DNA or spindle under laser-scanning confocal microscopy. The remaining half from the vitrified-thawed MO treatment groups was washed three times in Brackett and Oliphant\'s fertilization medium and in vitro fertilized. Cleavage and blastocyst rates were recorded 48 h post-fertilization. Results demonstrated that vitrification damaged MO zona pellucida (ZP), microtubule (MT), meiotic spindle (MS), and caused chromosomal fragmentation. Both the cleavage (84%) and blastocyst rates (50%) of the control group were significantly higher compared to the vitrified-thawed treatment groups. However, extending the incubation period of vitrified MO to 120 min after thawing (prior to fertilization) improved cleavage (65%) and blastocyst (13%) rates 48 h post-fertilization. Fertilizing vitrified MO immediately (0 min group) after thawing resulted in the lowest cleavage (42%) and blastocyst (1.9%) rates. In conclusion, vitrification reduces the subsequent fertilization rate of MO, however, a prolonged post-thawing incubation period (120 min) improves survival, cleavage and blastocyst formation rates, and enhances the reorganization of MO\'s cytoskeleton (MT and MS).     Pairwise comparison matrices and the error-free property of the decision maker

    Pairwise comparison is a popular assessment method either for deriving criteria-weights or for evaluating alternatives according to a given criterion. In real-world applications consistency of the comparisons rarely happens: intransitivity can occur. The aim of the paper is to discuss the relationship between the consistency of the decision maker—described with the error-free property—and the consistency of the pairwise comparison matrix (PCM). The concept of error-free matrix is used to demonstrate that consistency of the PCM is not a sufficient condition of the error-free property of the decision maker. Informed and uninformed decision makers are defined. In the first stage of an assessment method a consistent or near-consistent matrix should be achieved: detecting, measuring and improving consistency are part of any procedure with both types of decision makers. In the second stage additional information are needed to reveal the decision maker’s real preferences. Interactive questioning procedures are recommended to reach that goal

    The effect of calcium on auxin depletion-induced tomato (Lycopersicon esculentum Mill.) pedicel abscission

    Indole-3-acetic acid (IAA) and calcium are the most important factors that instigate plant organ abscission. This study aimed to elucidate the mechanisms that underlie the effects of IAA and calcium on delayed abscission in tomato. The results showed a clear trend towards reduced abscission rates with increased concentrations of IAA, and the applications on pedicel proximal or distal side also resulted in a different abscission. IAA combined with calcium significantly improved inhibition in contrast to IAA only, while IAA combined with magnesium exhibited little increased inhibition. 1-NNaphthylphthalamic acid (NPA), a polar auxin transport inhibitor, accelerated the abscission. IAA transported basipetally through an assay with 4 mm long pedicel sections indicated that the average transport intensity of [3H]-IAA applied to the distal pedicel end was 65 Bq h–1 and the average velocity was 5.29 mm h-1. When the proximal side was incubated in [3H]-IAA, its average transport intensity reduced to 19.53 Bq h–1 and the average velocity was only 1.92 mm h-1. Calcium treatment enhanced IAA transportation, as shown by significantly enhancing the transport intensity, but it had no effect on velocity.Keywords: Indole-3-acetic acid (IAA), calcium, abscission, tomat

    On reducing inconsistency of pairwise comparison matrices below an acceptance threshold

    A recent work of the authors on the analysis of pairwise comparison matrices that can be made consistent by the modification of a few elements is continued and extended. Inconsistency indices are defined for indicating the overall quality of a pairwise comparison matrix. It is expected that serious contradictions in the matrix imply high inconsistency and vice versa. However, in the 35-year history of the applications of pairwise comparison matrices, only one of the indices, namely CR proposed by Saaty, has been associated to a general level of acceptance, by the well known ten percent rule. In the paper, we consider a wide class of inconsistency indices, including CR, CM proposed by Koczkodaj and Duszak and CI by Pel\'aez and Lamata. Assume that a threshold of acceptable inconsistency is given (for CR it can be 0.1). The aim is to find the minimal number of matrix elements, the appropriate modification of which makes the matrix acceptable. On the other hand, given the maximal number of modifiable matrix elements, the aim is to find the minimal level of inconsistency that can be achieved. In both cases the solution is derived from a nonlinear mixed-integer optimization problem. Results are applicable in decision support systems that allow real time interaction with the decision maker in order to review pairwise comparison matrices.Comment: 20 page
