    Antimicrobial peptide expression in a wild tobacco plant reveals the limits of host-microbe-manipulations in the field

    Plant-microbe associations are thought to be beneficial for plant growth and resistance against biotic or abiotic stresses, but for natural ecosystems, the ecological analysis of microbiome function remains in its infancy. We used transformed wild tobacco plants (Nicotiana attenuata) which constitutively express an antimicrobial peptide (Mc-AMP1) of the common ice plant, to establish an ecological tool for plant-microbe studies in the field. Transgenic plants showed in planta activity against plant-beneficial bacteria and were phenotyped within the plants´ natural habitat regarding growth, fitness and the resistance against herbivores. Multiple field experiments, conducted over 3 years, indicated no differences compared to isogenic controls. Pyrosequencing analysis of the root-associated microbial communities showed no major alterations but marginal effects at the genus level. Experimental infiltrations revealed a high heterogeneity in peptide tolerance among native isolates and suggests that the diversity of natural microbial communities can be a major obstacle for microbiome manipulations in nature

    Investigation on the Mechanical and Tribological Properties of Aluminium-Tin Based Plain Bearing Material

    The purpose of this study is to investigate the Mechanical and Tribological properties of plain bearing alloys used especially in internal combustion engines. The mechanical properties namely Tensile strength and Hardness were investigated according to standard procedure. The sliding friction and wear properties of aluminium-tin alloy against high carbon high chromium steel were investigated at different normal loads as (29.43 N, 33.35 N and 36.25 N). Tests were carried in oil lubricated conditions with a sliding speed of 1 m/s. Prior to experimentation, the circulating engine oil 20w40 was heated to temperature of 800C using heater. The frictional behavior and wear property of aluminium-tin alloy were studied by means of pin-on-disk tribometer. The weight loss of the specimen was measured and wear and friction characteristics were calculated with respect to time, depth of wear track, sliding speed and bearing load. To determine the wear mechanism, the worn surfaces of the samples were examined using Scanning Electron Microscope (SEM). The optimum wear reduction was obtained at different normal loads and at same sliding speed

    Exploring the genetic variability, heritability, gene action for yield related traits and ToLCNDV resistance on F3 and F4 generations in cucumber [Cucumis sativus L.]

    Cucumber is traditionally cultivated in India and is recognized as a primary centre of origin, belonging to the gourd family, Cucurbitaceae. The study focused on assessing genetic variability in F3 and F4 cucumber populations and examining correlations among various characters including ToLCNDV resistance. In both F3 and F4 generations, per cent disease index, fruit weight and vine length exhibited high heritability and genetic advance as per cent of mean. In F4 generation, yield per plant, fruit girth, fruit length, number of branches and number of fruits per plant also showed high heritability and genetic advance as per cent of mean. Yield related traits viz., sex ratio, number of fruits per vine, yield per plant and vine length exhibited complementary gene action. Days to first harvest and crop duration demonstrated negatively skewed distribution suggesting duplicate gene action and rapid genetic gain under selection. Leptokurtic distribution in yield per plant and associated traits implied the involvement of few genes for genetic inheritance of the above traits. Significant positive associations were found between yield per plant and number of fruits per plant, fruit weight and number of branches per plant suggesting potential early selection for these characters to enhance overall yield

    Characterization of Salivary Glutathione reductase in Normal Individuals and its Implications on Smokers

    Abstract: The assay of saliva is an increasing area of research with implications for basic and clinical purposes. Although this biological fluid is easy to manipulate and collect, careful attention must be directed to limit variation in specimen integrity. In this study, glutathione reductase (GR) activity of saliva obtained from smokers and non smokers of both the sex of various age groups were assessed. The investigation of salivary GR from non smokers revealed a pH optimum of 6.8, temperature optimum as 37ºC and a low K m of 0.058 mM for the substrate (Oxidized glutathione, GSSG). A significant reduction in the salivary GR activity has been observed from smokers of both acute and chronic than the non smokers. A drastic decrease in the GR activity was noticed in chronic smokers than the acute smokers, proving the possibility of utilizing the enzyme as a diagnostic biomarker for detecting the oral, throat and neck cancers. This optimized developed protocol was also found to be simple and cost effective

    A survey on the knowledge, perceptions and practices regarding unwanted medicine disposal among pharmacists in Sri Lanka

    Background: Unwanted medicines are defined as expired, unused, damaged or contaminated pharmaceutical products. Improper disposal of unwanted medicines leads to many health and environmental hazards. The World Health Organisation recommends that unwanted medicines should always be disposed properly. The main objective of this study was to assess the knowledge, practices and perceptions on the disposal of unwanted medicines among pharmacists in Sri Lanka.Methods: A cross-sectional study was carried out among pharmacists in 40 private retail pharmacies in the Northern, Eastern and the Western provinces within a period of three months. The pharmacies were selected via stratified randomised sampling in each district. The most experienced pharmacist in each pharmacy was recruited for data collection. A pre-tested, self-administered questionnaire was used. The ethics approval was obtained (Ref: EC-12-190). The data was represented using simple descriptive statistics.Results: The data was collected from 40 pharmacies. Among the pharmacists, 65% were males. The majority answered that burning and landfill as the most appropriate methods of disposal for most of the types of medicinal waste. A significant number of pharmacists were not aware about the method of disposal for anti-infective agents and anti-neoplastic agents. The majority perceived the seriousness of environmental damage caused by disposal via trash or sink. A majority was not agreeing to have pharmacies as collecting centers for unwanted medicines. A discrepancy between the pharmacists’ perceptions and the practices was observed.Conclusions: The level of knowledge, practices and perceptions among pharmacists on unwanted medicines disposal was substandard and needs attention

    Genetic diversity and risk factors for the transmission of antimicrobial resistance across human, animals and environmental compartments in East Africa: a review.

    BACKGROUND The emergence and spread of antimicrobial resistance (AMR) present a challenge to disease control in East Africa. Resistance to beta-lactams, which are by far the most used antibiotics worldwide and include the penicillins, cephalosporins, monobactams and carbapenems, is reducing options for effective control of both Gram-positive and Gram-negative bacteria. The World Health Organization, Food and Agricultural Organization and the World Organization for Animal Health have all advocated surveillance of AMR using an integrated One Health approach. Regional consortia also have strengthened collaboration to address the AMR problem through surveillance, training and research in a holistic and multisectoral approach. This review paper contains collective information on risk factors for transmission, clinical relevance and diversity of resistance genes relating to extended-spectrum beta-lactamase-producing (ESBL) and carbapenemase-producing Enterobacteriaceae, and Methicillin-resistant Staphylococcus aureus (MRSA) across the human, animal and environmental compartments in East Africa. MAIN BODY The review of the AMR literature (years 2001 to 2019) was performed using search engines such as PubMed, Scopus, Science Direct, Google and Web of Science. The search terms included 'antimicrobial resistance and human-animal-environment', 'antimicrobial resistance, risk factors, genetic diversity, and human-animal-environment' combined with respective countries of East Africa. In general, the risk factors identified were associated with the transmission of AMR. The marked genetic diversity due to multiple sequence types among drug-resistant bacteria and their replicon plasmid types sourced from the animal, human and environment were reported. The main ESBL, MRSA and carbapenem related genes/plasmids were the CTX-Ms (45.7%), SCCmec type III (27.3%) and IMP types (23.8%), respectively. CONCLUSION The high diversity of the AMR genes suggests there may be multiple sources of resistance bacteria, or the possible exchange of strains or a flow of genes amongst different strains due to transfer by mobile genetic elements. Therefore, there should be harmonized One Health guidelines for the use of antibiotics, as well as regulations governing their importation and sale. Moreover, the trend of ESBLs, MRSA and carbapenem resistant (CAR) carriage rates is dynamic and are on rise over time period, posing a public health concern in East Africa. Collaborative surveillance of AMR in partnership with regional and external institutions using an integrated One Health approach is required for expert knowledge and technology transfer to facilitate information sharing for informed decision-making

    The attention towards the utilization of sustainable feedstocks for polymer synthesis has grown exponentially in recent years. One of the spotlighted monomers derived from renewable resources is 2,5-furandicarboxylic acid (FDCA), one of the most promising bio-based monomers, due to its resemblance to petroleum-based terephthalic acid. Very interesting synthetic routes using this monomer have been reported in the last two decades. Combining the use of bio-based monomers and non-toxic chemicals via enzymatic polymerizations can lead to a robust and favorable approach towards a greener technology of bio-based polymer production. In this chapter, a brief introduction to FDCA-based monomers and enzymatic polymerizations is given, particularly focusing on furan-based polymers and their polymerization. In addition, an outline of the recent developments in the field of enzymatic polymerizations is discussed. </p
