    Forests can prevent and/or mitigate hydrogeomorphic hazards in mountainous landscapes. Their effect is particularly relevant in the case of shallow landslides phenomena, where plants decrease the water content of the soil and increase its mechanical strength. Although such an effect is well known from ages, its quantification is a relatively new challenge. The present dissertation faces the effect on hillslope stability of several forest species typical of Lombardy\u2019s mountain forests. Such effects are estimate in terms of additional root cohesion by the classical Wu (1976) and Waldron (1977) model (W&W) and by Fiber Bundle Model approach (FBM). Samples of European beech (Fagus sylvatica L.), Norway spruce (Picea abies (L.) Karst.), European larch (Larix decidua Mill.), sweet chestnut (Castanea sativa Mill.) and European hop-hornbeam (Ostrya carpinifolia Scop.), Sycamore Maple (Acer pseudoplatanus L.), Grey Alder or Speckled Alder (Alnus incana (L.) Moench), European Ash (Fraxinus excelsior L.), Manna Ash (Fraxinus ornus L.), Black Locust (Robinia pseudoacacia L.), were taken in different locations of Lombardy (Northern Italy) to estimate root tensile strength, Root Area Ratio the root cohesion distribution within the soil. Besides the increasing of knowledge concerning the ability of the mentioned species to reinforce soils, that are fundamental to estimate the stability of forested hillslopes in the Alps, the dissertation reaches some more general results. Referring to root tensile strength, the obtained results confirm that such property is related to root size by a power law relationship. Such relationship, however, is not invariant with species but shows dependence with site\u2019s characteristics, although the observed variability is very high and no specific factor has been resulted as a key factor. Considering roots with a diameter greater than 1 mm, European beech roots seem to be more resistant than Norway spruce, Sweet chestnut and European Ash. Referring to Root Area Ratio, the results herein presented showed a great variability within the same species at the same location and among different locations. Despite the number of rooted profiles investigated, such variability hindered the possibility to identify a specie-specific or site-specific behaviour. Concerning root cohesion modelling, the original W&W model seems to overestimate root reinforcement because it assumes that all the roots crossing the shear surface break at the same time. FBM approach, instead, accounting for the progressive breaking of roots represents a promising perspective in root cohesion evaluation. Root cohesion values present the same species and site variability according to RAR values. The values herein obtained, moreover, are consistent with the results of direct shear tests and back-analysis data and, finally, they are significant for slope stabilisation at depth where shallow landslides generally occur

    Detection of Wolbachia in the Tick Ixodes ricinus is Due to the Presence of the Hymenoptera Endoparasitoid Ixodiphagus hookeri

    The identification of micro-organisms carried by ticks is an important issue for human and animal health. In addition to their role as pathogen vectors, ticks are also the hosts for symbiotic bacteria whose impact on tick biology is poorly known. Among these, the bacterium Wolbachia pipientis has already been reported associated with Ixodes ricinus and other tick species. However, the origins of Wolbachia in ticks and their consequences on tick biology (known to be very diverse in invertebrates, ranging from nutritional symbionts in nematodes to reproductive manipulators in insects) are unknown. Here we report that the endoparasitoid wasp Ixodiphagus hookeri (Hymenoptera, Chalcidoidea, Encyrtidae) – strictly associated with ticks for their development - is infested at almost 100% prevalence by a W. pipientis strain belonging to a Wolbachia supergroup that has already been reported as associated with other hymenopteran parasitoids. In a natural population of I. ricinus that suffers high parasitism rates due to I. hookeri, we used specific PCR primers for both hymenopteran and W. pipientis gene fragments to show that all unfed tick nymphs parasitized by I. hookeri also harbored Wolbachia, while unparasitized ticks were Wolbachia-free. We demonstrated experimentally that unfed nymphs obtained from larvae exposed to I. hookeri while gorging on their vertebrate host also harbor Wolbachia. We hypothesize that previous studies that have reported W. pipientis in ticks are due to the cryptic presence of the endoparasitoid wasp I. hookeri. This association has remained hidden until now because parasitoids within ticks cannot be detected until engorgement of the nymphs brings the wasp eggs out of diapause. Finally, we discuss the consequences of this finding for our understanding of the tick microbiome, and their possible role in horizontal gene transfer among pathogenic and symbiotic bacteria

    The Guinea Pig as a model for sporadic Alzheimer's Disease (AD): the impact of cholesterol intake on expression of AD-related genes

    Extent: 12p.We investigated the guinea pig, Cavia porcellus, as a model for Alzheimer’s disease (AD), both in terms of the conservation of genes involved in AD and the regulatory responses of these to a known AD risk factor - high cholesterol intake. Unlike rats and mice, guinea pigs possess an Aβ peptide sequence identical to human Aβ. Consistent with the commonality between cardiovascular and AD risk factors in humans, we saw that a high cholesterol diet leads to up-regulation of BACE1 (β-secretase) transcription and down-regulation of ADAM10 (α-secretase) transcription which should increase release of Aβ from APP. Significantly, guinea pigs possess isoforms of AD-related genes found in humans but not present in mice or rats. For example, we discovered that the truncated PS2V isoform of human PSEN2, that is found at raised levels in AD brains and that increases γ-secretase activity and Aβ synthesis, is not uniquely human or aberrant as previously believed. We show that PS2V formation is up-regulated by hypoxia and a high-cholesterol diet while, consistent with observations in humans, Aβ concentrations are raised in some brain regions but not others. Also like humans, but unlike mice, the guinea pig gene encoding tau, MAPT, encodes isoforms with both three and four microtubule binding domains, and cholesterol alters the ratio of these isoforms. We conclude that AD-related genes are highly conserved and more similar to human than the rat or mouse. Guinea pigs represent a superior rodent model for analysis of the impact of dietary factors such as cholesterol on the regulation of AD-related genes.Mathew J. Sharman, Seyyed H.     BHPR research: qualitative1. Complex reasoning determines patients' perception of outcome following foot surgery in rheumatoid arhtritis

    Background: Foot surgery is common in patients with RA but research into surgical outcomes is limited and conceptually flawed as current outcome measures lack face validity: to date no one has asked patients what is important to them. This study aimed to determine which factors are important to patients when evaluating the success of foot surgery in RA Methods: Semi structured interviews of RA patients who had undergone foot surgery were conducted and transcribed verbatim. Thematic analysis of interviews was conducted to explore issues that were important to patients. Results: 11 RA patients (9 ♂, mean age 59, dis dur = 22yrs, mean of 3 yrs post op) with mixed experiences of foot surgery were interviewed. Patients interpreted outcome in respect to a multitude of factors, frequently positive change in one aspect contrasted with negative opinions about another. Overall, four major themes emerged. Function: Functional ability & participation in valued activities were very important to patients. Walking ability was a key concern but patients interpreted levels of activity in light of other aspects of their disease, reflecting on change in functional ability more than overall level. Positive feelings of improved mobility were often moderated by negative self perception ("I mean, I still walk like a waddling duck”). Appearance: Appearance was important to almost all patients but perhaps the most complex theme of all. Physical appearance, foot shape, and footwear were closely interlinked, yet patients saw these as distinct separate concepts. Patients need to legitimize these feelings was clear and they frequently entered into a defensive repertoire ("it's not cosmetic surgery; it's something that's more important than that, you know?”). Clinician opinion: Surgeons' post operative evaluation of the procedure was very influential. The impact of this appraisal continued to affect patients' lasting impression irrespective of how the outcome compared to their initial goals ("when he'd done it ... he said that hasn't worked as good as he'd wanted to ... but the pain has gone”). Pain: Whilst pain was important to almost all patients, it appeared to be less important than the other themes. Pain was predominately raised when it influenced other themes, such as function; many still felt the need to legitimize their foot pain in order for health professionals to take it seriously ("in the end I went to my GP because it had happened a few times and I went to an orthopaedic surgeon who was quite dismissive of it, it was like what are you complaining about”). Conclusions: Patients interpret the outcome of foot surgery using a multitude of interrelated factors, particularly functional ability, appearance and surgeons' appraisal of the procedure. While pain was often noted, this appeared less important than other factors in the overall outcome of the surgery. The present work estimates the effect of some forest species on hillslope stability in terms of additional root cohesion by means of a model based on the classical Wu and Waldron approach (Wu in Alaska Geotech Rpt No 5 Dpt Civ Eng Ohio State Univ Columbus, USA, 1976; Waldron in Soil Sci Soc Am J 41:843-849, 1977). The model is able to account for root distribution with depth and non-simultaneous root breaking. Samples of European beech (Fagus sylvatica L.), Norway spruce (Picea abies (L.) Karst.), European larch (Larix decidua Mill.), sweet chestnut (Castanea sativa Mill.) and European hop-hornbeam (Ostrya carpinifolia Scop.), were taken from different locations of Lombardy (Northern Italy) to estimate root tensile strength, the Root Area Ratio and the root cohesion distribution in the soil. The results show that, in spite of its dramatic variability within the same species at the same location and among different locations, root cohesion can be coherently interpreted using the proposed method. The values herein obtained are significant for slope stabilisation, are consistent with the results of direct shear tests and back-analysis data, and can be used for the estimation of the stability of forested hillslopes in the Alps
