    Exploring Brain-Derived Progenitor Cells as a therapeutic delivery system to Glioblastoma

    Glioblastoma (GBM) is a devastating incurable malignant brain cancer in need of new treatments. We have begun to investigate the feasibility of a primary adult cell type (Brain-Derived Progenitor Cells, BDPCs) as a novel therapeutic delivery system to GBM. Our objective was to track the viability of BDPCs after intratumoral infusion into syngeneic orthotopic rat GBM tumours using non-invasive bioluminescence imaging (BLI). We hypothesize rat BDPCs will survive greater than 1 week following infusion into orthotopic F98 GBM tumors. BDPCs harvested from the cortex of adult Fischer rats were expanded in culture then engineered to co-express firefly Luciferase for BLI as well as the fluorescence protein tdTomato. In vitro assays displayed consistent lentiviral engineering of transgenes as well as statistically significant GBM-homing by BDPCs (p \u3c 0.01). All animals showed in vivo BLI signal until the study’s endpoint, confirming viable BDPCs were still present. Histological examination revealed small numbers of fluorescent BDPCs at the tumours’ invading edges in frozen coronal sections

    Variable δ15N Diet-Tissue Discrimination Factors among Sharks: Implications for Trophic Position, Diet and Food Web Models

    The application of stable isotopes to characterize the complexities of a species foraging behavior and trophic relationships is dependent on assumptions of δ15N diet-tissue discrimination factors (∆15N). As ∆15N values have been experimentally shown to vary amongst consumers, tissues and diet composition, resolving appropriate speciesspecific ∆15N values can be complex. Given the logistical and ethical challenges of controlled feeding experiments for determining ∆ 15N values for large and/or endangered species, our objective was to conduct an assessment of a range of reported ∆ 15N values that can hypothetically serve as surrogates for describing the predator-prey relationships of four shark species that feed on prey from different trophic levels (i.e., different mean δ 15N dietary values). Overall, the most suitable species-specific ∆ 15N values decreased with increasing dietary-δ 15N values based on stable isotope Bayesian ellipse overlap estimates of shark and the principal prey functional groups contributing to the diet determined from stomach content analyses. Thus, a single ∆ 15N value was not supported for this speciose group of marine predatory fishes. For example, the ∆ 15N value of 3.7‰ provided the highest percent overlap between prey and predator isotope ellipses for the bonnethead shark (mean diet δ 15N = 9‰) whereas a ∆ 15N value \u3c 2.3‰ provided the highest percent overlap between prey and predator isotope ellipses for the white shark (mean diet δ 15N = 15‰). These data corroborate the previously reported inverse ∆ 15N-dietary δ 15N relationship when both isotope ellipses of principal prey functional groups and the broader identified diet of each species were considered supporting the adoption of different ∆ 15N values that reflect the predators’ δ 15N-dietary value. These findings are critical for refining the application of stable isotope modeling approaches as inferences regarding a species’ ecological role in their community will be influenced with consequences for conservation and management actions

    Future biogeochemical forcing in Eastern Siberia: cooling or warming?

    Over-proportional warming in the northern high latitudes, and large carbon stocks in boreal and (sub)arctic ecosystems have raised concerns as to whether substantial positive climate feedbacks from biogeochemical process responses should be expected. Such feedbacks occur if increasing temperatures lead to e.g., a net release of CO2 or CH4. However, temperature-enhanced emissions of biogenic volatile organic compounds (BVOC) have been shown to contribute to a cooling feedback via growth of secondary organic aerosol (SOA), and related aerosol forcings. Combining measurements in Eastern Siberia with model-based estimates of vegetation and permafrost dynamics, BVOC emissions and aerosol growth, we show here that the additional climate forcing from changes in ecosystem CO2 balance and BVOC-SOA interactions nearly cancel on a regional scale. The interactions between emissions and vegetation dynamics that underlie individual forcing estimates are complex and highlight the importance of addressing ecosystem-climate feedbacks in consistent, process-based model frameworks that account for a multitude of system processes

    Muon Catalyzed Fusion in 3 K Solid Deuterium

    Muon catalyzed fusion in deuterium has traditionally been studied in gaseous and liquid targets. The TRIUMF solid-hydrogen-layer target system has been used to study the fusion reaction rates in the solid phase of D_2 at a target temperature of 3 K. Products of two distinct branches of the reaction were observed; neutrons by a liquid organic scintillator, and protons by a silicon detector located inside the target system. The effective molecular formation rate from the upper hyperfine state of μd\mu d and the hyperfine transition rate have been measured: λ~(3/2)=2.71(7)stat.(32)syst.μ/s\tilde{\lambda}_(3/2)=2.71(7)_{stat.}(32)_{syst.} \mu/s, and λ~(3/2)(1/2)=34.2(8)stat.(1)syst.μ/s\tilde{\lambda}_{(3/2)(1/2)} =34.2(8)_{stat.}(1)_{syst.} \mu /s. The molecular formation rate is consistent with other recent measurements, but not with the theory for isolated molecules. The discrepancy may be due to incomplete thermalization, an effect which was investigated by Monte Carlo calculations.     A prospective study of asthma incidence and its predictors: the RHINE study.

Please click on the hyperlink in Additional LinkThe objective of this longitudinal study was to estimate the incidence rate of asthma, and to compare the incidence between subjects with or without baseline reporting of certain respiratory symptoms. A follow-up of the random population samples in the European Respiratory Health Survey (ECRHS) in Sweden, Norway, Denmark, Iceland and Estonia was conducted in 1999-2001, in a population aged 30-54 yrs at follow-up (n=14,731). Asthma was defined as reporting either asthma or physician-diagnosed asthma, and a reported year when asthma symptoms were first noticed. Incidence rates, incidence rate ratios and hazard ratios were calculated with 95% confidence intervals. The incidence rate of asthma was 2.2 cases per 1,000 person-yrs. The incidence was higher among females (2.9 cases.1,000 person-yrs(-1)) than among males (1.5 cases.1,000 person-yrs(-1)). When subjects with baseline reporting of wheezing were excluded, the incidence rate decreased to 1.7 cases.1,000 person-yrs(-1), with a further decrease to 1.5 cases.1,000 person-yrs(-1) after exclusion of subjects with wheezing, nocturnal dyspnoea, chest tightness and cough. There was a strong association between onset of asthma and wheezing at baseline. In this prospective, population-based study, the incidence rate of asthma in the whole population sample ranged 1.5-2.2.1,000 person-yrs(-1), with a higher incidence range among females. The incidence was dependent on the extent to which subjects with respiratory symptoms were excluded from follow-up.
