    A metabolic cage for the hindlimb suspended rat

    Hindlimb suspension has been successfully used to simulate the effects of microgravity in rats. The cage and suspension system developed by E. R. Holton is designed to produce a headward shift of fluid and unload the hindlimbs in rodents, causing changes in bone and muscle similar to those in animals and humans exposed to spaceflight. While the Holton suspension system simulates many of the conditions observed in the spaceflight animal, it does not provide for the collection of urine and feces needed to monitor some metabolic activities. As a result, only limited information has been gathered on the nutritional status, and the gastrointestinal and renal function of animals using that model. Although commercial metabolic cages are available, they are usually cylindrical and require a centrally located suspension system and thus, do not readily permit movement of the rats. The limited floor space of commercial cages may affect comparisons with studies using the Holton model which has more than twice the living space of most commercially available cages. To take advantage of the extra living space and extensive data base that has been developed with the Holton model, Holton's cage was modified to make urine and fecal collections possible

    Colonial palynomorphs from the Upper Ordovician of north-eastern Iran : ‘thalli’, coenobial Chlorophyceae (Hydrodictyaceae) or cyanobacteria?

    This study documents ‘colonial’ palynomorphs from the Upper Ordovician Ghelli Formation of north-eastern Iran. The aggregates of organic-walled microfossils come from the Katian Armoricochitina nigerica–Ancyrochitina merga chitinozoan biozones of this formation. The ‘colonial’ microfossils can be classified as acritarchs and/or cryptospores, but they cannot be attributed to a particular biological group. Some specimens resemble ‘thalli’ of putative spores, such as Grododowon orthogonalis Strother 2017. Other clusters may suggest an affinity to green algal groups, in particular to colonial chlorophyceaen algae, most probably belonging to Hydrodictyaceae. Some specimens also show morphological similarities with cyanobacterial groups. There is so far no evidence to relate these ‘colonial’ palynomorphs to primitive land plants, but we hypothesise that they were possibly produced by ancient green algal lineages with some kind of subaerial existence

    Effects of epitaxial strain on the growth mechanism of YBa2Cu3O7-x thin films in [YBa2Cu3O7-x / PrBa2Cu3O7-x] superlattices

    We report on the growth mechanism of YBa2Cu3O7-x (YBCO). Our study is based on the analysis of ultrathin, YBa2Cu3O7-x layers in c-axis oriented YBa2Cu3O7-x / PrBa2Cu3O7-x superlattices. We have found that the release of epitaxial strain in very thin YBCO layers triggers a change in the dimensionality of the growth mode. Ultrathin, epitaxially strained, YBCO layers with thickness below 3 unit cells grow in a block by block two dimensional mode coherent over large lateral distances. Meanwhile, when thickness increases, and the strain relaxes, layer growth turns into three dimensional, resulting in rougher layers and interfaces.Comment: 10 pages + 9 figures, accepted in Phys.     Air pollution exposure during critical time periods in gestation and alterations in cord blood lymphocyte distribution: a cohort of livebirths

    <p>Abstract</p> <p>Background</p> <p>Toxic exposures have been shown to influence maturation of the immune system during gestation. This study investigates the association between cord blood lymphocyte proportions and maternal exposure to air pollution during each gestational month.</p> <p>Methods</p> <p>Cord blood was analyzed using a FACSort flow cytometer to determine proportions of T lymphocytes (CD3<sup>+ </sup>cells and their subsets, CD4<sup>+ </sup>and CD8<sup>+</sup>), B lymphocytes (CD19<sup>+</sup>) and natural killer (NK) cells. Ambient air concentrations of 12 polycyclic aromatic hydrocarbons (PAH) and particulate matter < 2.5 micrometer in diameter (PM<sub>2.5</sub>) were measured using fixed site monitors. Arithmetic means of these pollutants, calculated for each gestational month, were used as exposure metrics. Data on covariates were obtained from medical records and questionnaires. Multivariable linear regression models were fitted to estimate associations between monthly PAH or PM<sub>2.5 </sub>and cord blood lymphocytes, adjusting for year of birth and district of residence and, in further models, gestational season and number of prior live births.</p> <p>Results</p> <p>The adjusted models show significant associations between PAHs or PM<sub>2.5 </sub>during early gestation and increases in CD3<sup>+ </sup>and CD4<sup>+ </sup>lymphocytes percentages and decreases in CD19<sup>+ </sup>and NK cell percentages in cord blood. In contrast, exposures during late gestation were associated with decreases in CD3<sup>+ </sup>and CD4<sup>+ </sup>fractions and increases in CD19<sup>+ </sup>and NK cell fractions. There was no significant association between alterations in lymphocyte distribution and air pollution exposure during the mid gestation.</p> <p>Conclusions</p> <p>PAHs and PM<sub>2.5 </sub>in ambient air may influence fetal immune development via shifts in cord blood lymphocytes distributions. Associations appear to differ by exposure in early versus late gestation.</p

    A multiple-tubes procedure is described for using PCR to determine the genotype of a very small DNA sample. I The procedure involves dividing the sample among several tubes, then ampwing and typing the contents of each tube separately. The results are analyzed by a statistical procedure which determines whether a genotype can be conclusively assigned to the DNA sample. Simulation studies show that this procedure usually gives correct results even when the number of double-stranded fragments in the sample is as small as 30. The procedure remains effective even in the presence of small amounts of laboratory contamination. We find that the multiple-tubes procedure is superior to the standard one-tube procedure, either when the sample is small or when laboratory contamination is a potential problem; and we recommend its use in these situations. Because the procedure is statistical, it allows the degree of certainty in the result to be quantified and may be useful in other PCR applications as well
