    Pharmacological and Nonpharmacological Factors Influencing Hypothalamic-Pituitary-Adrenocortical Axis Reactivity in Acutely Depressed Psychiatric In-patients, Measured by the Dex-CRH Test

    The most consistent biological findings in patients with depression are abnormalities in the hypothalamic-pituitary-adrenal (HPA)-axis, which can be measured using the combined dexamethasone-suppression/CRH-stimulation (Dex-CRH) test. The reactivity of the HPAaxis in this test, however, ranges over several orders of magnitude in depressed patients with comparable severity of symptoms. In this present study, we investigate which factors influence the magnitude of the response in the Dex-CRH test in 235 acutely depressed inpatients. We first examined the effects of common confounders shown to influence the HPA-axis, such as caffeine and nicotine consumption, acute stressors during the test, weight, gender, and age. Of all these variables, only female sex and nicotine consumption were positively correlated with the cortisol or ACTH response, respectively. As for the effects of psychopharmacological treatment, only the intake of carbamazepine and the fact of having relapsed under an established pharmacotherapy significantly increased the response in the Dex-CRH test, whereas the presence or absence of antidepressant treatment, the type of antidepressant treatment, or the number of ineffective antidepressant treatment trials during the index episode up to admission did not have any effect. We also found a positive correlation of the number of previous episodes, the overall HAM-D score and the severity of somatic/vegetative symptoms with the results in the Dex-CRH test. These results underline that in depressed patients this test is not majorly influenced by disease-unrelated factors. In addition, current antidepressant treatment does not appear to affect test outcome in the absence of clinical response. The influence of the number of previous episodes and relapse under pharmacotherapy suggests that HPA-axis reactivity may be altered by repetitive states of hypercortisolemia or continuous antidepressant treatment. Finally, more severe vegetative symptoms are associated with an enhanced HPA-axis activity

    Towards a consolidation of worldwide journal rankings - A classification using random forests and aggregate rating via data envelopment analysis

    AbstractThe question of how to assess research outputs published in journals is now a global concern for academics. Numerous journal ratings and rankings exist, some featuring perceptual and peer-review-based journal ranks, some focusing on objective information related to citations, some using a combination of the two. This research consolidates existing journal rankings into an up-to-date and comprehensive list. Existing approaches to determining journal rankings are significantly advanced with the application of a new classification approach, ‘random forests’, and data envelopment analysis. As a result, a fresh look at a publication׳s place in the global research community is offered. While our approach is applicable to all management and business journals, we specifically exemplify the relative position of ‘operations research, management science, production and operations management’ journals within the broader management field, as well as within their own subject domain

    Capturing the sounds of an urban greenspace

    Acoustic data can be a source of important information about events and the environment in modern cities. To date, much of the focus has been on monitoring noise pollution, but the urban soundscape contains a rich variety of signals about both human and natural phenomena. We describe the CitySounds project, which has installed enclosed sensor kits at several locations across a heavily used urban greenspace in the city of Edinburgh. The acoustic monitoring components regularly capture short clips in real-time of both ultrasonic and audible noises, for example encompassing bats, birds and other wildlife, traffic, and human. The sounds are complemented by collecting other data from sensors, such as temperature and relative humidity. To ensure privacy and compliance with relevant legislation, robust methods render completely unintelligible any traces of voice or conversation that may incidentally be overheard by the sensors. We have adopted a variety of methods to encourage community engagement with the audio data and to communicate the richness of urban soundscapes to a general audience

    A trans-acting locus regulates an anti-viral expression network and type 1 diabetes risk

    Combined analyses of gene networks and DNA sequence variation can provide new insights into the aetiology of common diseases that may not be apparent from genome-wide association studies alone. Recent advances in rat genomics are facilitating systems-genetics approaches. Here we report the use of integrated genome-wide approaches across seven rat tissues to identify gene networks and the loci underlying their regulation. We defined an interferon regulatory factor 7 (IRF7)-driven inflammatory network (IDIN) enriched for viral response genes, which represents a molecular biomarker for macrophages and which was regulated in multiple tissues by a locus on rat chromosome 15q25. We show that Epstein-Barr virus induced gene 2 (Ebi2, also known as Gpr183), which lies at this locus and controls B lymphocyte migration, is expressed in macrophages and regulates the IDIN. The human orthologous locus on chromosome 13q32 controlled the human equivalent of the IDIN, which was conserved in monocytes. IDIN genes were more likely to associate with susceptibility to type 1 diabetes (T1D)-a macrophage-associated autoimmune disease-than randomly selected immune response genes (P = 8.85 x 10(-6)). The human locus controlling the IDIN was associated with the risk of T1D at single nucleotide polymorphism rs9585056 (P = 7.0 x 10(-10); odds ratio, 1.15), which was one of five single nucleotide polymorphisms in this region associated with EBI2 (GPR183) expression. These data implicate IRF7 network genes and their regulatory locus in the pathogenesis of T1D
