    Versatile compact atomic source for high resolution dual atom interferometry

    We present a compact 87^{87}Rb atomic source for high precision dual atom interferometers. The source is based on a double-stage magneto-optical trap (MOT) design, consisting of a 2-dimensional (2D)-MOT for efficient loading of a 3D-MOT. The accumulated atoms are precisely launched in a horizontal moving molasses. Our setup generates a high atomic flux (>1010>10^{10} atoms/s) with precise and flexibly tunable atomic trajectories as required for high resolution Sagnac atom interferometry. We characterize the performance of the source with respect to the relevant parameters of the launched atoms, i.e. temperature, absolute velocity and pointing, by utilizing time-of-flight techniques and velocity selective Raman transitions.Comment: uses revtex4, 9 pages, 12 figures, submitted to Phys.     Identification of LRRC8 heteromers as an essential component of the volume-regulated anion channel VRAC

    Regulation of cell volume is critical for many cellular and organismal functions, yet the molecular identity of a key player, the volume-regulated anion channel VRAC, has remained unknown. A genome-wide siRNA screen in mammalian cells identified LRRC8A as a VRAC component. LRRC8A formed heteromers with other LRRC8 multispan membrane proteins. Genomic disruption of LRRC8A ablated VRAC currents. Cells with disruption of all five LRRC8 genes required LRRC8A co-transfection with other LRRC8 isoforms to reconstitute VRAC currents. The isoform combination determined VRAC inactivation kinetics. Taurine flux and regulatory volume decrease also depended on LRRC8 proteins. Our work shows that VRAC defines a class of anion channels, suggests that VRAC is identical to the volume-sensitive organic osmolyte/anion channel VSOAC, and explains the heterogeneity of native VRAC currents

    Thermal responses of single zone offices on existing near-extreme summer weather data

    There have been a number of attempts in the past to define “near extreme” weather for facilitating overheating analysis in free running buildings. The most recently efforts include CIBSE latest release of Design Summer Year (DSY) weather using multiple complete weather years and a newly proposed composite DSY. This research aims to assess how various single zone offices respond to these new definitions of near extreme weathers. Parametric studies were carried out on single zone offices through which four sampling sets of models were employed to examine the thermal responses of dry bulb temperature, global solar radiation & wind speed collectively. London weather data from 1976 to 1995 were used and the overheating assessments were made based on CIBSE Guide A & BS EN 15251. The research discovers that solar radiation and wind both influence the predicted indoor warmth with solar radiation has obvious stronger impacts than wind. No perfect correlation was found from observation and Spearman’s rank order analysis on the ranks between the weather warmth and the predicted indoor warmth. The ranks made using multiple weather parameters show better correlation than some of the dry bulb temperature only metrics. The research also discovers that the Test Reference Year weather behaves warmer than expected. It is also found that a single complete year can not represent the near-extreme consistently and there is no evidence a composite DSY is better statistically. These findings support the notion of using multiple complete warm weather years for overheating assessments

    Pulmonary vasoconstrictor action of KCNQ potassium channel blockers

    KCNQ channels have been widely studied in the nervous system, heart and inner ear, where they have important physiological functions. Recent reports indicate that KCNQ channels may also be expressed in portal vein where they are suggested to influence spontaneous contractile activity. The biophysical properties of K+ currents mediated by KCNQ channels resemble a current underlying the resting K+ conductance and resting potential of pulmonary artery smooth muscle cells. We therefore investigated a possible role of KCNQ channels in regulating the function of pulmonary arteries by determining the ability of the selective KCNQ channel blockers, linopirdine and XE991, to promote pulmonary vasoconstriction. Linopirdine and XE991 both contracted rat and mouse pulmonary arteries but had little effect on mesenteric arteries. In each case the maximum contraction was almost as large as the response to 50 mM K+. Linopirdine had an EC50 of around 1 μM and XE991 was almost 10-fold more potent. Neither removal of the endothelium nor exposure to phentolamine or α,β-methylene ATP, to block α1-adrenoceptors or P2X receptors, respectively, affected the contraction. Contraction was abolished in Ca2+-free solution and in the presence of 1 μM nifedipine or 10 μM levcromakalim

    International Public Health Research Involving Interpreters: a Case Study from Bangladesh

    Background: Cross-cultural and international research are important components of public health research, but the challenges of language barriers and working with interpreters are often overlooked, particularly in the case of qualitative research. Methods: A case-study approach was used to explore experiences of working with an interpreter in Bangladesh as part of a research project investigating women's experiences of emergency obstetric care. The case study: Data from the researcher's field notes provided evidence of experiences in working with an interpreter and show how the model of interviewing was adapted over time to give a more active role to the interpreter. The advantages of a more active role were increased rapport and "flow" in interviews. The disadvantages included reduced control from the researcher's perspective. Some tensions between the researcher and interpreter remained hard to overcome, irrespective of the model used. Independent transcription and translation of the interviews also raised questions around accuracy in translation. Conclusion: The issues examined in this case study have broader implications for public health research. Further work is needed in three areas: 1) developing effective relationships with interpreters; 2) the impact of the interpreter on the research process; and 3) the accuracy of the translation and level of analysis needed in any specific public health research. Finally, this paper highlights the importance to authors of reflecting on the potential impact of translation and interpretation on the research process when disseminating their research
