    Functional crosstalk in culture between macrophages and trigeminal sensory neurons of a mouse genetic model of migraine

    Background: Enhanced activity of trigeminal ganglion neurons is thought to underlie neuronal sensitization facilitating the onset of chronic pain attacks, including migraine. Recurrent headache attacks might establish a chronic neuroinflammatory ganglion profile contributing to the hypersensitive phenotype. Since it is difficult to study this process in vivo, we investigated functional crosstalk between macrophages and sensory neurons in primary cultures from trigeminal sensory ganglia of wild-type (WT) or knock-in (KI) mice expressing the Cacna1a gene mutation (R192Q) found in familial hemiplegic migraine-type 1. After studying the number and morphology of resident macrophages in culture, the consequences of adding host macrophages on macrophage phagocytosis and membrane currents mediated by pain-transducing P2X3 receptors on sensory neurons were examined.Results: KI ganglion cultures constitutively contained a larger number of active macrophages, although no difference in P2X3 receptor expression was found. Co-culturing WT or KI ganglia with host macrophages (active as much as resident cells) strongly stimulated single cell phagocytosis. The same protocol had no effect on P2X3 receptor expression in WT or KI co-cultures, but it largely enhanced WT neuron currents that grew to the high amplitude constitutively seen for KI neurons. No further potentiation of KI neuronal currents was observed.Conclusions: Trigeminal ganglion cultures from a genetic mouse model of migraine showed basal macrophage activation together with enhanced neuronal currents mediated by P2X3 receptors. This phenotype could be replicated in WT cultures by adding host macrophages, indicating an important functional crosstalk between macrophages and sensory neurons. \ua9 2012 Franceschini et al.; licensee BioMed Central Ltd

    Addressing the persistent inequities in immunization coverage.

    A key focus of the health-related sustainable development goal (SDG) 3 is universal health coverage (UHC), including access to safe, effective, quality, and affordable essential medicines and vaccines. However, the challenges to achieving UHC are substantial, especially with increased demands on the health sector and with most budgets being static or shrinking. Immunization programmes have been successful in reaching children worldwide. For example, 86% of the world’s infants had received three doses of diphtheria-tetanus-pertussis (DTP3) vaccine in 2018. The experiences from such programmes can contribute to UHC, and as these programmes strive to adapt to new global strategic frameworks, such as Gavi, the Vaccine Alliance’s strategy Gavi and the World Health Organization’s (WHO) Immunization Agenda 2030, these efforts can inform the progressive realization of UHC. Immunization programmes that can sustain regular levels of contact between health providers and beneficiaries at the community level have enabled new vaccines to be added to routine immunization schedules and other interventions to be delivered to children and their families. In addition, experiences from both polio campaigns and the child health days strategy show that incorporating additional interventions into campaigns can increase coverage of these interventions as well as of vaccinations

    Lambda production in central Pb+Pb collisions at CERN-SPS energies

    In this paper we present recent results from the NA49 experiment for Λ\Lambda and Λˉ\bar{\Lambda} hyperons produced in central Pb+Pb collisions at 40, 80 and 158 A\cdotGeV. Transverse mass spectra and rapidity distributions for Λ\Lambda are shown for all three energies. The shape of the rapidity distribution becomes flatter with increasing beam energy. The multiplicities at mid-rapidity as well as the total yields are studied as a function of collision energy including AGS measurements. The ratio Λ/π\Lambda/\pi at mid-rapidity and in 4π\pi has a maximum around 40 A\cdotGeV. In addition, Λˉ\bar{\Lambda} rapidity distributions have been measured at 40 and 80 A\cdotGeV, which allows to study the Λˉ\bar{\Lambda}/Λ\Lambda ratio.Comment: SQM proceedings. J. Phys. G: Nucl.     Speech Spectrum's Correlation with Speakers' Eysenck Personality Traits

    The current study explored the correlation between speakers' Eysenck personality traits and speech spectrum parameters. Forty-six subjects completed the Eysenck Personality Questionnaire. They were instructed to verbally answer the questions shown on a computer screen and their responses were recorded by the computer. Spectrum parameters of /sh/ and /i/ were analyzed by Praat voice software. Formant frequencies of the consonant /sh/ in lying responses were significantly lower than that in truthful responses, whereas no difference existed on the vowel /i/ speech spectrum. The second formant bandwidth of the consonant /sh/ speech spectrum was significantly correlated with the personality traits of Psychoticism, Extraversion, and Neuroticism, and the correlation differed between truthful and lying responses, whereas the first formant frequency of the vowel /i/ speech spectrum was negatively correlated with Neuroticism in both response types. The results suggest that personality characteristics may be conveyed through the human voice, although the extent to which these effects are due to physiological differences in the organs associated with speech or to a general Pygmalion effect is yet unknown

    Evaluation of a new arterial pressure-based cardiac output device requiring no external calibration

    <p>Abstract</p> <p>Background</p> <p>Several techniques have been discussed as alternatives to the intermittent bolus thermodilution cardiac output (CO<sub>PAC</sub>) measurement by the pulmonary artery catheter (PAC). However, these techniques usually require a central venous line, an additional catheter, or a special calibration procedure. A new arterial pressure-based cardiac output (CO<sub>AP</sub>) device (FloTrac™, Vigileo™; Edwards Lifesciences, Irvine, CA, USA) only requires access to the radial or femoral artery using a standard arterial catheter and does not need an external calibration. We validated this technique in critically ill patients in the intensive care unit (ICU) using CO<sub>PAC </sub>as the method of reference.</p> <p>Methods</p> <p>We studied 20 critically ill patients, aged 16 to 74 years (mean, 55.5 ± 18.8 years), who required both arterial and pulmonary artery pressure monitoring. CO<sub>PAC </sub>measurements were performed at least every 4 hours and calculated as the average of 3 measurements, while CO<sub>AP </sub>values were taken immediately at the end of bolus determinations. Accuracy of measurements was assessed by calculating the bias and limits of agreement using the method described by Bland and Altman.</p> <p>Results</p> <p>A total of 164 coupled measurements were obtained. Absolute values of CO<sub>PAC </sub>ranged from 2.80 to 10.80 l/min (mean 5.93 ± 1.55 l/min). The bias and limits of agreement between CO<sub>PAC </sub>and CO<sub>AP </sub>for unequal numbers of replicates was 0.02 ± 2.92 l/min. The percentage error between CO<sub>PAC </sub>and CO<sub>AP </sub>was 49.3%. The bias between percentage changes in CO<sub>PAC </sub>(ΔCO<sub>PAC</sub>) and percentage changes in CO<sub>AP </sub>(ΔCO<sub>AP</sub>) for consecutive measurements was -0.70% ± 32.28%. CO<sub>PAC </sub>and CO<sub>AP </sub>showed a Pearson correlation coefficient of 0.58 (<it>p </it>< 0.01), while the correlation coefficient between ΔCO<sub>PAC </sub>and ΔCO<sub>AP </sub>was 0.46 (<it>p </it>< 0.01).</p> <p>Conclusion</p> <p>Although the CO<sub>AP </sub>algorithm shows a minimal bias with CO<sub>PAC </sub>over a wide range of values in an inhomogeneous group of critically ill patients, the scattering of the data remains relative wide. Therefore, the used algorithm (V 1.03) failed to demonstrate an acceptable accuracy in comparison to the clinical standard of cardiac output determination.</p

    Peer assessing individual contributions in a group project

    Graduates are expected to have good academic knowledge but also the professional skills required in the workplace. One such ‘soft’ skill is the ability to give constructive criticism and provide meaningful but professional feedback. This is particularly relevant when working in a team within industry, where peers need to influence each other to improve their project outcomes and chances of success. The development of student’s skills to generate such feedback should be supported within higher education. Specifically the IPAC Consortium investigates the use of Individual Peer Assessed Contribution to group work. In this context, students create an output directed to their own peers (i.e. a form of external-facing assessment), and prepares students for similar practices in industry. This paper, linked to the roundtable session on external-facing assessments proposed by Grindle and Tong, investigates staff and student perceptions on such practice.
