Hisztamin hatása a sejtdifferenciációra, összehasonlító vizsgálatok tumor - és embrionális őssejteken = Effect of histamine on cell differentiation, comparative study of tumor-and embronic stem cells
Célkitűzésünk a genetikailag hisztamin hiányos és WT dermatofibrosarcoma tumorsejtek és embrionális őssejtek differenciációs markereinek összehasonlítása volt. Az indukált tumorokból alapított sejttenyészetekből tumor spheroid formájában fenntartható klónokat hoztunk létre, melyek jellemzőikben hasonlítottak az embrionális testet (EB) képző őssejtekre. A továbbiakban microarray analízissel a letapadó, illetve spheroidként/EB-ként növő sejtek génexpressziós mintázatát vizsgáltuk, különös tekintettel az egyedfejlődésben szerepet játszó, és a sejt önmegújulásért felelős transzkripciós faktorokra. Mivel az array adatok igen nagyszámú gén aktivitásában mutattak különbséget, ezért az Ingenuity pathway analízis nyomán csak a Wnt jelátviteli útra koncentráltunk a továbbiakban. Az így kiemelt Id2, Sox2, Nestin, Col2A1 génhálózat expressziós tendenciáit real-time PCR-rel és Western blottal próbáltuk validálni, azonban a 3 módszerrel detektált változások tendenciái nem vagy csak részben mutattak egyezést. Mivel a spheroid- és EB-képzésben a sejt-mátrix kapcsolatoknak is szerepe van (erre utalhat a Col2A1) ezért ebbe az irányba terjesztettük ki kutatásainkat, az adherens és spheroid/EB sejtek adhéziós viselkedését vizsgálva, különös tekintettel egy másik (a hisztaminnal összefüggésben már igazolt szerepű) mátrix komponensre, a fibulin5-re. A HDC KO sejtek fokozottabb adhézióval és fibulin5 expresszióval rendelkeznek, ez összhangban áll a korábban melanoma sejteken tett megfigyelésünkkel. | Our aim was to compare the differentiation markers of BalB/c WT and HDC KO dermatofibrosarcoma tumor cells and embryonic stem cells with the same genetic background. From the induced tumors cell cultures and non-adhesive spheroid forming clones were established, which were similar embryoid bodies (EBs). Subsequently microarray analyses on Agilent platform were carried out to determine the gene expression patterns of the adherent and spheroid or EB forming cells. We focused our interest to those transcription factors involved in cellular self-renewal and in early embryogenesis. As the array data showed differences in hundreds of genes, we concentrated our efforts - after using Ingenuity pathway analysis - the Wnt pathway, important in self-renewal and tumorigenesis. Id2, Sox2, Nestin and Col2A1 elements of this network were used for validation via real-time PCR and Western blotting. Unfortunately the tendencies observed here did not fit well the data derived from microarray analysis. Since cell-matrix connections are important in both spheroid and EB formation (reflected by the Col2A1 expression, too) we expand our efforts to study the adhesive behaviour of adherent and spheroid/body forming cells in connection with an other matrix component fibulin5, its role related to histamine has been proven earlier by us. The HDC KO cells had better adhesion characteristics and increased FBLN5 expression, which is in good correlation our earlier observations made on melanoma cells
Doctor-Patient Knowledge Transfer: Innovative Technologies and Policy Implications
The aim of this study was to empirically investigate the barriers in doctor-patient communication and knowledge
transfer and the role of innovative technologies in overcoming these barriers. We applied qualitative research
methods.
Our results show that patients extensively use information sources, primarily the Internet before the visits. Patients
regularly apply a self-diagnosis regarding their diseases. This implies several risks as many of them are not able to
properly interpret the found information and at the same time the information might not be reliable. To overcome
these risks efforts are required within the fields of technology developments for making web sites more reliable and
improvement of the health culture of patients, as well.
Our research identified the most significant barriers of doctor-patient communication including limited time, the
patients’ distress, inadequate health culture and prior knowledge, as well as poor communication skills of some
clinicians. Technology might help clinicians to use their limited time more effectively.
In the long term, innovative technology solutions might take over some tasks of the health care personnel if they
provide reliable health information adapted to the patient’s health, emotional and psychosocial status. Barriers of access to the new technology should be identified and addressed otherwise it would increase the already existing knowledge gap between patients and doctors
Comparison of working length control consistency between hand K-files and Mtwo NiTi rotary system
Background: The purpose of this study was to investigate the consistency of working length control between hand
instrumentation in comparison to engine driven Mtwo nickel-titanium rotary files.
Material and Methods: Forty extracted maxillary molars were selected and divided onto two parallel groups. The
working lengths of the mesiobuccal root canals were estimated. The teeth were fixed in a phantom head. The root
canal preparation was carried out group 1 (n=20) with hand K-files, (VDW, Munich, Germany) and group 2 (n=20)
with Mtwo instruments (VDW, Munich, Germany). Vestibulo-oral and mesio-distal directional x-ray images were
taken before the preparation with #10 K-file, inserted into the mesiobuccal root canal to the working length, and
after preparation with #25, #30 and #40 files. Working lenght changes were detected with measurements between
the radiological apex and the instrument tips.
Results: In the Mtwo group a difference in the working competency (
p
<0.05) could be noticed only in the vestibulo-
oral direction from #10 to #40 file. The hand instrument group showed a significant difference in working length
competency for each larger file size (
p
<0.05) (ANOVA). Regression analysis in the hand instrumentation group
indicated a working length decrease with a mean of 0,2 mm after each consecutive file size (
p
<0.01).
Conclusions: The outcome of our trial indicated a high consistency in working length control for root canal preparation under simulated clinical condition using Mtwo rotary files. Mtwo NiTi rotary file did therefore proved to be
more accurate in comparison to the conventional hand instrumentation
Innovation Management in Central and Eastern Europe: Technology Perspectives and EU Policy Implications
The purpose of this study is to investigate empirically the role of innovation activity in Central and Eastern
Europea (CEE). We also identified those internal and external factors, which might cause improvements in
innovation performance of CEE companies. Our main focus was on technology-based innovations within the
healthcare industry. We applied qualitative research methods. Our findings demonstrate that CEE companies within the healthcare industry have significant contribution to
European Union’s innovation performance. We found that key success factors of these organizations are based
on four elements: knowledge management, access to financial resources, managing formal and informal
networks, as well as achieving synergies between technological and non-technological innovations
Innovációmenedzsment és innovációs stratégiák – a vállalati tudás szerepe az innovációs folyamatokban (Innovation management and innovation strategies – the role of corporate knowledge in innovative processes)
A szerzők kutatásának célja, hogy az innovációmenedzsment és az innovációs stratégiák összefüggéseiben
rávilágítsanak a magyarországi vállalatokat jellemző stratégiai irányokra és azonosítsák a vállalati
tudás szerepét az innovációs folyamatokban. Az induktív logikán alapuló, kvalitatív interjúkra építkező
módszertan segítségével történt az adatok gyűjtése és elemzése. Kutatási eredményeik alapján megállapítható,
hogy a magyar vállalatok többsége tisztában van az innováció stratégiai jelentőségével és a tudás
innovációfokozó erejével. Azonban a bizonytalan szabályozási környezet, a negatív piaci hangulat, a finanszírozási forrásokhoz való hozzáférés és a globális verseny megnehezítik a hazai KKV-k innovációs
törekvéseit. Ugyanakkor a multinacionális vállalatok élen járnak mind a technológiai, mind pedig a nem
technológiai innovációk terén is. A nem technológiai innováció területén komoly fejlődési lehetőségek körvonalazódhatnak a magyar KKV-k számára is, hiszen ezek kevésbé költségesek és időigényesek, és a belső erőforrásokat felhasználva a szervezeti tudásbázisra is épülhetnek. ____ The aim of the authors’ research is to highlight the characteristic strategic directions of Hungarian companies and identify the role of corporate knowledge in the innovative processes in the context of innovation management and innovative strategies. The collection and analysis of data were helped by a methodology which is based on inductive logic and qualitative interviews. The diagnosis, based on there search results,was that the majority of Hungarian companies are aware of the strategic importance of innovation and the innovation enhancing power of knowledge
Társadalmi innováció a joggyakorlatban: Coaching Action Research a munkavédelemben
Kutatásunkban (1) kidolgoztunk egy olyan, big data alapú módszertant és adatbázist, amely segíti a vállalkozásokat a munkavédelmi szabályok értelmezésében; (2) egy pilot projekt keretében felmértük, hogy a legfontosabb érintettek körében milyen a fogadókészség ennek használatára, illetve milyen feltételek mellett használnák inkább az adatbázist; (3) egy coaching folyamat keretében segítenénk az adatbázis bevezetését, a használatának elterjesztését, valamint action research-öt folytatva monitoroznánk a használók reakcióit és értékelnénk a bevezetés tanulságait. A kutatás során mesterséges intelligencia alapú gépi tanítás (Support Vector Machine) segítéségével modellt tanítottunk arra, hogy a számítógépes algoritmus a jövőben képes legyen a balesetek osztályozását elvégezni

Microbiological investigations on the water of a thermal bath at Budapest
Thermal baths are unique aquatic environments combining a wide variety of natural and anthropogenic ecological factors, which also appear in their microbiological state. There is limited information on the microbiology of thermal baths in their complexity, tracking community shifts from the thermal wells to the pools. In the present study, the natural microbial community of well and pool waters in Gellért bath was studied in detail by cultivation-based techniques. To isolate bacteria, 10% R2A and minimal synthetic media (with “bath water”) with agar–agar and gellan gum were used after prolonged incubation time; moreover, polyurethane blocks covered with media were also applied. Strains were identified by sequencing their 16S rRNA gene after grouping them by amplified rDNA restriction analysis. From each sample, the dominance of Alphaproteobacteria was characteristic though their diversity differed among samples. Members of Actinobacteria, Firmicutes, Beta- and Gamma-proteobacteria, Deinococcus–Thermus, and Bacteroidetes were also identified. Representatives of Deinococcus–Thermus phylum appeared only in the pool water. The largest groups in the pool water belonged to the Tistrella and Chelatococcus genera. The most dominant member in the well water was a new taxon, its similarity to Hartmannibacter diazotrophicus as closest relative was 93.93%
Chemomechanical preparation by hand instrumentation and by Mtwo engine-driven rotary files, an ex vivo study
Objective: To compare the disinfecting efficacy of the sodium hypochlorite irrigation by root canal preparation with
stainless steel hand files, taper 0.02 and nickel-titanium Mtwo files with taper 0.04-0.06.
Study design: 40 extracted human teeth were sterilized, and then inoculated with Enterococcus faecalis (ATCC
29212). After 6 day incubation time the root canals were prepared by hand with K-files (n=20) and by engine-driven
Mtwo files (VDW, Munich, Germany) (n=20). Irrigation was carried out with 2.5% NaOCl in both cases. Samples
were taken and determined in colony forming units (CFU) from the root canals before and after the preparation
with instruments #25 and #35.
Results: Significant reduction in bacterial count was determined after filing at both groups. The number of bacteria
kept on decreasing with the extension of apical preparation diameter. There was no significant difference between
the preparation sizes in the bacterial counts after hand or engine-driven instrumentation at the same apical size.
Statistical analysis was carried out with Mann-Whitney test, paired t-test and independent sample t-test.
Conclusions: Significant reduction in CFU was achieved after the root canal preparation completed with 2.5%
NaOCl irrigation, both with stainless steel hand or nickel-titanium rotary files. The root canal remained slightly
infected after chemo mechanical preparation in both groups
