    Design and Fabrication of Three-Dimensional Scaffolds for Tissue Engineering of Human Heart Valves

    We developed a new fabrication technique for 3-dimensional scaffolds for tissue engineering of human heart valve tissue. A human aortic homograft was scanned with an X-ray computer tomograph. The data derived from the X-ray computed tomogram were processed by a computer-aided design program to reconstruct a human heart valve 3-dimensionally. Based on this stereolithographic model, a silicone valve model resembling a human aortic valve was generated. By taking advantage of the thermoplastic properties of polyglycolic acid as scaffold material, we molded a 3-dimensional scaffold for tissue engineering of human heart valves. The valve scaffold showed a deviation of only +/- 3-4% in height, length and inner diameter compared with the homograft. The newly developed technique allows fabricating custom-made, patient-specific polymeric cardiovascular scaffolds for tissue engineering without requiring any suture materials. Copyright (c) 2008 S.     Culture in embryonic kidney serum and xeno-free media as renal cell carcinoma and renal cell carcinoma cancer stem cells research model

    The use of fetal bovine serum hinders obtaining reproducible experimental results and should also be removed in hormone and growth factor studies. In particular hormones found in FBS act globally on cancer cell physiology and influence transcriptome and metabolome. The aim of our study was to develop a renal carcinoma serum free culture model optimized for (embryonal) renal cells in order to select the best study model for downstream auto-, para- or endocrine research. Secondary aim was to verify renal carcinoma stem cell culture for this application. In the study, we have cultured renal cell carcinoma primary tumour cell line (786-0) as well as human kidney cancer stem cells in standard 2D monolayer cultures in Roswell Park Memorial Institute Medium or Dulbecco’s Modified Eagle’s Medium and Complete Human Kidney Cancer Stem Cell Medium, respectively. Serum-free, animal-component free Human Embryonic Kidney 293 media were tested. Our results revealed that xeno-free embryonal renal cells optimized culture media provide a useful tool in RCC cancer biology research and at the same time enable effective growth of RCC. We propose bio-mimic RCC cell culture model with specific serum-free and xeno-free medium that promote RCC cell viability

    Modelling the Galactic Bar Using Red Clump Giants

    The color-magnitude diagrams of 7×105\sim 7 \times 10^5 stars obtained for 12 fields across the Galactic bulge with the OGLE project reveal a well-defined population of bulge red clump giants. We find that the distributions of the apparent magnitudes of the red clump stars are systematically fainter when moving towards lower galactic ll fields. The most plausible explanation of this distinct trend is that the Galactic bulge is a bar, whose nearest end lies at positive galactic longitude. We model this Galactic bar by fitting for all fields the observed luminosity functions in the red clump region of the color-magnitude diagram. We find that almost regardless of the analytical function used to describe the 3-D stars distribution of the Galactic bar, the resulting models have the major axis inclined to the line of sight by 2030deg20-30\deg, with axis ratios corresponding to x0:y0:z0=3.5:1.5:1x_0:y_0:z_0=3.5:1.5:1. This puts a strong constraint on the possible range of the Galactic bar models.     Identifying Very Metal-Rich Stars with Low-Resolution Spectra: Finding Planet-Search Targets

    We present empirical calibrations that estimate stellar metallicity, effective temperature and surface gravity as a function of Lick/IDS indices. These calibrations have been derived from a training set of 261 stars for which (1) high-precision measurements of [Fe/H], T_eff and log g have been made using spectral-synthesis analysis of HIRES spectra, and (2) Lick indices have also been measured. Our [Fe/H] calibration, which has precision 0.07 dex, has identified a number of bright (V < 9) metal-rich stars which are now being screened for hot Jupiter-type planets. Using the Yonsei-Yale stellar models, we show that the calibrations provide distance estimates accurate to 20% for nearby stars. This paper outlines the second tier of the screening of planet-search targets by the N2K Consortium, a project designed to identify the stars most likely to harbor extrasolar planets. Discoveries by the N2K Consortium include the transiting hot Saturn HD 149026 b (Sato et al. 2005, astro-ph/0507009) and HD 88133 b (Fischer et al. 2005). See Ammons et al.     Histone H4 lysine 16 acetylation controls central carbon metabolism and diet-induced obesity in mice

    Noncommunicable diseases (NCDs) account for over 70% of deaths world-wide. Previous work has linked NCDs such as type 2 diabetes (T2D) to disruption of chromatin regulators. However, the exact molecular origins of these chronic conditions remain elusive. Here, we identify the H4 lysine 16 acetyltransferase MOF as a critical regulator of central carbon metabolism. High-throughput metabolomics unveil a systemic amino acid and carbohydrate imbalance in Mof deficient mice, manifesting in T2D predisposition. Oral glucose tolerance testing (OGTT) reveals defects in glucose assimilation and insulin secretion in these animals. Furthermore, Mof deficient mice are resistant to diet-induced fat gain due to defects in glucose uptake in adipose tissue. MOF-mediated H4K16ac deposition controls expression of the master regulator of glucose metabolism, Pparg and the entire downstream transcriptional network. Glucose uptake and lipid storage can be reconstituted in MOF-depleted adipocytes in vitro by ectopic Glut4 expression, PPARγ agonist thiazolidinedione (TZD) treatment or SIRT1 inhibition. Hence, chronic imbalance in H4K16ac promotes a destabilisation of metabolism triggering the development of a metabolic disorder, and its maintenance provides an unprecedented regulatory epigenetic mechanism controlling diet-induced obesity

    An Investigation of Be/X-ray Pulsars with OGLE-III Data

    We have studied five seasons of OGLE-III data for eight SMC Be/X-ray pulsars for which no other survey data were available. We have determined orbital periods for four of these binary systems, one of which also shows nonradial pulsations. Optical identification of SMC X-2 is reconsidered, but no periods were found for either of the two possible candidates

    Discovery of a high state AM CVn binary in the Galactic Bulge Survey

    We report on the discovery of a hydrogen-deficient compact binary (CXOGBS J175107.6-294037) belonging to the AM CVn class in the Galactic Bulge Survey. Deep archival X-ray observations constrain the X-ray positional uncertainty of the source to 0.57 arcsec, and allow us to uniquely identify the optical and UV counterpart. Optical spectroscopic observations reveal the presence of broad, shallow He i absorption lines while no sign of hydrogen is present, consistent with a high state system. We present the optical lightcurve from Optical Gravitational Lensing Experiment monitoring, spanning 15 years. It shows no evidence for outbursts; variability is present at the 0.2 mag level on timescales ranging from hours to weeks. A modulation on a timescale of years is also observed. A Lomb-Scargle analysis of the optical lightcurves shows two significant periodicities at 22.90 and 23.22 min. Although the physical interpretation is uncertain, such timescales are in line with expectations for the orbital and superhump periods. We estimate the distance to the source to be between 0.5 - 1.1 kpc. Spectroscopic follow-up observations are required to establish the orbital period, and to determine whether this source can serve as a verification binary for the eLISA gravitational wave mission.Comment: Accepted for publication in MNRAS Letter

    The large magnitude and the sign correlation effect in the parity non-conserving resonant scattering of epithermal neutrons from 232^{232}Th is discussed in terms of a non-collective 2p1h2p-1h local doorway model.     A Post-AGB Star in the Small Magellanic Cloud Observed with the Spitzer Infrared Spectrograph

    We have observed an evolved star with a rare combination of spectral features, MSX SMC 029, in the Small Magellanic Cloud (SMC) using the low-resolution modules of the Infrared Spectrograph on the Spitzer Space Telescope. A cool dust continuum dominates the spectrum of MSX SMC 029. The spectrum also shows both emission from polycyclic aromatic hydrocarbons (PAHs) and absorption at 13.7 micron from C2H2, a juxtaposition seen in only two other sources, AFGL 2688 and IRAS 13416-6243, both post-asymptotic giant branch (AGB) objects. As in these sources, the PAH spectrum has the unusual trait that the peak emission in the 7-9 micron complex lies beyond 8.0 micron. In addition, the 8.6 micron feature has an intensity as strong as the C-C modes which normally peak between 7.7 and 7.9 micron. The relative flux of the feature at 11.3 micron to that at 8 micron suggests that the PAHs in MSX SMC 029 either have a low ionization fraction or are largely unprocessed. The 13-16 micron wavelength region shows strong absorption features similar to those observed in the post-AGB objects AFGL 618 and SMP LMC 11. This broad absorption may arise from the same molecules which have been identified in those sources: C2H2, C4H2, HC3N, and C6H6. The similarities between MSX SMC 029, AFGL 2688, and AFGL 618 lead us to conclude that MSX SMC 029 has evolved off the AGB in only the past few hundred years, making it the third post-AGB object identified in the SMC.Comment: 4 figures, Fig. 4 color; to appear in the 20 November 2006 Astrophysical Journal Letter
