Three-dimensional dust radiative-transfer models: The Pinwheel Nebula of WR104
We present radiative-transfer modelling of the dusty spiral Pinwheel Nebula
observed around the Wolf-Rayet/OB-star binary WR104. The models are based on
the three-dimensional radiative-transfer code TORUS, modified to include an
adaptive mesh that allows us to adequately resolve both the inner spiral turns
(sub-AU scales) and the outer regions of the nebula (distances of 10^4 AU from
the central source). The spiral model provides a good fit to both the spectral
energy distribution and Keck aperture masking interferometry, reproducing both
the maximum entropy recovered images and the visibility curves. We deduce a
dust creation rate of 8+-1 x 10^{-7} solar masses per year, corresponding to
approximately 2% by mass of the carbon produced by the Wolf-Rayet star.
Simultaneous modelling of the imaging and spectral data enables us to constrain
both the opening-angle of the wind-wind collision interface and the dust grain
size. We conclude that the dust grains in the inner part of the Pinwheel nebula
are small (~100A), in agreement with theoretical predictions, although we
cannot rule out the presence of larger grains (~1 micron) further from the
central binary. The opening angle of the wind-wind collision interface appears
to be about 40 degrees, in broad agreement with the wind parameters estimated
for the central binary. We discuss the success and deficiencies of the model,
and the likely benefits of applying similar techniques to the more the more
complex nebulae observed around other WR/O star binaries.Comment: 10 pages, accepted by MNRA
Alanine scanning mutagenesis of a high-affinity nitrate transporter highlights the requirement for glycine and asparagine residues in the two nitrate signature motifs
Common to all of the nitrate nitrite porter family are two conserved motifs in transmembrane helices 5 and 11 termed NS (nitrate signature) 1 and NS2. Although perfectly conserved substrate-interacting arginine residues have been described in transmembrane helices 2 and 8, the role of NSs has not been investigated. In the present study, a combination of structural modelling of NrtA (nitrate transporter from Aspergillus nidulans) with alanine scanning mutagenesis of residues within and around the NSs has been used to shed light on the probable role of conserved residues in the NSs. Models show that Asn 168 in NS1 and Asn 459 in NS2 are positioned approximately midway within the protein at the central pivot point in close proximity to the substrate-binding residues Arg 368 and Arg 87 respectively, which lie offset from the pivot point towards the cytoplasmic face. The Asn 168 /Arg 368 and Asn 459 /Arg 87 residue pairs are relatively widely separated on opposite sides of the probable substrate translocation pore. The results of the present study demonstrate the critical structural contribution of several glycine residues in each NS at sites of close helix packing. Given the relative locations of Asn 168 /Arg 368 and Asn 459 /Arg 87 pairs, the validity of the models and possible role of the NSs together with the substrate-binding arginine residues are discusse

Using chlorhexidine varnish to prevent early childhood caries in American Indian children
Objectives: To test the efficacy of 10% chlorhexidine (CHX) dental varnish applied to the mothers' dentition in preventing caries in American Indian children. Methods: This was a placebo‐controlled, double‐blind, randomized clinical trial. Mother–child pairs were enrolled when the child was 4.5‐6.0 months. Mothers received 4 weekly applications of the study treatment (CHX or placebo) followed by single applications when her child was age 12 and 18 months. Children received caries examinations at enrollment, 12, 18 and 24 months. Analyses were limited to the intent‐to‐treat (ITT) group: children whose mothers received the first study treatment and who received at least one post‐baseline exam. The outcome variable was the number of new carious surfaces (NNCS) at the child's last visit. Wilcoxon nonparametric and Fisher's exact tests were used to test differences between the active and placebo groups. Results: We randomized 414 mother–child pairs, with 367 (88.6%) included in the ITT group (active = 188, placebo = 179). The proportion of children caries‐free at their final exam was 51.1% and 50.8% for the active and placebo groups ( P > 0.99). The mean NNCS for the active and placebo groups was 3.82 (standard deviation [SD] = 8.18) and 3.80 (SD = 6.08), respectively ( P = 0.54). The proportion with NNCS > 6 was 18.1% for active children versus 27.9% for placebo (relative risk [RR] = 0.65, P = 0.03). The number needed to treat to shift one child from NNCS > 6 to a lower severity was 10.2. Conclusions: In this population CHX varnish did not reduce the mean NNCS or proportion of children with caries, but did reduce the proportion with severe caries.Peer Reviewedhttp://deepblue.lib.umich.edu/bitstream/2027.42/96744/1/jphd348.pd
Thermal activation between Landau levels in the organic superconductor -(BEDT-TTF)SFCHCFSO
We show that Shubnikov-de Haas oscillations in the interlayer resistivity of
the organic superconductor -(BEDT-TTF)SF
CHCFSO become very pronounced in magnetic fields ~60~T.
The conductivity minima exhibit thermally-activated behaviour that can be
explained simply by the presence of a Landau gap, with the
quasi-one-dimensional Fermi surface sheets contributing negligibly to the
conductivity. This observation, together with complete suppression of chemical
potential oscillations, is consistent with an incommensurate nesting
instability of the quasi-one-dimensional sheets.Comment: 6 pages, 4 figure
Inhomogeneous Superconductivity in Condensed Matter and QCD
Inhomogeneous superconductivity arises when the species participating in the
pairing phenomenon have different Fermi surfaces with a large enough
separation. In these conditions it could be more favorable for each of the
pairing fermions to stay close to its Fermi surface and, differently from the
usual BCS state, for the Cooper pair to have a non zero total momentum. For
this reason in this state the gap varies in space, the ground state is
inhomogeneous and a crystalline structure might be formed. This situation was
considered for the first time by Fulde, Ferrell, Larkin and Ovchinnikov, and
the corresponding state is called LOFF. The spontaneous breaking of the space
symmetries in the vacuum state is a characteristic feature of this phase and is
associated to the presence of long wave-length excitations of zero mass. The
situation described here is of interest both in solid state and in elementary
particle physics, in particular in Quantum Chromo-Dynamics at high density and
small temperature. In this review we present the theoretical approach to the
LOFF state and its phenomenological applications using the language of the

Relationship of an hRAD54 gene polymorphism (2290 C/T) in an Ecuadorian population with chronic myelogenous leukemia
The hRAD54 gene is a key member of the RAD52 epistasis group involved in repair of double-strand breaks (DSB) by homologous recombination (HR). Thus, alterations of the normal function of these genes could generate genetic instability, shifting the normal process of the cell cycle, leading the cells to develop into cancer. In this work we analyzed exon 18 of the hRAD54 gene, which has been previously reported by our group to carry a silent polymorphism, 2290 C/T (Ala730Ala), associated to meningiomas. We performed a PCR-SSCP method to detect the polymorphism in 239 samples including leukemia and normal control population. The results revealed that the 2290 C/T polymorphism has frequencies of 0.1 for the leukemia and 0.1 for the control group. These frequencies show no statistical differences. Additionally, we dissected the leukemia group in chronic myelogenous leukemia (CML) and acute lymphoblastic leukemia (ALL) to evaluate the polymorphism. The frequencies found in these subgroups were 0.14 for CML and 0.05 for ALL. We found statistically significant differences between CML patients and the control group (p < 0.05) but we did not find significant differences between ALL and the control group (p > 0.05). These results suggest a possible link between the 2290 C/T polymorphism of the hRAD54 gene and CML
CAS9 transcriptional activators for target specificity screening and paired nickases for cooperative genome engineering
Prokaryotic type II CRISPR-Cas systems can be adapted to enable targeted genome modifications across a range of eukaryotes.1–7. Here we engineer this system to enable RNA-guided genome regulation in human cells by tethering transcriptional activation domains either directly to a nuclease-null Cas9 protein or to an aptamer-modified single guide RNA (sgRNA). Using this functionality we developed a novel transcriptional activation–based assay to determine the landscape of off-target binding of sgRNA:Cas9 complexes and compared it with the off-target activity of transcription activator–like (TAL) effector proteins8, 9. Our results reveal that specificity profiles are sgRNA dependent, and that sgRNA:Cas9 complexes and 18-mer TAL effector proteins can potentially tolerate 1–3 and 1–2 target mismatches, respectively. By engineering a requirement for cooperativity through offset nicking for genome editing or through multiple synergistic sgRNAs for robust transcriptional activation, we suggest methods to mitigate off-target phenomena. Our results expand the versatility of the sgRNA:Cas9 tool and highlight the critical need to engineer improved specificity
