    THE IMPACT OF PRODUCT VARIETY ON RETAILER OPERATIONAL PERFORMANCE AND SALES: EVIDENCE FROM CHINA.

    Providing higher levels of product variety has long been shown to generate increased revenues for both retail and manufacturing firms. However, recent research has also shown that higher levels of product variety can have a negative impact on firm operational performance. This dissertation is a two essay study using archival data provided by a single retail firm based in Shanghai, China, on the effects of product variety on retailer inventory levels, stock out rates, and sales. The first essay examines how product variety, as measured by the number of SKUs carried in the retailer’s product category assortment, affects inventory levels, stock out rates, and sales. The second essay investigates whether different types of product variety (namely brands, sizes, and product lines) impacts store inventory levels, stock out rates, and sales differently. The first essay investigates how the size of the product assortment impacts inventory levels, stock out rates, and sales. Greater product variety has the potential to generate higher revenue for the retailer, but also brings the potential for more complications in inventory and supply chain management processes. While previous research has examined this relationship within a manufacturing context, no research has investigated the tradeoff in a retail context. Also, this research is the first to consider the impact of product variety on a firm’s inventory levels. This is an important inclusion as inventory levels directly impact the stock out rate of a retailer. Furthermore, this paper investigates whether characteristics of a product category, such as the hedonic or utilitarian nature of the product category, moderate the relationship between product variety, operational performance, and sales. Using simultaneous equations and a three stage least squares regression methodology, results suggest that product variety has a positive relationship with inventory levels, stock out rates, and sales. Finally, the relationship between a product categories’ stock out rate and sales is stronger for hedonic product categories than utilitarian product categories. In the second essay, this dissertation examines whether the relationship between product variety, inventory levels, stock out rates and sales differs between different types of product variety. In particular, this essay investigates whether brand variety has a larger impact on retailer inventory levels, stock out rates, and sales than do size variety or product line variety. Again, using a simultaneous equation model and a three stage least squares methodology, the results suggest that brand variety is associated with higher inventory levels, lower stock out rates and higher sales than size or product line variety in the retail context

    Measurement of three-dimensional temperature fields above heated surfaces by holographic interferometry

    Optical holography can be used to record multi-directional interferometric data, which provides a basis for measuring three-dimensional, asymmetric temperature or density fields in fluids. If continuous optical pathlength data are available over a 180[deg] angle of view, the temperature or density field is shown to be equal to the inverse Radon transform of the data. A procedure for computing limited-resolution reconstructions of the field in terms of discrete data collected over a limited angle of view is presented.The technique of holographic interferometry was used to map isothermal contours in the developing convective plume above heated, horizontal, rectangular surfaces.     Saccharomyces cerevisiae Rad9 Acts as a Mec1 Adaptor to Allow Rad53 Activation

    SummaryBackground: The DNA damage checkpoint is a protein kinase-based signaling system that detects and signals physical alterations in DNA. Despite having identified many components of this signaling cascade, the exact mechanisms by which checkpoint kinases are activated after DNA damage, as well as the role of the checkpoint mediators, remain poorly understood.Results: To elucidate the mechanisms that underlie the MEC1 and RAD9-dependent activation of Rad53, the Saccharomyces cerevisiae ortholog of Chk2, we mapped and characterized in vivo phosphorylation sites present on Rad53 after DNA damage by mass spectrometry. We find that Rad53 requires for its activation multisite phosphorylation on a number of typical and atypical Mec1 phosphorylation sites, thus confirming that Rad53 is a direct target of Mec1, the mammalian ATR homolog. Moreover, by using biochemical reconstitution experiments, we demonstrate that efficient and direct phosphorylation of Rad53 by Mec1 is only observed in the presence of purified Rad9, the archetypal checkpoint mediator. We find that the stimulatory activity of Rad9 requires a phospho- and FHA-dependent interaction with Rad53, which allows Rad53 to be recognized as a substrate for Mec1.Conclusions: Our results indicate that Rad9 acts as a bona fide signaling adaptor that enables Rad53 phosphorylation by Mec1. Given the high degree of conservation of checkpoint signaling in eukaryotes, we propose that one of the critical functions of checkpoint mediators such as MDC1, 53BP1, or Brca1 is to act as PIKK adaptors during the DNA damage response

    The Physiologic Effects of Multiple Simultaneous Electronic Control Device Discharges

    Objectives: Law enforcement and military personnel use electronic control devices to control non-compliant and actively resistive subjects. The TASER® Shockwave is a new electronic control device designed specifically as an area denial device capable of delivering multiple simultaneous discharges. This is the first study to examine the effects of multiple simultaneous device discharges in humans.Methods: Volunteers were exposed to multiple (two to three), simultaneous 5-second discharges from the Shockwave device to the chest, back, chest to abdomen, or thighs. Blood was analyzed before and after discharge for pH, lactate, potassium, creatine kinase (CK), and troponin. Continuous spirometry was performed before, during, and after the discharge. In addition, electrocardiograms (ECGs) before and after discharge were recorded, and echocardiography was used to determine the rhythm during discharge.Results: Small elevations of lactate occurred. Moderate increases in CK at 24 hours occurred and appeared to be related to the number of simultaneous discharges. There was a trend to a decrease in minute ventilation in the volunteers exposed to two simultaneous discharges, but it did not reach statistical significance. ECG changes only reflected an increase in vagal tone, and there was no evidence of capture by echocardiography. Five-second, simultaneous, multiple exposures to the TASER Shockwave device were reasonably tolerated by our human volunteers.Conclusion: Our study suggests that this device may have a reasonable risk/benefit ratio when used to protect an area from a threat.
