Pleyotropic antiaggregant effects of an innovative antiarrhythmic of class III SS-68, an indole derivative
A new indole derivative with the lab code-number SS-68 demonstrates a significant antiarrhythmic (anti-fibrillation) activity associated with the predominant influence on potassium and calcium conductivity of cardiomyocytes plasmalemmas. The preliminary data give evidence of SS-68 possessing an antiaggregant activity. The influence of SS-68 on the platelet formation stage of hemostasis was determined by assessment of the anti-platelet effect (measuring platelet aggregation with Born/ O’Brien method), the dynamics of intracellular concentration of calcium ions in platelets was studied with FURA-2/АМ fluorescent prob
Part I
The problem of heart rhythm disturbances is one of the most urgent topics of modern cardiology. According to the currently available concepts, 1,2- and 1,3-disubstituted aminoindole derivatives, which compound 2-phenyl-1-(3-pyrrolidin-1-il-propyl)-1H-indole hydrochloride (SS-68) belongs to, are a promising chemical group in terms of their cardio-pharmacological activit
Pharmacokinetic studies derived indole SS-68 with antiarrhythmic and antianginal properties
A method of quantitative determination of SS-68 derivative of indole in rabbit blood plasma by high performance liquid chromatography with tandem mass spectrometric detector (HPLCMS/ MS). Conducted pharmacokinetic studies SS-68 in the body of rabbits. Set the main pharmacokinetic parameters of the substance that allow you to optimize the future use of it’s as a potential drug in cardiology practic
Anti-inflammatory and analgesic propertiesof SS-68 indole derivative
The experiments with rats show that when administered intravenously (5 mg/kg) and intragastrically (5, 10, 15 and 20 mg/kg), SS-68 compound exerts an antiinflammatory effect (AIE) on models of acute inflammation, caused by carrageenin and serotonin, and exerts no AIE in case of edema induced by complete Freund‘s adjuvant (CFA) and histamine, except for intragastric administration at a dose of 20 mg/kg. By its antiphlogogenic effect in case of carrageenin and serotonin edemas, SS-68 is comparable to diclofenac (5, 10 and 15 mg/kg intragastrically) and indomethacin (10 mg kg intragastriccally), and exceeds piroxicam (20 mg/kg intragastrically). By reference to its therapeutic index (TI), in the first case SS-68 exceeds diclofenac, indomethacin and piroxicam 2.2, 15.6 and 5.6 times, and in the second case it exceeds them 1.8, 12.8 and 4.4 times, respectivel
The influence of the vagus nerve and indole derivative SS-68 on excitation processes in the SA node
The aim of the study was to identify the components of the takeover process of central rhythmogenesis by the SA node in the conditions of atrial fibrillation when stimulating the vagus nerve and using substance SS-6
Distance learning: opportunities and challenges in quarantine.
The urgent introduction of distance education into the learning process due to the COVID-19 pandemic has become a spontaneous experiment, requiring urgent decisions, effective mechanisms to implement into the educational process and analysis of ongoing educational process. The possibilities of modern digital technologies made it possible to quickly set up the information component of education and to controle student’s knowledge. A detailed analysis of the communication between the faculty members of the department among themselves and analysis of the features of teaching and self-education of students in the field of medical radiology and oncology based on answers and comments with using Google classroom, Socrative, Messenger Telegram, etc. was made. Issues of communicative interaction between faculty members and students, psychological aspects of distance communication, language of instruction, logistics of distance education and methodological problems are described in details. It has been suggested that distance education may be equally effective for theoretical subjects in full-time study form, but when studying clinical disciplines, the priority should be given to traditional methods of teaching, involving student's bedside training. Taking into account technical possibilities and high risk of development of extreme and emergency situations, it is suggested to introduce permanent readiness for distance education, to create all – Ukrainian distance imitation service of patients’ follow-up. The importance of using the state language in training as the basis for mastering the professional Ukrainian and English language as the basis for mastering international medical experience is indicated

The compound SS-68 attenuates electrophysiological effects of m3 cholinoreceptors stimulation in rat left atrial myocardium
In the present study we have investigated influence of SS-68, which has been previously demonstrated to produce potent antiarrhythmic effects in mammalian myocardium, on the electrophysiological effects of selective M3 cholinoreceptors stimulation in rat atrial myocardium. M3 receptors were activated with muscarinic agonist pilocarpine (10 jM) in the presence of highly selective blocker of M2 cholinoreceptors methoctramine (100 nM). In isolated left atrial preparations M3 stimulation produced reduction of action potential duration by 20 ± 2,2% of control duration. SS-68 (10 jM) decreased intensity of action potential shortening more than twice - to 7,9 ± 1,1% of control action potential duration. SS-68 also suppressed action potential shortening produced by pilocarpine in the absence of M2 antagonist. Thus, compound SS-68 attenuates effect of joint activation of M2 and M3 receptors as well as the effect of selective M3 stimulation
SS-68 IMPACT ON THE CONTRACTIVE ACTIVITY OF ISOLATED VESSELS
In the experiments on isolated circular segments of the thoracic aorta of a rat it has been proved that indole SS-66 derivative in 10-5 and 10-4 М concentrations can relax smooth muscle cells (SMS) due to its impact on the SMS potassium conduction thus facilitating their hyperpolarization. It results in the block of voltage-sensitive calcium channels, the decrease in calcium input and the SMS relaxing. Lack of the NO-synthase Nω-nitro-L-arginine blocker impact on this process indicates of the lack of the SS-68 impact on the processes connected with the endothelium-dependant SMS relaxation
