    Realistic Expectations with Brain Computer Interfaces

    PurposeThis paper describes the research underpinning the development and evaluation of a brain computer interface (BCI) system designed to be suitable for domestic use by people with acquired brain injury in order to facilitate control of their home environment. The purpose of the research is to develop a robust and user‐friendly BCI system which was customisable in terms of user ability, preferences and functionality. Specifically the human interface was designed to provide consistent visual metaphors in usage, while applications change, for example, from environmental control to entertainment and communications.Design/methodology/approachThe research took a user centred design approach involving representative end‐users throughout the design and evaluation process. A qualitative study adopting user interviews alongside interactive workshops highlighted the issues that needed to be addressed in the development of a user interface for such a system. User validation then underpinned prototype development.FindingsThe findings of the research indicate that while there are still significant challenges in translating working BCI systems from the research laboratories to the homes of individuals with acquired brain injuries, participants are keen to be involved in the deign and development of such systems. In its current stage of development BCI is multi‐facetted and uses complex software, which poses a significant usability challenge. This work also found that the performance of the BCI paradigm chosen was considerably better for those users with no disability than for those with acquired brain injury. Further work is required to identify how and whether this performance gap can be addressed.Research limitations/implicationsThe research had significant challenges in terms of managing the complexity of the hardware and software set‐up and transferring the working systems to be tested by participants in their home. Furthermore, the authors believe that the development of assistive technologies for the disabled user requires a significant additional level of personalisation and intensive support to the level normally required for non‐disabled users. Coupled with the inherent complexity of BCI, this leads to technology that does not easily offer a solution to both disabled and non‐disabled users.Originality/valueThe research contributes additional findings relating to the usability of BCI systems. The value of the work is to highlight the practical issues involved in translating such systems to participants where the acquired brain injury can impact on the ability of the participant to use the BCI system.</jats:sec

    G = E:What GWAS Can Tell Us about the Environment

    As our understanding of genetics has improved, genome-wide association studies (GWAS) have identified numerous variants associated with lifestyle behaviours and health outcomes. However, what is sometimes overlooked is the possibility that genetic variants identified in GWAS of disease might reflect the effect of modifiable risk factors as well as direct genetic effects. We discuss this possibility with illustrative examples from tobacco and alcohol research, in which genetic variants that predict behavioural phenotypes have been seen in GWAS of diseases known to be causally related to these behaviours. This consideration has implications for the interpretation of GWAS findings

    Enhanced Leak Detection

    A key requirement for Veeder-Root’s Enhanced Leak Detection System is that it be able to test in situ for the presence of leaks at gasoline dispensing facilities. Aside from the obvious issues of safety and lost product, this functionality is obligatory for compliance with environmental standards mandated by federal and state oversight bodies, such as the California State Water Resources Control Board (SWRCB). The SWRCB demands a testing procedure that includes conditions as close to operational as possible, while still using environmentally safe gases as a test fluid. Although the test parameters (e.g., pressure) are allowed to deviate from operating conditions in order to facilitate the test procedure, a prescribed rescaling of the test thresholds must then be applied to account for the deviation. Whether the test is run at operation conditions or in a slightly different parameter regime, the fact that the testing must be done on the product and return lines after installation at a service station presents significant challenges in devising an effective test strategy

    Psychotropic medication use in adolescents with intellectual disability living in the community

    Purpose Information on the use of psychotropic medications in adolescents with intellectual disability is scant. Such information can guide interventions to improve psychotropic medication use in this population. We investigated the prevalence of, and factors associated with, psychotropic medication use in adolescents with intellectual disability in Australia who live in the community. Methods Cross-sectional data were obtained from adolescents with intellectual disability living in the community in South East Queensland, Australia, between February 2007 and September 2010. Self-reported information on medication use was extracted from a health screening tool. Demographic and medical data were collected through parent/caregiver surveys. Medications were classified according to the Anatomical Therapeutic Chemical classification system. Psychopathology was assessed using the Developmental Behaviour Checklist Short Form. Logistic regression analysis was used to assess the association of demographic and medical characteristics with psychotropic medication use. Results There were 176 participants (median age = 16 years, range = 11–19 years; 55% male). Psychotropics were used by 20% of participants. Psychostimulants were the commonest psychotropic class, used by 9% of participants. Multipsychotropic prescribing was not common with only seven participants using more than one psychotropic agent. After adjusting for potentially confounding variables, use of psychotropic medications was significantly associated with male gender (adjusted odds ratio = 3.6; 95% confidence interval = 1.3–9.5) and having major behaviour problems (3.1; 1.1–8.9). Conclusions Adolescents with intellectual disability use a wide range of psychotropic medications. Being male and having major behaviour problems are associated with the use of psychotropic medications. Research examining the rationale for psychotropic prescribing in this population is needed.     Investigating causality in the association between 25(OH)D and schizophrenia

    Vitamin D deficiency is associated with increased risk of schizophrenia. However, it is not known whether this association is causal or what the direction of causality is. We performed two sample bidirectional Mendelian randomization analysis using single nucleotide polymorphisms (SNPs) robustly associated with serum 25(OH)D to investigate the causal effect of 25(OH)D on risk of schizophrenia, and SNPs robustly associated with schizophrenia to investigate the causal effect of schizophrenia on 25(OH)D. We used summary data from genome-wide association studies and meta-analyses of schizophrenia and 25(OH)D to obtain betas and standard errors for the SNP-exposure and SNP-outcome associations. These were combined using inverse variance weighted fixed effects meta-analyses. In 34,241 schizophrenia cases and 45,604 controls, there was no clear evidence for a causal effect of 25(OH)D on schizophrenia risk. The odds ratio for schizophrenia per 10% increase in 25(OH)D conferred by the four 25(OH)D increasing SNPs was 0.992 (95% CI: 0.969 to 1.015). In up to 16,125 individuals with measured serum 25(OH)D, there was no clear evidence that genetic risk for schizophrenia causally lowers serum 25(OH)D. These findings suggest that associations between schizophrenia and serum 25(OH)D may not be causal. Therefore, vitamin D supplementation may not prevent schizophrenia.</p

    In vitro bacterial vaginosis biofilm community manipulation using endolysin therapy

    Bacterial vaginosis (BV) affects approximately 26% of women of childbearing age globally, presenting with 3–5 times increased risk of miscarriage and two-fold risk of pre-term birth. Antibiotics (metronidazole and clindamycin) are typically employed to treat BV; however the success rate is low due to the formation of recalcitrant polymicrobial biofilms. As a novel therapeutic, promising results have been obtained in vitro using Gardnerella endolysins, although to date their efficacy has only been demonstrated against simple biofilm models. In this study, a four-species biofilm was developed consisting of Gardnerella vaginalis, Fannyhessea vaginae, Prevotella bivia and Mobiluncus curtisii. Biofilms were grown in NYC III broth and treated using antibiotics and an anti-Gardnerella endolysin (CCB7.1) for 24 h. Biofilm composition, viability and structure were assessed using colony counts, live/dead qPCR and scanning electron microscopy. All species colonised biofilms to varying degrees, with G. vaginalis being the most abundant. Biofilm composition remained largely unchanged when challenged with escalated concentrations of conventional antibiotics. A Gardnerella-targeted endolysin candidate (CCB7.1) showed efficacy against several Gardnerella species planktonically, and significantly reduced viable G. vaginalis within polymicrobial biofilms at 1 to 4X pMIC (p &lt; 0.05 vs. vehicle control). Collectively, this study highlights the resilience of biofilm-embedded pathogens against the currently used antibiotics and provides a polymicrobial model that allows for more effective pre-clinical screening of BV therapies.
