Te Aka Roa o Te Oranga, the far reaching vines of wellness: The development of a framework to evaluate alcohol and drug treatment for Māori
The impact of alcohol and other drug
problems for Māori is well documented.
Substance use has been implicated in a
range of physical and mental health
problems, and a variety of negative social
statistics such as high rates of imprisonment. To date there has been
little systematic documentation of treatment
practices, and limited operationalisation of
Māori health frameworks. The evaluation
of the outcomes of alcohol and other drug
treatments is an area in which there is a
paucity of documentation, in terms of
methods and frameworks for evaluation,
and actual data. Te Aka Roa O Te Oranga (TAROTO)
was developed from a range of projects
undertaken by the National Addiction
Centre. The TARATO evaluation framework
embraces a holistic perspective: developed
to examine the interaction between the
client, whānau, practitioner, and
service/organisation. The aim of the
framework is to clarify the complex
relationships and interactions between
stakeholders within the context of
treatment. It will also help to elucidate the
strengths and weaknesses of individual
services. Within this framework, a range of
indicators and outcomes of “successful
treatment” will be explored. The current project is the first phase of
a broader project that will make a
significant contribution to improvements in
Māori health via further developing
effective treatments of alcohol and other
drug related problems
Identification of early gene expression changes in primary cultured neurons treated with topoisomerase I poisons.
Topoisomerase 1 (TOP1) poisons like camptothecin (CPT) are currently used in cancer chemotherapy but these compounds can have damaging, off-target effects on neurons leading to cognitive, sensory and motor deficits. To understand the molecular basis for the enhanced sensitivity of neurons to CPT, we examined the effects of compounds that inhibit TOP1-CPT, actinomycin D (ActD) and β-lapachone (β-Lap)-on primary cultured rat motor (MN) and cortical (CN) neurons as well as fibroblasts. Neuronal cells expressed higher levels of Top1 mRNA than fibroblasts but transcript levels are reduced in all cell types after treatment with CPT. Microarray analysis was performed to identify differentially regulated transcripts in MNs in response to a brief exposure to CPT. Pathway analysis of the differentially expressed transcripts revealed activation of ERK and JNK signaling cascades in CPT-treated MNs. Immediate-early genes like Fos, Egr-1 and Gadd45b were upregulated in CPT-treated MNs. Fos mRNA levels were elevated in all cell types treated with CPT; Egr-1, Gadd45b and Dyrk3 transcript levels, however, increased in CPT-treated MNs and CNs but decreased in CPT-treated fibroblasts. These transcripts may represent new targets for the development of therapeutic agents that mitigate the off-target effects of chemotherapy on the nervous system
The crustose red algal genus Peyssonnelia (Peyssonneliales, Rhodophyta) in the Azores : from five to one species
The family Peyssonneliaceae comprises a worldwide group of non-calcified to calcified, crust-forming red algae of great ecological significance. Of the genera currently recognized in the family, Peyssonnelia has been widely considered to contain the largest number of species, with five members reported for the Azores. Using rbcL as a molecular marker, we here report on the taxonomic identity of recent collections of Peyssonneliaceae from the Azorean islands of São Miguel, Graciosa and Pico, and compare those specimens in a worldwide context. Only a single Peyssonnelia species, P. squamaria, is confirmed for the Azorean archipelago, with three different haplotypes. Although the populations in the Azores are genetically different from those occurring in the Mediterranean, this separation appears to be relatively recent

VCU is a large, public, urban research university situated in the middle of a capital city. Its faculty, staff, student body, alumni, and the surrounding community are remarkably diverse as are the academic offerings. It is, and should be viewed as, the intellectual and cultural engine of the region. Our project proposes the creation of a large-scale, high-profile speaker series designed to highlight emerging trends and provide students, faculty, staff, alumni and the Richmond community with a forum for conversation. The speaker series will cover topics that are critically engaging, have national relevance, and introduce ideas that propel the next generation of leaders. In addition to a large speaking engagement, the speaker series will also incorporate other activities to cultivate interactions and build relationships such as classroom lectures, book signings, and a dinner through the development office. The speaker series will host at least one speaker annually, with the addition of a second speaker as the event builds momentum. At least one of the lectures will occur at the beginning of the traditional academic semester, allowing for the greatest opportunity for participation across VCU and Richmond. Internal support from VCU students, faculty, staff, and colleges will ensure that the project is connected to the mission, vision, goals, and pursuits of VCU. A speaker series committee will help sustain and coordinate efforts across the university and community. Committee members will include stakeholders that require buy-in and cooperation for activities that complement the speaker series (e.g., other lectures, panel discussions, classroom activities). A survey will be used to gain insights into topics and speakers of interest. The committee will review the survey responses in order to make informed decisions during the planning process. The ongoing presence of hosting influential speakers will allow VCU to emerge into the national spotlight as thought-leaders. This speaker series will serve many purposes. First, the series will serve to inspire VCU students, faculty, staff, and the Richmond. Through frank and open conversations attendees will be exposed to new concepts and ideas. Second, the series will unite the diverse groups that make up VCU and the Richmond community. The lecture series will expose attendees to new ideas and open doors for possible opportunities for collaboration through classroom and community engagement activities related to the topics discussed. Third, the series will serve as a cultural conduit, solidly connecting the VCU and Richmond communities around engaging ideas of importance. Opening a new market-place of ideas will ensure that the students of VCU interact with new information in exciting and transformative ways
Factors influencing participation in a vascular disease prevention lifestyle program among participants in a cluster randomized trial
BackgroundPrevious research suggests that lifestyle intervention for the prevention of diabetes and cardiovascular disease (CVD) are effective, however little is known about factors affecting participation in such programs. This study aims to explore factors influencing levels of participation in a lifestyle modification program conducted as part of a cluster randomized controlled trial of CVD prevention in primary care.MethodsThis concurrent mixed methods study used data from the intervention arm of a cluster RCT which recruited 30 practices through two rural and three urban primary care organizations. Practices were randomly allocated to intervention (n = 16) and control (n = 14) groups. In each practice up to 160 eligible patients aged between 40 and 64 years old, were invited to participate. Intervention practice staff were trained in lifestyle assessment and counseling and referred high risk patients to a lifestyle modification program (LMP) consisting of two individual and six group sessions over a nine month period. Data included a patient survey, clinical audit, practice survey on capacity for preventive care, referral and attendance records at the LMP and qualitative interviews with Intervention Officers facilitating the LMP. Multi-level logistic regression modelling was used to examine independent predictors of attendance at the LMP, supplemented with qualitative data from interviews with Intervention Officers facilitating the program.ResultsA total of 197 individuals were referred to the LMP (63% of those eligible). Over a third of patients (36.5%) referred to the LMP did not attend any sessions, with 59.4% attending at least half of the planned sessions. The only independent predictors of attendance at the program were employment status - not working (OR: 2.39 95% CI 1.15-4.94) and having high psychological distress (OR: 2.17 95% CI: 1.10-4.30). Qualitative data revealed that physical access to the program was a barrier, while GP/practice endorsement of the program and flexibility in program delivery facilitated attendance.ConclusionBarriers to attendance at a LMP for CVD prevention related mainly to external factors including work commitments and poor physical access to the programs rather than an individuals’ health risk profile or readiness to change. Improving physical access and offering flexibility in program delivery may enhance future attendance. Finally, associations between psychological distress and attendance rates warrant further investigation
Mobility and Variation in Chalcolithic North Gujarat, India (Ca 3600 – 1800 Bc)
Nine relatively obscure sites in the northern plain of Gujarat, India: Loteshwar, Santhli, Datrana, Nagwada, Langhnaj, Zekhada, Ratanpura and Kanewal; demonstrate a broad range of material culture traditions present in this region throughout the fourth through second millennia BC. This diversity results from the numerous economic strategies employed by the inhabitants of this region, the most important of which is mobility. Most of the sites reviewed in this work are the remains of temporary occupations, which are usually ascribed to pastoral nomads. Although pastoralism was an important subsistence strategy, a closer examination of the material culture and features at these sites shows there was a spectrum of approaches to mobility, which were related to different economic strategies. This work will show that despite many similarities, these sites do not represent a homogenous set of pastoralist camps. Instead, they document manifold activities, reflected through the uses of material culture and space
Transcriptome profiling of spinal muscular atrophy motor neurons derived from mouse embryonic stem cells.
Proximal spinal muscular atrophy (SMA) is an early onset, autosomal recessive motor neuron disease caused by loss of or mutation in SMN1 (survival motor neuron 1). Despite understanding the genetic basis underlying this disease, it is still not known why motor neurons (MNs) are selectively affected by the loss of the ubiquitously expressed SMN protein. Using a mouse embryonic stem cell (mESC) model for severe SMA, the RNA transcript profiles (transcriptomes) between control and severe SMA (SMN2+/+;mSmn-/-) mESC-derived MNs were compared in this study using massively parallel RNA sequencing (RNA-Seq). The MN differentiation efficiencies between control and severe SMA mESCs were similar. RNA-Seq analysis identified 3,094 upregulated and 6,964 downregulated transcripts in SMA mESC-derived MNs when compared against control cells. Pathway and network analysis of the differentially expressed RNA transcripts showed that pluripotency and cell proliferation transcripts were significantly increased in SMA MNs while transcripts related to neuronal development and activity were reduced. The differential expression of selected transcripts such as Crabp1, Crabp2 and Nkx2.2 was validated in a second mESC model for SMA as well as in the spinal cords of low copy SMN2 severe SMA mice. Furthermore, the levels of these selected transcripts were restored in high copy SMN2 rescue mouse spinal cords when compared against low copy SMN2 severe SMA mice. These findings suggest that SMN deficiency affects processes critical for normal development and maintenance of MNs
