    Common Knowledge: The Epistemology of American Realism

    My dissertation, Common Knowledge: The Epistemology of American Realism, focuses on realist fiction (primarily the novel) at the end of the nineteenth century. Its motivating claim is that the central descriptive and thematic imperative of realism--to depict life as it is rather than in some idealized form--emerged in response to crises in the status of knowledge that resulted from an attempt by writers and readers to come to a common understanding of the relationship between private experience and an increasingly fragmented social world. While William Dean Howells\u27s definition of realism as a form of writing that displays fidelity to experience and probability of motive assumes a correspondence between writing and the real, my dissertation argues that realism\u27s primary aesthetic achievement was its response to a pervasive sense of epistemological uncertainty. Accordingly, Common Knowledge engages the tensions embodied in interpenetrating depictions of social conflict and shared knowledge. On one hand, much recent scholarship has been devoted to demonstrating realism\u27s commitment to documenting the intensified class conflict characteristic of the last decade of the nineteenth century. On the other hand, much scholarship has also been dedicated to portraying realism as an articulation of bourgeois gentility that remained largely ignorant of the stakes of such conflicts. In studies of the novels of Howells, Henry James, Harold Frederic, and Charles Chesnutt, I attempt to synthesize those two interpretations of American realism, preferring to read oscillations between social concern and reified class privilege as indications of a fundamental ambivalence about the reliability of social knowledge. Common Knowledge entwines readings of fiction with elaborations on the critical, technological, and aesthetic discourses of epistemological uncertainty that emerge from them, documenting how recognitions of socio-economic, racial, and ontological difference both rely on and throw into question the possibility of a shared knowledge of the world

    Turnstile File Transfer: A Unidirectional System for Medium-Security Isolated Clusters

    Data transfer between isolated clusters is imperative for cybersecurity education, research, and testing. Such techniques facilitate hands-on cybersecurity learning in isolated clusters, allow cybersecurity students to practice with various hacking tools, and develop professional cybersecurity technical skills. Educators often use these remote learning environments for research as well. Researchers and students use these isolated environments to test sophisticated hardware, software, and procedures using full-fledged operating systems, networks, and applications. Virus and malware researchers may wish to release suspected malicious software in a controlled environment to observe their behavior better or gain the information needed to assist their reverse engineering processes. The isolation prevents harm to networked systems. However, there are times when the data is required to move in such quantities or speeds that it makes downloading onto an intermediate device untenable. This study proposes a novel turnstile model, a mechanism for one-way file transfer from one enterprise system to another without allowing data leakage. This system protects data integrity and security by connecting the isolated environment to the external network via a locked-down interconnection. Using medium-security isolated clusters, the researchers successfully developed a unidirectional file transfer system that acts as a one-way “turnstile” for secure file transfer between systems not connected to the internet or other external networks. The Turnstile system (source code available at github.com/monnin/turnstile) provides unidirectional file transfer between two computer systems. The solution enabled data to be transferred from a source system to a destination system without allowing any data to be transferred back in the opposite direction. The researchers found an automated process of transferring external files to isolated clusters optimized the transfer speed of external files to isolated clusters using Linux distributions and commands

    Mutations in the store-operated Ca²⁺ entry pore protein ORAI1 have been reported to cause myopathies in human patients but the mechanism involved is not known. Cardiomyocytes express ORAI1 but its role in heart function is also unknown. Using reverse genetics in zebrafish, we demonstrated that inactivation of the highly conserved zebrafish orthologue of ORAI1 resulted in severe heart failure, reduced ventricular systolic function, bradycardia and skeletal muscle weakness. Electron microscopy of Orai1-deficient myocytes revealed progressive skeletal muscle instability with loss of myofiber integrity and ultrastructural abnormalities of the z-disc in both skeletal and cardiac muscle. Isolated Orai1-deficient cardiomyocytes showed loss of the calcineurin-associated protein calsarcin from the z-discs. Furthermore, we found mechanosignal transduction was affected in Orai1-depleted hearts, indicating an essential role for ORAI1 in establishing the cardiac signaling transduction machinery at the z-disc. Our findings identify ORAI1 as an important regulator of cardiac and skeletal muscle function and provide evidence linking ORAI1-mediated calcium signaling to sarcomere integrity and cardiomyocyte function

    Effects of a ketogenic diet during pregnancy on embryonic growth in the mouse

    BACKGROUND: The increasing use of the ketogenic diet (KD), particularly by women of child-bearing age, raises a question about its suitability during gestation. To date, no studies have thoroughly investigated the direct implications of a gestational ketogenic diet on embryonic development. METHODS: To fill this knowledge gap we imaged CD-1 mouse embryos whose mothers were fed either a Standard Diet (SD) or a KD 30 days prior to, as well as during gestation. Images were collected at embryonic days (E) 13.5 using Optical Projection Tomography (OPT) and at E17.5 using Magnetic Resonance Imaging (MRI). RESULTS: An anatomical comparison of the SD and KD embryos revealed that at E13.5 the average KD embryo was volumetrically larger, possessed a relatively larger heart but smaller brain, and had a smaller pharynx, cervical spinal cord, hypothalamus, midbrain, and pons, compared with the average SD embryo. At E17.5 the KD embryo was found to be volumetrically smaller with a relatively smaller heart and thymus, but with enlarged cervical spine, thalamus, midbrain and pons. CONCLUSION: A ketogenic diet during gestation results in alterations in embryonic organ growth. Such alterations may be associated with organ dysfunction and potentially behavioral changes in postnatal life
