    Design of Super Twisting Integral Sliding Mode Control for Industrial Robot Manipulator

    In the present work, integral sliding mode based continuous control algorithm is extended to multi input multi output system. The typical integral sliding mode control (ISMC) contains nominal control with discontinuous feedback control due to which overall control becomes discontinuous in nature. The proposed controller is a fusion of two continuous terms and one of which is able to handle, estimate and reject the disturbance successfully. A proposed robust ISMC technique is applied for industrial robot manipulators which utilizes interactive manipulation activity. Here, robust position tracking control obtained via ISMC principle for two link IRM scheme influenced by parametric uncertainties and external disturbances. The proposed ISMC design replaces the discontinuous part by continuous control, which super twisting control is able to handle the disturbance rejection completely. The effectiveness of the proposed control technique is tested under uncertain conditions and comparison study with other controllers has been done. The simulation result shows that the tracking error is effectively minimized by the proposed technique in presence of uncertain conditions

    Regulation of virulence factors by quorum sensing in Vibrio harveyi.

    Vibrio harveyi is an important aquatic pathogen that produces several virulence factors. In this study, the effect of quorum sensing, bacterial cell-to-cell communication, on the production of the virulence factors caseinase, gelatinase, lipase, hemolysin, and phospholipase, was investigated. The activity of virulence factors was studied through enzymatic plate assays using V. harveyi wild type and mutants with constitutively maximal or minimal quorum sensing activity. The results showed that quorum sensing negatively regulates phospholipase activity as higher activity was observed in mutants with minimal quorum sensing activity than in the mutant with maximal quorum sensing activity. Reverse transcriptase real-time PCR with specific primers revealed that the expression level of three phospholipase genes was 2-fold lower in the mutant with minimal quorum sensing activity than in the mutant with maximal quorum sensing activity. As far as we know, this is the first report of quorum sensing regulation of phospholipase. Finally, caseinase and gelatinase activity were positively regulated by quorum sensing, which is consistent with previous reports, and lipase and hemolysin activity were found to be independent of quorum sensing. Hence, the regulation is different for different virulence factors, with some being either positively or negatively regulated, and others being independent of quorum sensing. This might reflect the need to produce the different virulence factors at different stages during infection

It has been observed that there is a considerable likelihood that dumping light weight solid waste items may pollute public areas where the public congregates momentarily. There is no ideal method for keeping an eye on and maintaining these locations. It is possible to create a self-controlled robot application to solve these issues. Here machine can identify garbage and separate it into several categories, such as metallic and non-metallic waste.Implemented system uses IR sensors to detect garbage, metal proximity sensors to distinguish between metallic and non-metallic junk, and moisture sensors to further divide garbage into wet and dry waste. Here robot is controlled by an ESP32 microcontroller

    Industry 4.0 the proclaimed fourth industrial revolution is unfolding at the moment. It is characterized by interconnectedness and vast amounts of available information. Industrial production has evolved enormously over the last centuries due to modern instruments. Hence issue of the instrument failure is very paramount in any industry. Even if one machine fails it halts the whole production. Overall, it may cost us with more man-hours, project delay, process latency and all this sums up as a huge loss. The life of the instruments should be taken care by continuously monitoring its health. Any faulty or unnatural disturbance in usage of the instrument may lead to its failure. Every instrument needs proper maintenance, even with the slight negligence towards the anomaly it may lead to instrument failure. In, predictive maintenance historic data is utilized and analyzed with the help of advance analytics and modelling techniques using Machine learning, moreover we can predict failures and can schedule the maintenance beforehand and predict failure in advance. With the help of relevant sensor dataset, we can estimate the remaining runtime of the instruments. This maintenance approach helps to lower the costs which are incurred due to system shut downs. It also ease the scheduling and maintenance activities.In this work, three different industrial case studies are considered like shell and tube type heat exchanger, plate type heat exchanger, and semiconductor manufacturing process.Here the predictive maintenance is carried out for heat exchanger by utilizing the concept of multi linear regression and time series analysis. For the semiconductor manufacturing dataset, support vector machine algorithm is implemented to find out the good and bad quality of semiconductor production slots

    Bioanalytical Method Development and Validation Study of Neuroprotective Extract of Kashmiri Saffron Using Ultra-Fast Liquid Chromatography-Tandem Mass Spectrometry (UFLC-MS/MS): In Vivo Pharmacokinetics of Apocarotenoids and Carotenoids

    Kashmir saffron (Crocus sativus L.), also known as Indian saffron, is an important Asian medicinal plant with protective therapeutic applications in brain health. The main bioactive in Kashmir or Indian Saffron (KCS) and its extract (CSE) are apocarotenoids picrocrocin (PIC) and safranal (SAF) with carotenoids, crocetin esters (crocins), and crocetins. The ultra-fast liquid chromatography(UFLC)- photodiode array standardization confirmed the presence of biomarkers PIC, trans-4-GG-crocin (T4C), trans-3-Gg-crocin (T3C), cis-4-GG-crocin (C4C), trans-2-gg-crocin (T2C), trans-crocetin (TCT), and SAF in CSE. This study’s objectives were to develop and validate a sensitive and rapid UFLC-tandem mass spectrometry method for PIC and SAF along T4C and TCT in rat plasma with internal standards (IS). The calibration curves were linear (R2 > 0.990), with the lower limit of quantification (LLOQ) as 10 ng/mL. The UFLC-MS/MS assay-based precision (RSD, <15%) and accuracy (RE, −11.03–9.96) on analytical quality control (QC) levels were well within the acceptance criteria with excellent recoveries (91.18–106.86%) in plasma samples. The method was applied to investigate the in vivo pharmacokinetic parameters after oral administration of 40 mg/kg CSE in the rats (n = 6). The active metabolite TCT and T4C, PIC, SAF were quantified for the first time with T3C, C4C, T2C by this validated bioanalytical method, which will be useful for preclinical/clinical trials of CSE as a potential neuroprotective dietary supplement

    Development and validation of a multigene variant profiling assay to guide targeted and immuno therapy selection in solid tumors.

    We present data on analytical validation of the multigene variant profiling assay (CellDx) to provide actionable indications for selection of targeted and immune checkpoint inhibitor (ICI) therapy in solid tumors. CellDx includes Next Generation Sequencing (NGS) profiling of gene variants in a targeted 452-gene panel as well as status of total Tumor Mutation Burden (TMB), Microsatellite instability (MSI), Mismatch Repair (MMR) and Programmed Cell Death-Ligand 1 (PD-L1) respectively. Validation parameters included accuracy, sensitivity, specificity and reproducibility for detection of Single Nucleotide Alterations (SNAs), Copy Number Alterations (CNAs), Insertions and Deletions (Indels), Gene fusions, MSI and PDL1. Cumulative analytical sensitivity and specificity of the assay were 99.03 (95% CI: 96.54-99.88) and 99.23% (95% CI: 98.54% - 99.65%) respectively with 99.20% overall Accuracy (95% CI: 98.57% - 99.60%) and 99.7% Precision based on evaluation of 116 reference samples. The clinical performance of CellDx was evaluated in a subsequent analysis of 299 clinical samples where 861 unique mutations were detected of which 791 were oncogenic and 47 were actionable. Indications in MMR, MSI and TMB for selection of ICI therapies were also detected in the clinical samples. The high specificity, sensitivity, accuracy and reproducibility of the CellDx assay is suitable for clinical application for guiding selection of targeted and immunotherapy agents in patients with solid organ tumors
