    Genetic and biochemical identification of a novel single-stranded DNA-binding complex in Haloferax volcanii

    Single-stranded DNA (ssDNA)-binding proteins play an essential role in DNA replication and repair. They use oligonucleotide/oligosaccharide-binding (OB)-folds, a five-stranded Ī²-sheet coiled into a closed barrel, to bind to ssDNA thereby protecting and stabilizing the DNA. In eukaryotes the ssDNA-binding protein (SSB) is known as replication protein A (RPA) and consists of three distinct subunits that function as a heterotrimer. The bacterial homolog is termed SSB and functions as a homotetramer. In the archaeon Haloferax volcanii there are three genes encoding homologs of RPA. Two of the rpa genes (rpa1 and rpa3) exist in operons with a novel gene specific to Euryarchaeota; this gene encodes a protein that we have termed RPA-associated protein (rpap). The rpap genes encode proteins belonging to COG3390 group and feature OB-folds, suggesting that they might cooperate with RPA in binding to ssDNA. Our genetic analysis showed that rpa1 and rpa3 deletion mutants have differing phenotypes; only Ī”rpa3 strains are hypersensitive to DNA damaging agents. Deletion of the rpa3-associated gene rpap3 led to similar levels of DNA damage sensitivity, as did deletion of the rpa3 operon, suggesting that RPA3 and RPAP3 function in the same pathway. Protein pull-downs involving recombinant hexahistidine-tagged RPAs showed that RPA3 co-purifies with RPAP3, and RPA1 co-purifies with RPAP1. This indicates that the RPAs interact only with their respective associated proteins; this was corroborated by the inability to construct rpa1 rpap3 and rpa3 rpap1 double mutants. This is the first report investigating the individual function of the archaeal COG3390 RPA-associated proteins (RPAPs). We have shown genetically and biochemically that the RPAPs interact with their respective RPAs, and have uncovered a novel single-stranded DNA-binding complex that is unique to Euryarchaeota

    Spatial Dynamics of Pandemic Influenza in a Massive Artificial Society

    EpiSimS is a massive simulation of the movements, activities, and social interactions of individuals in realistic synthetic populations, and of the dynamics of contagious disease spread on the resulting social contact network. This paper describes the assumptions and methodology in the EpiSimS model. It also describes and presents a simulation of the spatial dynamics of pandemic influenza in an artificial society constructed to match the demographics of southern California. As an example of the utility of the massive simulation approach, we demonstrate a strong correlation between local demographic characteristics and pandemic severity, which gives rise to previously unanticipated spatial pandemic hotspots. In particular, the average household size in a census tract is strongly correlated with the clinical attack rate computed by the simulation. Public heath agencies with responsibility for communities having relatively high population per household should expect to be more severely hit by a pandemic.Agent Based Modeling, Computer Simulation, Epidemic Simulation, Public Health Policy

    Preparing Faculty For Overseas Assignments

    With increased globalization, more colleges and universities are establishing faculty exchanges, study abroad programs, collaborative research efforts, and technical assistance programs with counterparts overseas resulting in enhanced faculty development, increased learning for students, revenue generation and prestige for the partnering institutions. If faculty are inadequately prepared and supported for overseas assignments valuable time and resources are wasted, student learning is impacted, and the image of the institution and faculty member may suffer. The purpose of this research was to determine how institutions can better prepare faculty for overseas assignments. To study this problem, faculty with international teaching experience and overseas country coordinators were surveyed and a literature review was conducted. Data were compiled to identify actions which can be taken to ensure greater success in overseas assignments. The researchers will share results exploring why educational institutions are expanding operations overseas, the negative impact of failing to adequately prepare and support faculty for overseas assignments, and actions which can be taken to ensure faculty are adequately prepared and supported for their overseas assignment.https://fuse.franklin.edu/ss2014/1021/thumbnail.jp

    Biobehavioral Indices of Emotion Regulation Relate to School Attitudes, Motivation, and Behavior Problems in a Low-Income Preschool Sample

    Effective emotion regulation may promote resilience and preschool classroom adjustment by supporting adaptive peer interactions and engagement in learning activities. We investigated how hypothalamus-pituitary-adrenal axis (HPA) regulation, cardiac reactivity, and classroom emotion displays related to adjustment among low-income preschoolers attending Head Start. A total of 62 four-year-olds completed a laboratory session including a baseline soothing video; emotion-eliciting slides/video clips, and recovery. Salivary cortisol, heart rate, and vagal tone were measured throughout. Independent coders used handheld computers to observe classroom emotion expression/regulation. Teachers rated child motivation, persistence/attention, learning attitudes, and internalizing/externalizing symptoms.
