Molecular cloning of doublesex genes of four cladocera (water flea) species
BACKGROUND: The gene doublesex (dsx) is known as a key factor regulating genetic sex determination in many organisms. We previously identified two dsx genes (DapmaDsx1 and DapmaDsx2) from a freshwater branchiopod crustacean, Daphnia magna, which are expressed in males but not in females. D. magna produces males by parthenogenesis in response to environmental cues (environmental sex determination) and we showed that DapmaDsx1 expression during embryonic stages is responsible for the male trait development. The D. magna dsx genes are thought to have arisen by a cladoceran-specific duplication; therefore, to investigate evolutionary conservation of sex specific expression of dsx genes and to further assess their functions in the environmental sex determination, we searched for dsx homologs in four closely related cladoceran species. RESULTS: We identified homologs of both dsx genes from, D. pulex, D. galeata, and Ceriodaphnia dubia, yet only a single dsx gene was found from Moina macrocopa. The deduced amino acid sequences of all 9 dsx homologs contained the DM and oligomerization domains, which are characteristic for all arthropod DSX family members. Molecular phylogenetic analysis suggested that the dsx gene duplication likely occurred prior to the divergence of these cladoceran species, because that of the giant tiger prawn Penaeus monodon is rooted ancestrally to both DSX1 and DSX2 of cladocerans. Therefore, this result also suggested that M. macrocopa lost dsx2 gene secondarily. Furthermore, all dsx genes identified in this study showed male-biased expression levels, yet only half of the putative 5’ upstream regulatory elements are preserved in D. magna and D. pulex. CONCLUSIONS: The all dsx genes of five cladoceran species examined had similar amino acid structure containing highly conserved DM and oligomerization domains, and exhibited sexually dimorphic expression patterns, suggesting that these genes may have similar functions for environmental sex determination in cladocerans

Introduction: In early-stage oral tongue squamous cell carcinoma (OTSCC), elective neck dissection (END) is recommended when occult lymph node metastasis is suspected; however, there is no unanimous consensus on the risks and benefits of END in such cases. The management of clinically node-negative (cN0) OTSCC remains controversial. This study, therefore, aimed to evaluate the efficacy of END and its impact on the quality of life (QoL) of patients with cN0 OTSCC.
Methods and analysis: This is a prospective, multicentre, nonrandomised observational study. The choice of whether to perform END at the same time as resection of the primary tumour is based on institutional policy and patient preference. The primary endpoint of this study is 3-year overall survival. The secondary endpoint are 3-year disease-specific survival, 3-year relapse-free survival and the impact on patient QoL. Propensity score-matching analysis will be performed to reduce selection bias.
Ethics and dissemination: This study was approved by the Clinical Research Review Board of the Nagasaki University. The protocol of this study was registered at the University Hospital Medical Information Network Clinical Trials Registry. The datasets generated during the current study will be available from the corresponding author on reasonable request. The results will be disseminated internationally, through scientific and professional conferences and in peer-reviewed medical journals

看護技術教育のための模擬患者(Simulated Patient :SP)養成の実際
医学教育の客観的臨床能力試験(OSCE:Objective Structured Clinical Examination)への導入から始まった模擬患者(Standardized/Simulated Patient)参画型学習は、看護学教育では2000年頃から対人関係やコミュニケーション学習を中心に導入されてきた。医学教育では独自の養成を行っている大学が少なくないが、看護学教育においては独自の養成をしている教育機関はまだ少ない現状にある。模擬患者の導入によって、看護学生が患者への理解や看護実践の理解が進み、学内演習と臨地実習との乖離を埋めることも期待されることから、訓練された模擬患者によるシミュレーション学習には一定の効果が見込まれる。 本稿では基礎看護技術教育への模擬患者(Simulated Patient:SP)導入を視野に入れ、平成24年度に本学の公開講座として看護学部基礎看護学分野が主催した『看護学生と学ぶ模擬患者養成講座』の実際を報告する。本講座は、現役SPが講師を務め、基礎看護学教員が看護技術教育で学生に伝えたいことや教育的ビジョンなどを伝え、相互に十分なディスカッションができるように協働して進めた。受講者は3名であった。看護学生の参加は1年生3名、4年生5名で任意の参加とした。本講座は6回コースであり、修了した受講生は、その後の基礎看護技術演習へのデビューに備えて、引き続いてフォローアップ講座3回を受講した。現在は、本学で継続した活動を続けている。この養成講座の実践を通して、SPの質保障は教育側の責任であるという観点からSP養成の意義を考察する
SofTouch: Turning Soft Objects into Touch Interfaces Using Detachable Photo Sensor Modules
Part 2: DeviceInternational audienceWe propose a system that turns everyday soft objects, such as cushions, into touch interfaces in a non-intrusive manner. The belt type sensor modules developed in this study, which can be attached to the outside of soft objects, comprise several photo reflective sensors to measure the reflection intensity from an object by emitting infrared light (IR LED). When the light is irradiated to a material with the same reflection coefficient, a reflection intensity that is inversely proportional to the square of the distance can be obtained. Our method uses the sensor modules to measure the change in distance from the sensor to the surface of the soft object, and the touch position is estimated using a Support Vector Machine (SVM). To evaluate our method, we measured the accuracy when touching nine points on a cushion. Using existing everyday soft objects, it is possible to create interfaces that not only blend into the living space naturally but also match the preferences of each user
Origin of aeolian dust emitted from the Tarim Basin based on the ESR signal intensity and crystallinity index of quartz: the recycling system of fine detrital material within the basin
The Tarim Basin is the major source of aeolian dust in the northern hemisphere. Glacial activity in the mountains, transportation by rivers and homogenization by wind are believed to be responsible for dust production within the basin. However, the major source(s) and homogenization process(es) are not clear. Moreover, provenance studies on fine fractions have never been conducted. Here, we measured electron spin resonance (ESR) signal intensity and the crystallinity index of quartz in fine ( 64 mu m) fractions of river sediments, dry lake sediments and mountain loess to examine the process(es) that produce aeolian dust. The result suggests that the coarse fraction of the river sediment was derived from the bedrock in the drainage area. The ESR signal intensity and crystallinity index of the fine fraction of river sediments from the Tian Shan Mountains and mountainous rivers in the westernmost Kunlun and Pamir mountains are also similar to the coarse fraction, suggesting the same sources. However, the ESR signal intensity and crystallinity index of the fine fraction of river sediments from the Kunlun Mountains are different from the coarse fraction and converge towards values close to the average for the fine fraction of river sediments and mountain loess. Convergence of the ESR and crystallinity index values for the fine fraction of river sediments from the Kunlun Mountains can be explained by contamination of the river sediments by aeolian dust. The convergent values resulted from the homogenization of fine detrital material by repeated recycling within the basin

Cancer stem cells have the capability of self-renewal and multipotency and are, therefore, associated with tumor heterogeneity, resistance to chemoradiation therapy, and metastasis. The hypothesis that multinucleated giant cells, which often emerge following chemo- and/or radiotherapy, serve as cancer stem cells has not been fully evaluated. Although a previous study demonstrated that these cells functioned as stem cells, only low levels of Yamanaka factors were expressed, contrasting with the high expression seen from their gestated first-generation mononuclear cells. Herein, we report a case of a plasmablastic neoplasm with multinucleated giant cells that were analyzed for stemness to test the above hypothesis. The patient was a male in his 80s who had a plasmablastic neoplasm that was not easily distinguishable as plasmablastic lymphoma versus plasma cell myeloma of plasmablastic type. Lymph node biopsy showed predominant mononuclear cell proliferation with admixed multinucleated giant cells. Immunohistochemistry and in situ hybridization showed that both multinucleated and mononuclear cells had the same profile: CD138(+), light chain restriction of κ>λ, cyclin D1(+), CD68(-), EBER-ISH (+). These results suggested that both cell types were neoplastic. In accordance with the previous study, the multinucleated giant cells showed low expression of Yamanaka factors, which were highly expressed in some of the mononuclear cells. Furthermore, the multinucleated giant cells showed a much lower proliferative activity (Mib1/Ki67 index) than the mononuclear cells. Based on these results, the multinucleated giant cells were compatible with cancer stem cells. This case is expected to expand the knowledge base regarding biology of cancer stem cells.</p
