Helper Response to Experimentally Manipulated Predation Risk in the Cooperatively Breeding Cichlid Neolamprologus pulcher
Background
We manipulated predation risk in a field experiment with the cooperatively breeding cichlid Neolamprologus pulcher by releasing no predator, a medium- or a large-sized fish predator inside underwater cages enclosing two to three natural groups. We assessed whether helpers changed their helping behaviour, and whether within-group conflict changed, depending on these treatments, testing three hypotheses: βpay-to-stayβ PS, βrisk avoidanceβ RA, or (future) reproductive benefits RB. We also assessed whether helper food intake was reduced under risk, because this might reduce investments in other behaviours to save energy.
Methodology/Principal Findings
Medium and large helpers fed less under predation risk. Despite this effect helpers invested more in territory defence, but not territory maintenance, under the risk of predation (supporting PS). Experimentally covering only the breeding shelter with sand induced more helper digging under predation risk compared to the control treatment (supporting PS). Aggression towards the introduced predator did not differ between the two predator treatments and increased with group member size and group size (supporting PS and RA). Large helpers increased their help ratio (helping effort/breeder aggression received, βpunishmentβ by the dominant pair in the group) in the predation treatments compared to the control treatment, suggesting they were more willing to PS. Medium helpers did not show such effects. Large helpers also showed a higher submission ratio (submission/ breeder aggression received) in all treatments, compared to the medium helpers (supporting PS).
Conclusions/Significance
We conclude that predation risk reduces helper food intake, but despite this effect, helpers were more willing to support the breeders, supporting PS. Effects of breeder punishment suggests that PS might be more important for large compared to the medium helpers. Evidence for RA was also detected. Finally, the results were inconsistent with RB

We report here that they are of at least six distinct types, we describe how each forms, and, looking forward, encourage the targeting of tumour vessel subsets that have lost their vascular endothelial growth factor-A (VEGF-A) dependency and so are likely unresponsive to anti-VEGF-A therapies
Microsatellite Support for Active Inbreeding in a Cichlid Fish
In wild animal populations, the degree of inbreeding differs between species and within species between populations. Because mating with kin often results in inbreeding depression, observed inbreeding is usually regarded to be caused by limited outbreeding opportunities due to demographic factors like small population size or population substructuring. However, theory predicts inclusive benefits from mating with kin, and thus part of the observed variation in inbreeding might be due to active inbreeding preferences. Although some recent studies indeed report kin mating preferences, the evidence is still highly ambiguous. Here, we investigate inbreeding in a natural population of the West African cichlid fish Pelvicachromis taeniatus which showed clear kin mating preferences in standardized laboratory experiments but no inbreeding depression. The presented microsatellite analysis reveals that the natural population has, in comparison to two reference populations, a reduced allelic diversity (Aβ=β3) resulting in a low heterozygosity (Hoβ=β0.167) pointing to a highly inbred population. Furthermore, we found a significant heterozygote deficit not only at population (Fisβ=β0.116) but also at subpopulation level (Fisβ=β0.081) suggesting that inbreeding is not only a by-product of population substructuring but possibly a consequence of behavioral kin preferences
Immediate, but Not Delayed, Microsurgical Skull Reconstruction Exacerbates Brain Damage in Experimental Traumatic Brain Injury Model
Moderate to severe traumatic brain injury (TBI) often results in malformations to the skull. Aesthetic surgical maneuvers may offer normalized skull structure, but inconsistent surgical closure of the skull area accompanies TBI. We examined whether wound closure by replacement of skull flap and bone wax would allow aesthetic reconstruction of the TBI-induced skull damage without causing any detrimental effects to the cortical tissue. Adult male Sprague-Dawley rats were subjected to TBI using the controlled cortical impact (CCI) injury model. Immediately after the TBI surgery, animals were randomly assigned to skull flap replacement with or without bone wax or no bone reconstruction, then were euthanized at five days post-TBI for pathological analyses. The skull reconstruction provided normalized gross bone architecture, but 2,3,5-triphenyltetrazolium chloride and hematoxylin and eosin staining results revealed larger cortical damage in these animals compared to those that underwent no surgical maneuver at all. Brain swelling accompanied TBI, especially the severe model, that could have relieved the intracranial pressure in those animals with no skull reconstruction. In contrast, the immediate skull reconstruction produced an upregulation of the edema marker aquaporin-4 staining, which likely prevented the therapeutic benefits of brain swelling and resulted in larger cortical infarcts. Interestingly, TBI animals introduced to a delay in skull reconstruction (i.e., 2 days post-TBI) showed significantly reduced edema and infarcts compared to those exposed to immediate skull reconstruction. That immediate, but not delayed, skull reconstruction may exacerbate TBI-induced cortical tissue damage warrants a careful consideration of aesthetic repair of the skull in TBI
Evaluating vital rate contributions to greater sage-grouse population dynamics to inform conservation
Species conservation efforts often use shortβterm studies that fail to identify the vital rates that contribute most to population growth. Although the greater sageβgrouse (Centrocercus urophasianus; sageβgrouse) is a candidate for protection under the U.S. Endangered Species Act, and is sometimes referred to as an umbrella species in the sagebrush (Artemisia spp.) biome of western North America, the failure of proposed management strategies to focus on key vital rates that may contribute most to achieving population stability remains problematic for sustainable conservation. To address this dilemma, we performed both prospective and retrospective perturbation analyses of a life cycle model based on a 12βyr study that encompassed nearly all sageβgrouse vital rates. To validate our population models, we compared estimates of annual finite population growth rates (Ξ») from our femaleβbased life cycle models to those attained from maleβbased lek counts. Postβfledging (i.e., after second year, second year, and juvenile) female survival parameters contributed most to past variation in Ξ» during our study and had the greatest potential to change Ξ» in the future, indicating these vital rates as important determinants of sageβgrouse population dynamics. In addition, annual estimates of Ξ» from femaleβbased life cycle models and maleβbased lek data were similar, providing the most rigorous evidence to date that lek counts of males can serve as a valid index of sageβgrouse population change. Our comparison of fixed and mixed statistical models for evaluating temporal variation in nest survival and initiation suggest that conservation planners use caution when evaluating shortβterm nesting studies and using associated fixedβeffect results to develop conservation objectives. In addition, our findings indicated that greater attention should be paid to those factors affecting sageβgrouse postβfledging females. Our approach demonstrates the need for more longβterm studies of species vital rates across the life cycle. Such studies should address the decoupling of sampling variation from underlying process (co)variation in vital rates, identification of how such variation drives population dynamics, and how decision makers can use this information to reβdirect conservation efforts to address the most limiting points in the life cycle for a given population.Peer reviewedNatural Resource Ecology and Managemen
The 3β² Splice Site of Influenza A Segment 7 mRNA Can Exist in Two Conformations: A Pseudoknot and a Hairpin
The 3β² splice site of influenza A segment 7 is used to produce mRNA for the M2 ion-channel protein, which is critical to the formation of viable influenza virions. Native gel analysis, enzymatic/chemical structure probing, and oligonucleotide binding studies of a 63 nt fragment, containing the 3β² splice site, key residues of an SF2/ASF splicing factor binding site, and a polypyrimidine tract, provide evidence for an equilibrium between pseudoknot and hairpin structures. This equilibrium is sensitive to multivalent cations, and can be forced towards the pseudoknot by addition of 5 mM cobalt hexammine. In the two conformations, the splice site and other functional elements exist in very different structural environments. In particular, the splice site is sequestered in the middle of a double helix in the pseudoknot conformation, while in the hairpin it resides in a two-by-two nucleotide internal loop. The results suggest that segment 7 mRNA splicing can be controlled by a conformational switch that exposes or hides the splice site
