    Axillary bud viability and dry matter production of Poa ligularis in Patagonian grasslands

    Poa ligularis Nees is one of the dominant perennial grass species in Patagonian grasslands. This is the first field study which investigated the effects of defoliation frequency on its number of viable axillary buds, and subsequent dry matter production during one growing season. Experiments were arranged in a randomized complete block design. Treatments included one defoliation height (10 cm), from one to five times a year, and undefoliated controls. Bud respiratory activity was examined usingthe tetrazolium test and the vital stain Evan's blue. At the end of the growing season, plants that have been defoliated more than three times showed significatively (p<0.05) lower number of respiratory active buds than unclipped controls. A high defoliation frequency increased bud death in stem bases. Both defoliated and undefoliated plants produced more (p<0.05) axillary buds at the periphery than at their center. Defoliation treatments increased shoot weight by 13.4% on average compared with undefoliated controls, but reduced in by 10% the average number of metabolically active axillary buds. This indicates that this reduction, however, was not high enough to constrain dry matter production.It appears that Poa ligularis could tolerate moderate grazing frequency and intensity without compromising its potential regrowth capacity. Longer-term studies of the effects of defoliation on dry matter production and bud metabolic activity are needed to understand the cumulative effects of grazing of P. ligularis in natural grasslands.Poa ligularis Nees es una de las especies de gramíneas perennes dominantes en los pastizales naturales de la Patagonia. Este es el primer estudio que investigó los efectos de la frequencia de defoliación sobre su número de yemas axilares viables, y subsiguiente producción de materia seca durante una estación de crecimiento. Los estudios se efectuaron utilizando un diseño de bloques completamente al azar. Los tratamientos incluyeron una altura de defoliación (10 cm), de una a cinco veces por año, además de controles no defoliados. La actividad respiratoria de las yemas fue examinada usando el método del tetrazolio y el colorante vital azul de Evans.Hacia el final de la estación de crecimiento, las plantas que habían sido defoliadas más de tres veces mostraron un número significativamente menor (p<0.05) de yemas con actividad respiratoria que los controles no defoliados. Una alta frecuencia de defoliación incrementó la muerte de yemas en las bases de tallos. Todas las plantas- defoliadas o no, produjeron más (p<0.05) yemas axilares en su periferia que en su centro. La defoliación incrementó 13.4% el peso de los tallos en promedio, pero redujo en 10% el número promedio de yemas axilares metabólicamente activas. Esto indica que esta reducción , sin embargo, no fue suficiente como para limitar la producción de materia seca. Parece que P. ligularis toleraría una frecuencia e intensidad moderada de pastoreo sin comprometer su capacidad potencial de rebrote.     Manoeuvring between anxiety and control: Patients&apos; experience of learning to live with diabetes: A lifeworld phenomenological study

    Abstract Research shows that people with diabetes want their lives to proceed as normally as possible, but some patients experience difficulty in reaching their desired goals with treatment. The learning process is a complex phenomenon interwoven into every facet of life. Patients and healthcare providers often have different perspectives in care which gives different expectations on what the patients need to learn and cope with. The aim of this study, therefore, is to describe the experience of learning to live with diabetes. Interviews were conducted with 12 patients afflicted with type 1 or type 2 diabetes. The interviews were then analysed with reference to the reflective lifeworld research approach. The analysis shows that when the afflicted realize that their bodies undergo changes and that blood sugar levels are not always balanced as earlier in life, they can adjust to their new conditions early. The afflicted must take responsibility for balancing their blood sugar levels and incorporating the illness into their lives. Achieving such goals necessitates knowledge. The search for knowledge and sensitivity to changes are constant requirements for people with diabetes. Learning is driven by the tension caused by the need for and dependence on safe blood sugar control, the fear of losing such control, and the fear of future complications. The most important responsibilities for these patients are aspiring to understand their bodies as lived bodies, ensuring safety and security, and acquiring the knowledge essential to making conscious choices

    Definition of the σW regulon of Bacillus subtilis in the absence of stress

    Bacteria employ extracytoplasmic function (ECF) sigma factors for their responses to environmental stresses. Despite intensive research, the molecular dissection of ECF sigma factor regulons has remained a major challenge due to overlaps in the ECF sigma factor-regulated genes and the stimuli that activate the different ECF sigma factors. Here we have employed tiling arrays to single out the ECF σW regulon of the Gram-positive bacterium Bacillus subtilis from the overlapping ECF σX, σY, and σM regulons. For this purpose, we profiled the transcriptome of a B. subtilis sigW mutant under non-stress conditions to select candidate genes that are strictly σW-regulated. Under these conditions, σW exhibits a basal level of activity. Subsequently, we verified the σW-dependency of candidate genes by comparing their transcript profiles to transcriptome data obtained with the parental B. subtilis strain 168 grown under 104 different conditions, including relevant stress conditions, such as salt shock. In addition, we investigated the transcriptomes of rasP or prsW mutant strains that lack the proteases involved in the degradation of the σW anti-sigma factor RsiW and subsequent activation of the σW-regulon. Taken together, our studies identify 89 genes as being strictly σW-regulated, including several genes for non-coding RNAs. The effects of rasP or prsW mutations on the expression of σW-dependent genes were relatively mild, which implies that σW-dependent transcription under non-stress conditions is not strictly related to RasP and PrsW. Lastly, we show that the pleiotropic phenotype of rasP mutant cells, which have defects in competence development, protein secretion and membrane protein production, is not mirrored in the transcript profile of these cells. This implies that RasP is not only important for transcriptional regulation via σW, but that this membrane protease also exerts other important post-transcriptional regulatory functions

    Protection of early phase hepatic ischemia-reperfusion injury by cholinergic agonists

    BACKGROUND: Cytokine production is critical in ischemia/reperfusion (IR) injury. Acetylcholine binds to macrophages and inhibits cytokine synthesis, through the cholinergic anti-inflammatory pathway. This study examined the role of the cholinergic pathway in cytokine production and hepatic IR- injury. METHODS: Adult male mice underwent 90-min of partial liver ischemia followed by reperfusion. The AChR agonists (1,1-dimethyl-4-phenyl-L-pioperazinium-iodide [DMPP], and nicotine) or saline-vehicle were administered i.p. before ischemia. Plasma cytokine tumor necrosis factor (TNF)-α, macrophage inflammatory protein-2, and Interleukin-6 were measured. Liver injury was assessed by plasma alanine transaminase (ALT) and liver histopathology. RESULTS: A reperfusion time-dependent hepatocellular injury occurred as was indicated by increased plasma-ALT and histopathology. The injury was associated with marked elevation of plasma cytokines/chemokines. Pre-ischemic treatment of mice with DMPP or nicotine significantly decreased plasma-ALT and cytokines after 3 h of reperfusion. After 6 h of reperfusion, the protective effect of DMPP decreased and reached a negligible level by 24 h of reperfusion, despite significantly low levels of plasma cytokines. Histopathology showed markedly diminished hepatocellular injury in DMPP- and nicotine-pretreated mice during the early-phase of hepatic-IR, which reached a level comparable to saline-treated mice at late-phase of IR. CONCLUSION: Pharmacological modulation of the cholinergic pathway provides a means to modulate cytokine production and to delay IR-induced heaptocellular injury

    Characterisation of the dip-bump structure observed in proton-proton elastic scattering at root s=8 TeV

    The TOTEM collaboration at the CERN LHC has measured the differential cross-section of elastic proton-proton scattering at root s = 8 TeV in the squared four-momentum transfer range 0.2 GeV2 &lt; vertical bar t vertical bar &lt; 1.9 GeV2. This interval includes the structure with a diffractive minimum ("dip") and a secondary maximum ("bump") that has also been observed at all other LHC energies, where measurements were made. A detailed characterisation of this structure for root s = 8 TeV yields the positions, vertical bar t vertical bar(dip) = (0.521 +/- 0.007) GeV2 and vertical bar t vertical bar(bump) = (0.695 +/- 0.026) GeV2, as well as the cross-section values, d sigma/dt vertical bar(dip) = (15.1 +/- 2.5) mu b/GeV2 and d sigma/dt vertical bar(bump) = (29.7 +/- 1.8) mu b/Ge-2, for the dip and the bump, respectively

    Fostering children affected by AIDS in Richards Bay, South Africa: A qualitative study of grandparents' experiences

    Grandparents are increasingly becoming the primary carers of children orphaned by the HIV epidemic in South Africa. Traditional family roles are being reversed as aging family members take responsibility for the physical and psychosocial needs of children. This study uses qualitative research to explore the experiences of grandparents fostering children orphaned by AIDS in Richards Bay, KwaZulu-Natal Province, South Africa. The idea was born after a local HIV support organisation (Richards Bay Family Care) observed a trend within their organisation of grandparents increasingly becoming foster parents for orphans. An exploratory study was conducted in the organisation's three target areas (two rural villages and urban Richards Bay); the ultimate aim was to explore options for improving financial and emotional support for this group. The qualitative research methods included: four focus group discussions with foster-carers and community leaders (including two pocket-chart voting exercises); 12 in-depth interviews with grandparent foster-carers; and ten key-informant interviews, mostly with staff who provide support services. Data analysis was by thematic framework. The needs of the grandparent foster-carers varied: the rural participants were essentially concerned with meeting children's basic needs (housing, food and education), while those in urban areas more often felt pressure to provide emotional and psychological support for orphans. In both groups, women were at the forefront of foster care. Important problems identified by the grandparents were child discipline and a feeling of disharmony in the intergenerational relationship. Government foster care grants were identified as a regular source of income for especially the rural foster-carers. The findings may help programme managers better understand the differences in the needs of urban and rural foster parents. The experiences of grandparents as carers of orphans affected by HIV or AIDS are unique considering their age, frailty and poverty status. More research is needed on the role played by men in fostering children orphaned by AIDS
