    Genetic variability and structure of SNP haplotypes in the DMPK gene in Yakuts and other ethnic groups of northern Eurasia in relation to myotonic dystrophy

    The genetic variability of the DMPK locus has been studied in relation to six SNP markers (rs2070736, rs572634, rs1799894, rs527221, rs915915, and rs10415988) in Yakuts with myotonic dystrophy (MD) in the Yakut population and in populations of northern Eurasia. Significant differences were observed in the allele frequencies between patients and a population sample of Yakuts for three SNP loci (rs915915, rs1799894, and rs0415988) associated with a high chance of disease manifestation. The odds ratios (OR) of MD development in representatives of the Yakut population for these three loci were 2.59 (95% CI, p = 0,004), 4.99 (95% CI, p = 0.000), and 3.15 (95% CI, p = 0.01), respectively. Haplotype TTTCTC, which is associated with MD, and haplotype GTCCTT, which was observed only in Yakut MD patients (never in MD patients of non-Yakut origin), were revealed. A low level of variability in the locus of DMRK gene in Yakuts (He = 0.283) compared with other examined populations was noted. An analysis of pairwise genetic relationships between populations revealed their significant differentiation for all the examined loci. In addition, a low level of differentiation in territorial groups of Yakut populations (FST = 0.79%), which was related to the high subdivision of the northern Eurasian population (FST = 11.83%), was observed

    Alkoxy compounds - XXI. Synthesis, configuration, and conformation of some 5-alkoxymethyl-2,5-dialkyl-1,3-dioxanes

    Twenty-six previously unreported 5-alkoxymethyl-2,5-dialkyl-1,3-dioxanes have been synthesized by the condensation of 2-alkoxymethyl-2-alkylpropane-1,3-diols with aldehydes. It has been shown by an analysis of the PMR spectra of these dioxanes that the 1,3-dioxanes considered are mixtures of two stereoisomers. In some cases these mixtures have been resolved into the individual isomers by vacuum fractionation in efficient columns. The configurations and conformations of the stereoisomeric 5-alkoxymethyl-2,5-dialkyl-1,3-dioxanes have been established by the NMR method; it has been shown that the lower-boiling isomers have the trans configuration and predominantly the chair conformation and the higherboiling isomers the cis configuration and predominantly an unsymmetrical boat conformation. © 1970 The Faraday Press, Inc

    Single-Dose Bioequivalence Study of Rivaroxaban-Containing Medicinal Products in Healthy Volunteers

    Therapeutically, new oral anticoagulants (NOACs) are considered to be non-inferior or superior to vitamin K antagonists (warfarin). NOACs are included in current guidelines for the treatment of various cardiovascular diseases. Rivaroxaban medicinal products have been shown to effectively fight thrombotic complications of the new coronavirus infection, COVID-19. The wide clinical use of rivaroxaban products motivates the development of generics.The aim of the study was to compare the pharmacokinetics and safety of rivaroxaban medicinal products in a single-dose bioequivalence study in healthy volunteers under fasting conditions.Materials and methods: the bioequivalence study compared single-dose oral administration of Rivaroxaban, 10 mg film-coated tablets (NovaMedica Innotech LLC, Russia), and the reference product Xarelto®, 10 mg filmcoated tablets (Bayer AG, Germany), in healthy volunteers under fasting conditions. The open, randomised, crossover trial included 46 healthy volunteers. Each of the medicinal products (the test product and the reference product) was administered once; blood samples were collected during the 48 h after the administration. The washout between the study periods lasted 7 days. Rivaroxaban was quantified in plasma samples of the volunteers by high performance liquid chromatography coupled with tandem mass spectrometry (HPLC-MS/MS).Results: no adverse events or serious adverse events were reported for the test and reference products during the study. The following pharmacokinetic parameters were obtained for Rivaroxaban and Xarelto®, respectively: Cmax of 134.6 ± 58.0 ng/mL and 139.9 ± 49.3 ng/mL, AUC0–48 of 949.7 ± 354.5 ng×h/mL and 967.6 ± 319.9 ng×h/mL, AUC 0–∞ of 986.9 ± 379.7 ng×h/mL and 1003.6 ± 320.4 ng×h/mL, T1/2 of 8.2 ± 3.2 h and 7.8 ± 3.3 h. The 90% confidence intervals for the ratios of Cmax, AUC0–48, and AUC0–∞ geometric means were 88.04–108.67%, 89.42–104.92% and 89.44–104.81%, respectively.Conclusions: the test product Rivaroxaban and the reference product Xarelto® were found to have similar rivaroxaban pharmacokinetics and safety profiles.     The development and application of modern integrated methods for studying the efficiency of heat recovery systems and their individual elements enables ensuring their maximum efficiency. These methods are based on a combination of methods of exergic analysis with statistical methods of experiment planning, methods of the theory of linear systems, thermodynamics of irreversible processes, etc. A method is developed for calculating the losses of exergic power in heat conduction processes for a gas-air plate heat exchanger. Losses of exergic power occur when heat is transferred through the cross-section of the heat recovery plate. The methodology is based on a complex approach combining exergy methods with methods of thermodynamics of irreversible processes. The mathematical model of the investigated processes includes the exergy equation, the balance equation of exergy and entropy, the equation of continuity, the equation of phase motion, the energy equation, the balance equation for enthalpies, the Gibbs equation and the heat equation for boundary conditions of the third kind for an unbounded plate. A plate of a gas-air plate heat exchanger was modelled as an unlimited plate thickness. When obtaining formulas for calculating the losses of exergic power, the local differential equation of the exergy balance obtained earlier was used. In this equation, one of the terms determines the losses of exergy power, which are caused by the irreversibility of the processes. Such losses are related to thermal conductivity, phase viscosity, phase-to-phase heat transfer and friction between phases. On the basis of this equation and the solution of the heat equation under boundary conditions of the third kind, formulas are obtained for calculating the losses of exergy power. Calculations of total losses of exergic power in a gas-air plate heat exchanger and losses of exergic power in heat conduction processes under various operating conditions of the boiler are performed. It is established that losses of exergic power in heat conduction processes in a gas-air plate heat exchanger make up 8.6-11.6 % of the total losses of exergy power and depend on the operating mode of the boiler. Changing in the necessary direction the thermophysical characteristics of the gas-air plate heat exchanger, it is possible to reduce the loss of exergy power.Розроблено методику розрахунку втрат ексергетичної потужності у процесах теплопровідності під час передачі теплоти через поперечний переріз пластини газоповітряного пластинчастого теплоутилізатора за граничних умов третього роду. Методику засновано на комплексному підході, що поєднує ексергетичні методи з методами термодинаміки незворотних процесів. Математична модель досліджуваних процесів включає рівняння ексергії, рівняння балансу ексергії та ентропії, рівняння нерозривності трифазної термодинамічної системи при зміні концентрації однієї з фаз, рівняння руху фаз, рівняння енергій, рівняння балансу ентальпій, рівняння Гіббса і рівняння теплопровідності за граничних умов третього роду. Для отримання формул для розрахунку втрат ексергетичної потужності використано локальне диференціальне рівняння балансу ексергії. У цьому рівнянні одна зі складових визначає втрати ексергетичної потужності, зумовлені незворотністю процесів і пов'язані з теплопровідністю, в'язкістю фаз, міжфазним теплообміном і тертям між фазами. На підставі цього рівняння і рішення рівняння теплопровідності за граничних умов третього роду для необмеженої пластини, якою моделювалася пластина газоповітряного пластинчастого теплоутилізатора, отримано формули для розрахунку втрат ексергетичної потужності. Виконано розрахунки загальних втрат ексергетичної потужності в газоповітряному пластинчастому теплоутилізаторі за різних режимів роботи котла і втрат ексергетичної потужності у процесах теплопровідності. Встановлено, що втрати ексергетичної потужності у процесах теплопровідності в газоповітряному пластинчатому теплоутилізаторі становлять 8,6-11,6 % від загальних втрат ексергетичної потужності і залежать від режиму роботи котла

    Optimized properties of live vaccine influenza reassortant strains obtained by reverse genetics

    Classical reassortment in developing chicken eggs is a well-established technique for obtaining LAIV strains. Naturally generated reassortant vaccine strains are characterized by high reproductive capacity, genetically stable characteristics of temperature sensitivity and cold resistance, which correspond to the characteristics of the MDV involved in crossing with the epidemic virus. Along with antigenic relevance, natural reassortment ensures attenuation of vaccine strains, good reproduction capacity in upper respiratory tract cells and inability to reproduction in the lower respiratory tract. With classical reassortment, the speed and efficiency of obtaining vaccine reassortants largely depend on the properties of epidemic virus, and therefore cannot be stable. The potential of reverse genetics is attractive because it allows to obtain vaccine reassortants quickly and efficiently, reduce the likelihood of spontaneous mutations; however, the vaccine strain is deprived of the advantages of natural selection, in which the most viable clones are selected. This study presents the results of comparatively assessed A(H3N2) LAIVs obtained in parallel by classical reassortment and reverse genetics according to criteria confirming that vaccine strains inherit the necessary properties that guarantee their harmlessness and high reproduction in chicken embryos. Strains for LAIV obtained by both methods retained all attenuating mutations inherited from the MDV, were highly reproductive at the optimal temperature, with temperature sensitivity corresponded to the MDV. However, strains obtained by reverse genetics, was observed to have partial loss of cold resistance in comparison with that of the MDV and classical reassortants. Reduced cold adaptation may negatively affect vaccine effectiveness. It is important that after several additional passages in chicken embryos at low temperature, the cold resistance of the vaccine strain, assembled by reverse genetics, was increased. Credibly that cold resistance is a phenotypic trait, the degree of manifestation of which depends on the temperature conditions of virus multiplication. The selective factor of reduced incubation temperature is missing in reverse genetics. In order for the cold-adapted phenotype to be fully realized, additional passages at low temperature of RG-reassortants are necessary. Thus, the reverse genetics method using plasmid technology allows to effectively prepare reassortant strains for LAIV. An important stage in obtaining vaccine strains using genetic engineering techniques should be the control of their cold-adapted phenotype and its optimization by additional passages at low temperature

    Low-temperature electron spin relaxation is studied by the optical orientation method in bulk n-GaAs with donor concentrations from 10^14 cm^{-3} to 5x10^17 cm^{-3}. A peculiarity related to the metal-to-insulator transition (MIT) is observed in the dependence of the spin lifetime on doping near n_D = 2x10^16 cm^{-3}.
