    Cu(II) and Ni(II) Interactions with the Terminally Blocked Hexapeptide Ac-Leu-Ala-His-Tyr-Asn-Lys-amide Model of Histone H2B (80–85)

    The N- and C-terminal blocked hexapeptide Ac-Leu-Ala-His-Tyr-Asn-Lys-amide (LAHYNK) representing the 80–85 fragment of histone H2B was synthesized and its interactions with Cu(II) and Ni(II) ions were studied by potentiometric, UV-Vis, CD, EPR, and NMR spectroscopic techniques in solution. Our data reveal that the imidazole N(3) nitrogen atom is the primary ligating group for both metal ions. Sequential amide groups deprotonation and subsequent coordination to metal ions indicated an {Nimidazole, 3Namide} coordination mode above pH∼9, in all cases. In the case of Cu(II)-peptide system, the almost exclusive formation of the predominant species CuL in neutral media accounting for almost 98% of the total metal ion concentration at pH 7.3 strongly indicates that at physiological pH values the sequence -LAHYNK- of histone H2B provides very efficient binding sites for metal ions. The imidazole pyrrole N(1) ionization (but not coordination) was also detected in species CuH−4L present in solution above pH ∼ 11

    The nation and the family: the impact of national identification and perceived importance of family values on homophobic attitudes in Lithuania and Scotland

    The meanings attached to the nation can be consequential for group members’ attitudes and beliefs. We examined how national identity definition can influence the extent of individuals’ homophobia with 159 Lithuanian and 176 Scottish university students who completed a questionnaire which measured their national identification, homophobia, and the extent to which they felt traditional family values were central to their nation’s identity. Consistent with nation-wide differences in the significance given to the family, Lithuanian participants perceived family values to be more important for their national identity and expressed higher levels of homophobia than did Scottish participants. Moreover, the relationship between level of national identification and homophobia was stronger in Lithuania than in Scotland. Analyses revealed that the perceived importance of family values helped explain the difference between homophobia levels in Lithuania and Scotland. In both sites we found an indirect effect of national identification on homophobia via the perceived importance of family values, but this effect was significantly stronger for Lithuanian participants. These findings illustrate the ways in which identification with the nation is relevant to attitudes concerning sexuality, and how this varies according to national context. Our work indicates that LGBT rights campaigns should be informed by the knowledge that homophobia may be perpetuated by national valorisation of the family

    A multi-method evaluation of the implementation of a cancer teamwork assessment and feedback improvement programme (MDT-FIT) across a large integrated cancer system

    Background: Globally, Multidisciplinary Teams (MDTs) are considered the gold standard for diagnosis and treatment of cancer and other conditions, but variability in performance has led to demand for improvement tools. MDT-FIT (Multidisciplinary Team Feedback for Improving Teamwork) is an improvement programme developed iteratively with over 100 MDTs (>= 1100 MDT-members). Complex interventions are often adapted to context, but this is rarely evaluated. We conducted a prospective evaluation of the implementation of MDT-FIT across an entire integrated care system (ICS).Methods: MDT-FIT was implemented within all breast cancer MDTs across an ICS in England (n = 10 MDTs; 275 medical, nursing, and administrative members). ICS managers coordinated the implementation across the three stages of MDT-FIT: set up; assessment (self-report by team members plus independent observational assessment); team-feedback and facilitated discussion to agree actions for improvement. Data were collected using process and systems logs, and interviews with a purposively selected range of participants. Analysis was theoretically grounded in evidence-based frameworks for implementation strategies and outcomes.Results: All 10 MDTs participated in MDT-FIT; 36 interviews were conducted. Data from systems and process logs covered a 9-month period. Adaptations to MDT-FIT by the ICS (e.g., coordination of team participation by ICS rather than individual hospitals; and reducing time protected for coordination) reduced Fidelity and Adoption of MDT-FIT. However, the Acceptability, Appropriateness and Feasibility of MDT-FIT remained high due to embedding implementation strategies in the development of MDT-FIT (e.g., stakeholder engagement, interactive support).Conclusions: This is a unique and comprehensive evaluation of the multi-site implementation of a complex team improvement programme. Findings support the imperative of considering implementation strategies when designing such programmes to minimize potentially negative impacts of adaptations in "real world" settings

    2023 EELS field tests at Athabasca Glacier as an icy moon analogue environment

    JPL is developing a versatile and highly intelligent Exobiology Extant Life Surveyor (EELS) robot that would enable access to subsurface oceans and near-surface liquid reservoirs through existing conduits, such as the vents at the south pole of Enceladus or the putative geysers on Europa. A key mobility requirement for future vent exploration missions will be the ability to carefully descend and hold position in the vent to collect and analyze samples while withstanding plume forces without human intervention. Furthermore, this must be accomplished in a highly uncertain environment, requiring versatile hardware and intelligent autonomy. To work towards that goal, we have prototyped the EELS 1.0 and EELS 1.5 robots for horizontal and vertical mobility, respectively, in icy terrain. Autonomous surface mobility of EELS 1.0 was previously validated in a variety of terrain, including snowy mountains, ice rinks, and desert sand. Vertical mobility of EELS 1.5 was developed on laboratory ice walls. This paper presents the first mobility trials for both robots on large-scale, natural icy terrain: the Athabasca Glacier located in Alberta, Canada, a terrestrial analogue to the surfaces and subsurfaces of icy moons. This paper provides a preliminary written record of the test campaign’s four major trials: 1) surface mobility with EELS 1.0, 2) vertical mobility with EELS 1.5, 3) science instrument validation, and 4) terramechanics experiments. During this campaign, EELS 1.5 successfully held position and descended ~1.5 m vertically in an icy conduit and EELS 1.0 demonstrated surface mobility on icy surfaces with undulations and slopes. A miniaturized capillary electrophoresis (CE) instrument built to the form factor of an EELS module was tested in flowing water on the glacier and successfully demonstrated automated sampling and in-situ analysis. Terramechanics experiments designed to better understand the interaction between different ice properties and the screws that propel the robot forwards were performed on horizontal and vertical surfaces. In this paper we report the outcomes of the four tests and discuss their implications for potential future icy missions. The field test also demonstrated EELS’s ability to support Earth science missions. Another potential near-term follow-on could be a technology demonstration on the Moon. This paper is a high level report on the execution of the field test. Data and results will be detailed in subsequent publications
