    Creativity as opening toward new beginnings

    The initiative to organize an issue of Academic Quarter on creativity originates from the 32th International Human Science Research Conference (IHSRC), which took place at the University of Aalborg in the Northern Denmark in August 2013. Here the overall theme of the conference was ’Creativity in Human Science Research, Methodology and Theory’. More than 150 human scientists from all over the world participated in the conference in Denmark. According to the conference flyer:   "All human sciences exist in a tension between tradition and renewal. At the conference, we hope that participants will discuss how to renew the human sciences creatively, and also to present ideas about what creativity is as a basic human phenomenon. How can phenomenological, hermeneutic and other human science traditions be respected and yet renewed in creative directions? How – and how much – should human scientists experiment with creative methodological practices when researching human phenomena? What role can the arts play? Are there limits to creativity? Can human beings become too creative – in life as well as in research? And what can human scientists actually contribute with to the current creativity discourse?"   Many of those who have contributed to the present issue attended the conference. This issue raises a fundamental question that must be of importance for every human scientist: What is creativity really? And how can and why should a more ‘creative’ approach in human science be understood and defended? In what ways can phenomenology and hermeneutics become more open to insights into the nature of creativity and how should we talk about creative ways of doing phenomenology, hermeneutics, and human science as such? In the current issue, you will find many different answers to what creativity is and what a creative human science approach could look like, and how to practice it.     Phosphodiesterases in non-neoplastic appearing colonic mucosa from patients with colorectal neoplasia

    BACKGROUND: Intracellular signaling through cyclic nucleotides, both cyclic AMP and cyclic GMP, is altered in colorectal cancer. Accordingly, it is hypothesized that an underlying mechanism for colorectal neoplasia involves altered function of phosphodiesterases (PDEs), which affects cyclic nucleotide degradation. Here we present an approach to evaluate the function of selected cyclic nucleotide-PDEs in colonic endoscopic biopsies from non-neoplastic appearing mucosa. METHODS: Biopsies were obtained from patients with and without colorectal neoplasia. Activities of PDEs were characterized functionally by measurements of transepithelial ion transport and their expression and localization by employing real-time qPCR and immunohistochemistry. RESULTS: In functional studies PDE subtype-4 displayed lower activity in colorectal neoplasia patients (p = 0.006). Furthermore, real-time qPCR analysis showed overexpression of subtype PDE4B (p = 0.002) and subtype PDE5A (p = 0.02) in colorectal neoplasia patients. Finally, immunohistochemistry for 7 PDE isozymes demonstrated the presence of all 7 isozymes, albeit with weak reactions, and with no differences in localization between colorectal neoplasia and control patients. Of note, quantification of PDE subtype immunostaining revealed a lower amount of PDE3A (p = 0.04) and a higher amount of PDE4B (p = 0.02) in samples from colorectal neoplasia patients. CONCLUSION: In conclusion, functional data indicated lower activity of PDE4 subtypes while expressional and abundance data indicated a higher expression of PDE4B in patients with colorectal neoplasia. We suggest that cyclic nucleotide-PDE4B is overexpressed as a malfunctioning protein in non-neoplastic appearing colonic mucosa from patients with colorectal neoplasia. If a predisposition of reduced PDE4B activity in colonic mucosa from colorectal neoplasia patients is substantiated further, this subtype could be a potential novel early diagnostic risk marker and may even be a target for future medical preventive treatment of colorectal cancer.     Combined activity of COX-1 and COX-2 is increased in non-neoplastic colonic mucosa from colorectal neoplasia patients

    Abstract Background Cyclooxygenase (COX) activity is increased in endoscopic normal colonic mucosa from patients with colorectal neoplasia (CRN). COX-2 is thought to be the predominant COX isozyme involved in neoplasia. Meanwhile, relative contributions of COX-1 and COX-2 isoforms are unknown. Knowledge about their mutual activity in colonic mucosa is important for diagnostics and targeted therapy for CRN. The aim of this study was to assess the relative function, expression and localization of COX-1 and COX-2 enzymes in colonic non-neoplastic human mucosa and thereby to potentially reveal a mucosal disease predisposition for better treatment. Methods Biopsies were pinched from normal appearing colonic mucosa in patients undergoing endoscopy. Ussing chamber technique was applied for an indirect assessment of epithelial activity, RT-qPCR for expression and immunohistochemistry for localization of COX-1 and COX-2 enzymes in patients without (ctrls) and with a history of CRN (CRN-pts). Results Combined COX-1 and COX-2 activity was higher in CRN-pts, p = 0.036. COX-2 was primarily localized in absorptive cells, while COX-1 appeared to be restricted to nonenteroendocrine tuft cells of the colonic epithelium. Conclusions In biopsies from endoscopic normal appearing colonic mucosa, combined activity of COX-1 and COX-2 enzymes is increased in CRN-pts compared with ctrls. This indicates that COX-1 and COX-2 together contribute to an increased proliferation process. Of note, in colonic epithelial cell lining, the COX-1 enzyme seems localized in tuft cells

    Elective induction of labor: A prospective observational study.

    The aim of the present study was to assess indications for induction and describe the characteristics and delivery outcome in medical compared to non-medical/elective inductions. During a three-month period, 1663 term inductions were registered in 24 delivery units in Norway. Inclusion criteria were singleton pregnancies with cephalic presentation at gestational age 37+0 and beyond. Indications, pre-induction Bishop scores, mode of delivery and adverse maternal and fetal outcomes were registered, and compared between the medically indicated and elective induction groups. Ten percent of the inductions were elective, and the four most common indications were maternal request (35%), a previous negative delivery experience or difficult obstetric history (19%), maternal fatigue/tiredness (17%) and anxiety (15%). Nearly half of these inductions were performed at 39+0-40+6 weeks. There were fewer nulliparous women in the elective compared to the medically indicated induction group, 16% vs. 52% (p<0.05). The cesarean section rate in the elective induction group was 14% and 17% in the medically indicated group (14% vs. 17%, OR = 0.8, 95% CI 0.5-1.3). We found that one in ten inductions in Norway is performed without a strict medical indication and 86% of these inductions resulted in vaginal delivery

    Synthesis of β‑Cyclodextrin Diazonium Salts and Electrochemical Immobilization onto Glassy Carbon and Gold Surfaces

    This study shows that diazotized β-cyclodextrin (β-CD) can be produced, isolated, and immobilized onto glassy carbon and gold surfaces. 4-(1,2,3-Triazol-4-yl)­benzenediazonium-β-CD tetrafluoroborate (pDz-β-CD) and 3-(1,2,3-triazol-4-yl)­benzenediazonium-β-CD tetrafluoroborate (mDz-β-CD) were successfully prepared by Cu<sup>(I)</sup>-catalyzed azide alkyne coupling (CuAAC) of 6-monodeoxy-6-monoazido-β-cyclodextrin (N<sub>3</sub>-β-CD) and 4-ethynylaniline and 3-ethynylaniline, respectively, followed by diazotization. The products were isolated and stored successfully for several months at −18 °C. The intermediates and products were verified by Attenuated Total Reflectance Fourier Transform Infrared, Nuclear Magnetic Resonance, and Heteronuclear Single Quantum Coherence.pDz-β-CD and mDz-β-CD were immobilized onto glassy carbon and gold surfaces facilitated by electrochemical reduction of the diazonium group. The thus generated aryl radical reacted with the surface. The modified gold surfaces were investigated by Polarization Modulation Infrared Reflection Absorption Spectroscopy and cyclic voltammetry employing the redox probe K<sub>3</sub>Fe­(CN)<sub>6</sub> to analyze the extent of blocking of the surfaces. Finally, the availability of the cavity of surface-immobilized β-CD was shown by complexation of ferrocene followed by cyclic voltametric analysis
