Estimation in sera by radioimmunoassay of a specific membrane antigen associated with a murine lymphoma.

Material with a molecular weight of less than 10(5) daltons has been isolated and partially purified from the ascitic fluid of DBA2 mice bearing a syngeneic lymphoma (SL2). This substance inhibits the cytotoxic action of an allogeneic antiserum directed specifically against SL2 cells. Material has been rendered radioactive with 125I and between 20 and 25% of the radioactivity is bound in a specific manner to the antiserum. The material which is referred to as 125I-TSTA has been used in a radioimmunoassay to measure the level of TSTA in the sera of mice bearing both ascitic and subcutaneous SL2 tumours. The level of circulating TSTA was found to be high immediatley following inoculation of live SL2 cells, probably because a large proportion of the injected cells autolyse. The serum concentration of TSTA then falls but 6-10 days later begins to rise again in parallel with the growth of the SL2 tumour either in the peritoneal cavity or subcutaneously. Following surgical removal of an intradermal SL2 tumour the level of TSTA in the serum falls rapidly. No evidence could be found that a significant proportion of the TSTA in the serum of tumour-bearing mice is completed with antibody. However, in the serum of DBA2 mice which have been hyperimmunized with irradiated SL2 cells there are antibodies which bind 125I-TSTA although syngeneic anti-SL2 sera, unlike alloantisera, do not show complement dependent lysis of SL2 cells

Characterisation of gastroenteritis associated adenoviruses in South Africa

Objective. To analyse adenovirus (Ad) numbers and types associated with paediatric gastro-enteritis in South AfricaSetting. Gauteng, 1994-1996.Methods. A total of 234 paediatric diarrhoeal stool samples were screened for Ad using commercial enzyme-linked iInmunosorbent assays (EUSAs). Adenoviral isolates were typed, where possibie, using restriction enzyme analysis.Results. Ad was detected in 23 (9.8%) specimens, of which 8 (34.8%) were found by subgroup F-specilic EUSA to contain Ad40 or 41. Six of these isolates were typed and 2 could not be typed. Of the remaining 15 specimens, 2 isolates had restriction profiles that did not correspond with known Ads, while 2were identified as Ad31 and 1 as a subgroup CAd. The remaining 10 specimens negative for Ad40/41 were noncultivable and could not be typed.Conclusions. The high percentage of non-eultivable Ads other than Ad40/41 is unusual, and may possibly indicate the prevalence of hexon variants of Ad40/41 or of emerging Ad types in South Africa

Measuring Attachment and Reflective Functioning in Early Adolescence:An Introduction to the Friends and Family Interview

Internal working models (IWMs; Bowlby, 1969/1982) develop before language and are, initially at least, pre-symbolic, nonverbal notions. With reflective functioning (RF; Fonagy, Steele, Steele, Moran, & Higgitt, 1991) we have the possibility to refashion IWMs based on language, but linguistic skills only develop between 18-24 months, and then steadily over time. Reliable instruments are available to assess these constructs in infancy and adulthood: The Strange Situation observational measure (Ainsworth, Blehar, Waters, & Wall, 1978) reveals the infant’s IWMs of his caregivers, while the Adult Attachment Interview (AAI; Main, Hesse, & Goldwyn, 2008; George, Kaplan, & Main, 1985) exposes the adult speaker’s capacity for RF. This paper addresses the middle ground of early adolescent children who are not yet mature enough to respond to a full AAI, but are too old to expect that an observational attachment measure would reveal much about their inner thoughts, feelings, and beliefs about attachment. We outline an interview protocol designed for 9 to 16-year old children, asking about self, friends, teachers, and family, with the aim of elucidating both IWMs, regarding earlier experience, and the extent of RF concerning past and present experiences. The protocol is the Friends and Family Interview (FFI; Steele & Steele, 2005), which has a mul-tidimensional scoring system to be elaborated with verbatim examples of response from both low-risk community samples, and higher-risk samples of youth
