    Characterization of the 2016-2017 dermatology standardized letter of recommendation

Objective: We aimed to analyze the reformatted standard letter of recommendation (SLOR) for dermatology residents to examine trends in grading and content based on the positions of the letter writers, their backgrounds, and their relationship with the applicant, as well as to evaluate the SLOR\u27s ability to discriminate applicants. Design: This was a retrospective characterization study of dermatology SLORs from the 2016-17 application cycle. Setting: We examined SLORs received by The Ohio State University, the University of Oklahoma, and Hofstra University Northwell Health dermatology residency programs. Participants: We included dermatology residency applicants and their letter writers from the 2016-17 application cycle. Results: A total of 141 SLORs were analyzed from 115 applicants. SLORs demonstrated grade inflation from letter writers of all backgrounds. Ratings for research potential and inquisitive nature were significantly lower than ratings for other categories. Letter writers with limited clinical and research contact graded applicants significantly lower than did writers who had more extensive contact. Word boxes were underutilized. Conclusion: The dermatology SLOR is useful in differentiating applicants, and ratings correlate with the relationships that letter writers have with their applicants. Residency programs should be aware of these findings when evaluating letters of recommendation for applicants

    Food Matters: Food Insecurity among Pregnant Adolescents and Infant Birth Outcomes

    Objectives: The objectives of this study are to: (1) document prevalence of food insecurity among pregnant adolescents; (2) determine if food insecurity is associated with adverse birth outcomes (i.e., lower birth weight, earlier gestational age) among their newborns; and (3) examine whether depressive symptoms, anxiety, nutrition and/or weight gain mediate these associations. Methods: Pregnant adolescents (14-21 years old; N-881) in prenatal care at community hospitals and health centers in New York City completed a health and psychosocial survey during second and third trimesters of pregnancy. Birth weight and gestational age were recorded from medical records. Results: Over one-half of the adolescents reported food insecurity. Path analyses demonstrated that food insecurity was associated with lower birth weight and earlier gestational age. Depressive symptoms mediated these associations. Conclusions: Pregnant adolescents experience high rates of food insecurity. Those who were food insecure experienced more depressive symptoms, which in turn predicted adverse birth outcomes. Programs and policies should target these vulnerable children to stem the multi-generational effects of food insecurity

    S -Nitrosothiol-Modified Dendrimers as Nitric Oxide Delivery Vehicles

    The synthesis and characterization of two generation-4 polyamidoamine (PAMAM) dendrimers with S-nitrosothiol exteriors are reported. The hyperbranched macromolecules were modified with either N-acetyl-D,L-penicillamine (NAP) or N-acetyl-L-cysteine (NACys) and analyzed via 1H and 13C-NMR, UV absorption spectroscopy, MALDI-TOF mass spectrometry, and size exclusion chromatography. Treatment of the dendritic thiols with nitrite solutions yielded the corresponding S-nitrosothiol nitric oxide (NO) donors (G4-SNAP, G4-NACysNO). Chemiluminescent NO detection demonstrated that the dendrimers were capable of storing ~2 μmol NO·mg−1 when exposed to triggers of S-nitrosothiol decomposition (e.g., light and copper). The kinetics of NO-release were found to be highly dependent on the structure of the nitrosothiol (i.e., tertiary vs. primary) and exhibited similar NO-release characteristics to classical small molecule nitrosothiols reported in the literature. As demonstration of utility, the ability of G4-SNAP to inhibit thrombin-mediated platelet aggregation was assayed. At equivalent nitrosothiol concentrations (25 μM), the G4-SNAP dendrimer resulted in a 62% inhibition of platelet aggregation, compared to only 17% for the small molecule NO donor. The multivalent NO storage, the dendritic effects exerted on nitrosothiol stability and reactivity, and the utility of dendrimers as drug delivery vehicles highlight the potential of these constructs as clinically useful S-nitrosothiol-based therapeutics

    Updated resonance photo-decay amplitudes to 2 GeV

    We present the results of an energy-dependent and set of single-energy partial-wave analyses of single-pion photoproduction data. These analyses extend from threshold to 2 GeV in the laboratory photon energy, and update our previous analyses to 1.8 GeV. Photo-decay amplitudes are extracted for the baryon resonances within this energy range. We consider two photoproduction sum rules and the contributions of two additional resonance candidates found in our most recent analysis of πN\pi N elastic scattering data.     Relationships between depth and δ15N of Arctic benthos vary among regions and trophic functional groups

This relationship is commonly attributed to preferential uptake of 14N from sinking particulate organic matter (POM) by microbes, and suggests that relationships between δ15N and water depth may be affected by local POM sources and flux dynamics. We examined the relationships between δ15N and water depth (20–500 m) for six trophic functional groups using a mixed effects modelling approach, and compared relationships between two contiguous Arctic marine ecosystems with different POM sources and sinking export dynamics: the Canadian Beaufort Sea and Amundsen Gulf. We demonstrate for the first time in the Arctic that δ15N values of mobile epifaunal carnivores increased as a function of depth when considered separately from benthopelagic and infaunal carnivores, which contrarily did not exhibit increasing δ15N with depth. The δ15N of suspension/filter feeders, infaunal deposit feeders and bulk sediment also increased with water depth, and the slopes of the relationships were steeper in the Amundsen Gulf than in the Beaufort Sea. We propose that regional differences in slopes reflect differences in POM sources exported to the benthos. In the Beaufort Sea, terrestrial POM discharged from the Mackenzie River quantitatively dominates the sedimentary organic matter across the continental shelf and slope, dampening change in δ15N of benthic POM with depth. In the Amundsen Gulf, we attribute a faster rate of change in δ15N of POM with increasing depth to larger contributions of marine-derived POM to the benthic sedimentary pool, which had likely undergone extensive biological transformation in the productive offshore pelagic zone.     Generation and evaluation of a Glaesserella (Haemophilus) parasuis capsular mutant

    Glaesserella (Haemophilus) parasuis is a commensal of the upper respiratory tract in pigs and also the causative agent of Glässer's disease, which causes significant morbidity and mortality in pigs worldwide. Isolates are characterized into 15 serovars by their capsular polysaccharide, which has shown a correlation to isolate pathogenicity. To investigate the role capsule plays in G. parasuis virulence and host interaction, a capsule mutant of the serovar 5 strain HS069 was generated (HS069Δcap) through allelic exchange following natural transformation. HS069Δcap was unable to cause signs of systemic disease during a pig challenge study and had increased sensitivity to complement killing and phagocytosis by alveolar macrophages. When compared to the parent strain, HS069Δcap produced more robust biofilm and adhered equivalently to 3D4/31 cells; however, it was unable to persistently colonize the nasal cavity of inoculated pigs, with all pigs clearing HS069Δcap by 5 days post-challenge. Our results indicate the importance of capsular polysaccharide to G. parasuis virulence as well as nasal colonization in pigs

    Bactericidal Efficacy of Nitric Oxide-Releasing Silica Nanoparticles

    The utility of nitric oxide (NO)-releasing silica nanoparticles as a novel antibacterial is demonstrated against Pseudomonas aeruginosa. Nitric oxide-releasing nanoparticles were prepared via co-condensation of tetraalkoxysilane with aminoalkoxysilane modified with diazeniumdiolate NO donors, allowing for the storage of large NO payloads. Comparison of the bactericidal efficacy of the NO-releasing nanoparticles to 1-[2-(carboxylato)pyrrolidin-1-yl]diazen-1-ium-1,2-diolate (PROLI/NO), a small molecule NO donor, demonstrated enhanced bactericidal efficacy of nanoparticle-derived NO and reduced cytotoxicity to healthy cells (mammalian fibroblasts). Confocal microscopy revealed that fluorescently-labeled NO-releasing nanoparticles associated with the bacteria, providing rationale for the enhanced bactericidal efficacy of the nanoparticles. Intracellular NO concentrations were measurable when the NO was delivered from nanoparticles as opposed to PROLI/NO. Collectively, these results demonstrate the advantage of delivering NO via nanoparticles for antimicrobial applications

    Interactive metagenomic visualization in a Web browser

    <p>Abstract</p> <p>Background</p> <p>A critical output of metagenomic studies is the estimation of abundances of taxonomical or functional groups. The inherent uncertainty in assignments to these groups makes it important to consider both their hierarchical contexts and their prediction confidence. The current tools for visualizing metagenomic data, however, omit or distort quantitative hierarchical relationships and lack the facility for displaying secondary variables.</p> <p>Results</p> <p>Here we present Krona, a new visualization tool that allows intuitive exploration of relative abundances and confidences within the complex hierarchies of metagenomic classifications. Krona combines a variant of radial, space-filling displays with parametric coloring and interactive polar-coordinate zooming. The HTML5 and JavaScript implementation enables fully interactive charts that can be explored with any modern Web browser, without the need for installed software or plug-ins. This Web-based architecture also allows each chart to be an independent document, making them easy to share via e-mail or post to a standard Web server. To illustrate Krona's utility, we describe its application to various metagenomic data sets and its compatibility with popular metagenomic analysis tools.</p> <p>Conclusions</p> <p>Krona is both a powerful metagenomic visualization tool and a demonstration of the potential of HTML5 for highly accessible bioinformatic visualizations. Its rich and interactive displays facilitate more informed interpretations of metagenomic analyses, while its implementation as a browser-based application makes it extremely portable and easily adopted into existing analysis packages.
