    Modulation of TCR signalling components occurs prior to positive selection and lineage commitment in iNKT cells

    iNKT cells play a critical role in controlling the strength and character of adaptive and innate immune responses. Their unique functional characteristics are induced by a transcriptional program initiated by positive selection mediated by CD1d expressed by CD4+CD8+ (double positive, DP) thymocytes. Here, using a novel Vα14 TCR transgenic strain bearing greatly expanded numbers of CD24hiCD44loNKT cells, we examined transcriptional events in four immature thymic iNKT cell subsets. A transcriptional regulatory network approach identified transcriptional changes in proximal components of the TCR signalling cascade in DP NKT cells. Subsequently, positive and negative selection, and lineage commitment, occurred at the transition from DP NKT to CD4 NKT. Thus, this study introduces previously unrecognised steps in early NKT cell development and separates, the events associated with modulation of the T cell signalling cascade prior to changes associated with positive selection and lineage commitment

    On the finite-size behavior of systems with asymptotically large critical shift

    Exact results of the finite-size behavior of the susceptibility in three-dimensional mean spherical model films under Dirichlet-Dirichlet, Dirichlet-Neumann and Neumann-Neumann boundary conditions are presented. The corresponding scaling functions are explicitly derived and their asymptotics close to, above and below the bulk critical temperature TcT_c are obtained. The results can be incorporated in the framework of the finite-size scaling theory where the exponent λ\lambda characterizing the shift of the finite-size critical temperature with respect to TcT_c is smaller than 1/ν1/\nu, with ν\nu being the critical exponent of the bulk correlation length.Comment: 24 pages, late

    Interference of Selected Palmer Amaranth ( Amaranthus palmeri

    Palmer amaranth (Amaranthus palmeri S. Wats.) has become difficult to control in row crops due to selection for biotypes that are no longer controlled by acetolactate synthase inhibiting herbicides and/or glyphosate. Early season interference in soybean [Glycine max (L.) Merr.] for 40 days after emergence by three glyphosate-resistant (GR) and three glyphosate-susceptible (GS) Palmer amaranth biotypes from Georgia and North Carolina was compared in the greenhouse. A field experiment over 2 years compared season-long interference of these biotypes in soybean. The six Palmer amaranth biotypes reduced soybean height similarly in the greenhouse but did not affect soybean height in the field. Reduction in soybean fresh weight and dry weight in the greenhouse; and soybean yield in the field varied by Palmer amaranth biotypes. Soybean yield was reduced 21% by Palmer amaranth at the established field density of 0.37 plant m−2. When Palmer amaranth biotypes were grouped by response to glyphosate, the GS group reduced fresh weight, dry weight, and yield of soybean more than the GR group. The results indicate a possible small competitive disadvantage associated with glyphosate resistance, but observed differences among biotypes might also be associated with characteristics within and among biotypes other than glyphosate resistance

    Model-based evaluation of the long-term cost-effectiveness of systematic case-finding for COPD in primary care

    Introduction'One-off' systematic case-finding for COPD using a respiratory screening questionnaire is more effective and cost-effective than routine care at identifying new cases. However, it is not known whether early diagnosis and treatment is beneficial in the longer term. We estimated the long-term cost-effectiveness of a regular case-finding programme in primary care.MethodsA Markov decision analytic model was developed to compare the cost-effectiveness of a 3-yearly systematic case-finding programme targeted to ever smokers aged ≥50 years with the current routine diagnostic process in UK primary care. Patient-level data on case-finding pathways was obtained from a large randomised controlled trial. Information on the natural history of COPD and treatment effects was obtained from a linked COPD cohort, UK primary care database and published literature. The discounted lifetime cost per quality-adjusted life-year (QALY) gained was calculated from a health service perspective.ResultsThe incremental cost-effectiveness ratio of systematic case-finding versus current care was £16 596 per additional QALY gained, with a 78% probability of cost-effectiveness at a £20 000 per QALY willingness-to-pay threshold. The base case result was robust to multiple one-way sensitivity analyses. The main drivers were response rate to the initial screening questionnaire and attendance rate for the confirmatory spirometry test.DiscussionRegular systematic case-finding for COPD using a screening questionnaire in primary care is likely to be cost-effective in the long-term despite uncertainties in treatment effectiveness. Further knowledge of the natural history of case-found patients and the effectiveness of their management will improve confidence to implement such an approach

    Associations Between Slower Walking Speed and T1ρ Magnetic Resonance Imaging of Femoral Cartilage Following Anterior Cruciate Ligament Reconstruction

    Objective: To determine whether walking speed, collected at 6 and 12 months following anterior cruciate ligament reconstruction (ACLR), is associated with inter-extremity differences in proteoglycan density, measured via T1ρ magnetic resonance imaging, in tibiofemoral articular cartilage 12 months following ACLR. Methods: Twenty-one individuals with a unilateral patellar-tendon autograft ACLR (10 women and 11 men, mean ± SD age 23.9 ± 2.7 years, mean ± SD body mass index 23.9 ± 2.7 kg/m2) were recruited for participation in this study. Walking speed was collected using 3-dimensional motion capture at 6 and 12 months following ACLR. The articular cartilage of the medial femoral condyle (MFC) and lateral femoral condyle and medial and lateral tibial condyles was manually segmented and subsectioned into 3 regions of interest (anterior, central, and posterior) based on the location of the meniscus in the sagittal plane. Inter-extremity mean T1ρ relaxation time ratios (T1ρ ACLR extremity / T1ρ contralateral extremity) were calculated and used for analysis. Pearson product-moment correlations were used to determine associations between walking speed and inter-extremity differences in T1ρ relaxation time ratios. Results: Slower walking speed 6 months post-ACLR was significantly associated with higher T1ρ relaxation time ratios in the MFC of the ACLR extremity 12 months following ACLR (posterior MFC, r = −0.51, P = 0.02; central MFC, r = −0.47, P = 0.04). Similarly, slower walking speed at 12 months post-ACLR was significantly associated with higher T1ρ relaxation time ratios in the posterior MFC ACLR extremity (r = −0.47, P = 0.04) 12 months following ACLR. Conclusion: Slower walking speed at 6 and 12 months following ACLR may be associated with early proteoglycan density changes in medial femoral compartment cartilage health in the first 12 months following ACLR

    A preliminary study of the effect of closed incision management with negative pressure wound therapy over high-risk incisions

    Background Certain postoperative wounds are recognised to be associated with more complications than others and may be termed high-risk. Wound healing can be particularly challenging following high-energy trauma where wound necrosis and infection rates are high. Surgical incision for joint arthrodesis can also be considered high-risk as it requires extensive and invasive surgery and postoperative distal limb swelling and wound dehiscence are common. Recent human literature has investigated the use of negative pressure wound therapy (NPWT) over high-risk closed surgical incisions and beneficial effects have been noted including decreased drainage, decreased dehiscence and decreased infection rates. In a randomised, controlled study twenty cases undergoing distal limb high-energy fracture stabilisation or arthrodesis were randomised to NPWT or control groups. All cases had a modified Robert-Jones dressing applied for 72 h postoperatively and NPWT was applied for 24 h in the NPWT group. Morphometric assessment of limb circumference was performed at six sites preoperatively, 24 and 72 h postoperatively. Wound discharge was assessed at 24 and 72 h. Postoperative analgesia protocol was standardised and a Glasgow Composite Measure Pain Score (GCPS) carried out at 24, 48 and 72 h. Complications were noted and differences between groups were assessed. Results Percentage change in limb circumference between preoperative and 24 and 72 h postoperative measurements was significantly less at all sites for the NPWT group with exception of the joint proximal to the surgical site and the centre of the operated bone at 72 h. Median discharge score was lower in the NPWT group than the control group at 24 h. No significant differences in GCPS or complication rates were noted. Conclusions Digital swelling and wound discharge were reduced when NPWT was employed for closed incision management. Larger studies are required to evaluate whether this will result in reduced discomfort and complication rates postoperatively

    Molecular epidemiology of methicillin-resistant Staphylococcus aureus isolated from Australian veterinarians

    This work investigated the molecular epidemiology and antimicrobial resistance of methicillinresistant Staphylococcus aureus (MRSA) isolated from veterinarians in Australia in 2009. The collection (n = 44) was subjected to extensive molecular typing (MLST, spa, SCCmec, dru, PFGE, virulence and antimicrobial resistance genotyping) and antimicrobial resistance phenotyping by disk diffusion. MRSA was isolated from Australian veterinarians representing various occupational emphases. The isolate collection was dominated by MRSA strains belonging to clonal complex (CC) 8 and multilocus sequence type (ST) 22. CC8 MRSA (ST8-IV [2B], spa t064; and ST612-IV [2B] , spa variable,) were strongly associated with equine practice veterinarians (OR = 17.5, 95% CI = 3.3-92.5, P < 0.001) and were often resistant to gentamicin and rifampicin. ST22-IV [2B], spa variable, were strongly associated with companion animal practice veterinarians (OR = 52.5, 95% CI = 5.2-532.7, P < 0.001) and were resistant to ciprofloxacin. A single pig practice veterinarian carried ST398-V [5C2], spa t1451. Equine practice and companion animal practice veterinarians frequently carried multiresistant-CC8 and ST22 MRSA, respectively, whereas only a single swine specialist carried MRSA ST398. The presence of these strains in veterinarians may be associated with specific antimicrobial administration practices in each animal species

    Defining optimal soybean seeding rates and associated risk across North America

    Soybean [Glycine max (L.) Merr.] seeding rate research across North America is typically conducted in small geo-political regions where environmental effects on the seeding rate × yield relationship are minimized. Data from 211 individual field studies (∼21,000 data points, 2007–2017) were combined from across North America ranging in yield from 1,000– 7,500 kg ha−1. Cluster analysis was used to stratify each individual field study into similar environmental (soil × climate) clusters and into high (HYL), medium (MYL), and low (LYL) yield levels. Agronomically optimal seeding rates (AOSR) were calculated and Monte Carlo risk analysis was implemented. Within the two northern most clusters the AOSR was higher in the LYL followed by the MYL and then HYL. Within the farthest south cluster, a relatively small (±15,000 seeds ha−1) change in seeding rate from the MYL was required to reach the AOSR of the LYL and HYL, respectively. The increase in seeding rate to reach the LYL AOSR was relatively greater (5x) than the decrease to reach the HYL AOSR within the northern most cluster. Regardless, seeding rates below the AOSR presented substantial risk and potential yield loss, while seeding rates above provided slight risk reduction and yield increases. Specific to LYLs and MYLs, establishing and maintaining an adequate plant stand until harvest maximized yield regardless of the seeding rate, while maximizing seed number was important with lower seeding rates. These findings will help growers manage their soybean seed investment by adjusting seeding rates based upon the productivity of the environment.Fil: Gaspar, Adam P..
