    CD62L (L-selectin) shedding for assessment of perioperative immune sensitivity in patients undergoing cardiac surgery with cardiopulmonary bypass

    OBJECTIVE: To investigate the suitability of blood granulocyte and monocyte sensitivity, as measured by the quantity of different agonists required to induce CD62L shedding, for assessment of perioperative immune changes in patients undergoing cardiac surgery with cardiopulmonary bypass. METHODS: Patients scheduled for aortocoronary bypass grafting or for valve surgery were included in this prospective observational study. Blood samples were drawn before anesthesia induction, directly after surgery and 48 hours after anesthesia induction. We determined the concentration of two different inflammatory stimuli--lipoteichoic acid (LTA) and tumor necrosis factor alpha (TNF)--required to induce shedding of 50% of surface CD62L from blood granulocytes and monocytes. In parallel monocyte surface human leukocyte antigen (HLA)-DR, and plasma interleukin (IL)-8, soluble (s)CD62L, soluble (s)Toll-like receptor (TLR)-2 and ADAM17 quantification were used to illustrate perioperative immunomodulation. RESULTS: 25 patients were enrolled. Blood granulocytes and monocytes showed decreased sensitivity to the TLR 2/6 agonist Staphylococcus aureus LTA immediately after surgery (p = 0.001 and p = 0.004 respectively). In contrast, granulocytes (p = 0.01), but not monocytes (p = 0.057) displayed a decreased postoperative sensitivity to TNF. We confirmed the presence of a systemic inflammatory response and a decreased immune sensitivity in the post-surgical period by measuring significant increases in the perioperative plasma concentration of IL-8 (p </= 0.001) and sTLR (p = 0.004), and decreases in monocyte HLA-DR (p<0.001), plasma sCD62L (p </= 0.001). In contrast, ADAM17 plasma levels did not show significant differences over the observation period (p = 0.401). CONCLUSIONS: Monitoring granulocyte and monocyte sensitivity using the "CD62L shedding assay" in the perioperative period in cardiac surgical patients treated with the use of cardiopulmonary bypass reveals common changes in sensitivity to TLR2/6 ligands and to TNF stimulus. Further long-term follow-up studies will address the predictive value of these observations for clinical purposes

    Sequential Delivery of Host-Induced Virulence Effectors by Appressoria and Intracellular Hyphae of the Phytopathogen Colletotrichum higginsianum

    Phytopathogens secrete effector proteins to manipulate their hosts for effective colonization. Hemibiotrophic fungi must maintain host viability during initial biotrophic growth and elicit host death for subsequent necrotrophic growth. To identify effectors mediating these opposing processes, we deeply sequenced the transcriptome of Colletotrichum higginsianum infecting Arabidopsis. Most effector genes are host-induced and expressed in consecutive waves associated with pathogenic transitions, indicating distinct effector suites are deployed at each stage. Using fluorescent protein tagging and transmission electron microscopy-immunogold labelling, we found effectors localised to stage-specific compartments at the host-pathogen interface. In particular, we show effectors are focally secreted from appressorial penetration pores before host invasion, revealing new levels of functional complexity for this fungal organ. Furthermore, we demonstrate that antagonistic effectors either induce or suppress plant cell death. Based on these results we conclude that hemibiotrophy in Colletotrichum is orchestrated through the coordinated expression of antagonistic effectors supporting either cell viability or cell death

    Individuelle und strukturelle Faktoren der Mitgliederbindung im Sportverein

    This article analyses the conditions influencing the commitment of members of sports clubs. It focuses not only on individual characteristics of members, but also on the corresponding structural conditions of sports clubs related to the individual decision to quit or continue their membership. The influences of both the individual and context levels on the commitment of members are estimated in different multi-level models. Results of these multi-level analyses indicate that commitment of members is not just an outcome of individual characteristics such as strong commitment to the club, positively perceived communication and cooperation, satisfaction with sports clubsʼ offers, or voluntary engagement. It is also influenced by club-specific structural conditions: commitment is more probable in rural sports clubs, and clubs who explicitly support sociability, whereas success-oriented sporting goals in clubs have a destabilizing effect.In diesem Beitrag werden Bedingungen analysiert, die die Mitgliederbindung in Sportvereinen beeinflussen. Neben individuellen Merkmalen interessieren dabei auch die Strukturbedingungen der Sportvereine, die im Zusammenhang mit der individuellen Wahlhandlung zwischen stabiler Mitgliedschaft oder Austritt stehen. Der Einfluss der Individual- und Kontextebene auf die Mitgliederbindung in Sportvereinen wird anhand unterschiedlicher Mehrebenenmodelle geschätzt. Die Analysen machen deutlich, dass sich die dauerhafte Mitgliedschaft in Sportvereinen nicht allein auf individuelle Merkmale der Mitgliedschaft, wie eine ausgeprägte Verbundenheit, ein positiv wahrgenommenes soziales Miteinander, die Zufriedenheit mit der Vereinsarbeit sowie die ehrenamtliche Mitarbeit zurückführen lässt. Darüber hinaus nehmen vereinsspezifische Strukturbedingungen Einfluss auf die Mitgliederbindung, wobei in ländlich geprägten Sportvereinen und in Vereinen, die Geselligkeit explizit fördern und in denen das Vereinsziel sportlicher Erfolg eher eine untergeordnete Rolle spielt, die Austrittswahrscheinlichkeit geringer ist

    Optimization of Coding Gain for Full-Response CPM Space-Time Codes

    Abstract — Conditions are derived under which M-ary fullresponse CPM space-time codes will attain full spatial diversity and optimal coding gain. General code construction rules are desirable due to the nonlinearity and inherent memory of CPM signals which make manual design or computer search difficult. Optimization of the coding gain for CPM space-time codes is shown to depend on the CPM frequency/phase shaping pulse, modulation index, and codewords. The modulation indices and phase shaping functions that optimize the coding gain are specified. Finally, optimization of ST-CPM codewords is discussed. I
